Abstract

Batik industries have negative impacts on the environment from the dye used. An alternative method for decolorization in batik wastewater using the fungi Pleurotus ostreatus was grown in baglog. Spent mushroom (baglog) still contains fungal mycelium of P. ostreatus and contaminant fungus such as Penicillium sp., and Aspergillus sp.). Those fungi have a potential to decoloring dyes in batik wastewater. The aim of this research was to determine the ability of fungal mycelium isolated from spent mushroom P. ostreatus at decoloring batik wastewater, and determine the most effective fungal mycelium to decoloring batik wastewater. The results showed that the mycelium isolated from sewage baglog P. ostreatus was able to decoloring batik wastewater. Penicillium sp. mycelium is most effective at decoloring batik wastewater.

Abstract

λ-cyhalothrin is phyrethroid insecticide that can pollute the waters environment and cause sublethal as well as lethal effects to non-target organisms such as tilapia. This research were aimed to study the LC50 96 h value and histopathological effects of λ-cyhalothrin on the gills, liver, and small intestine of tilapia. In this research fish were divided into control group and the treatment groups of λ-cyhalothrin at the concentration of 3, 6, 9, and 12 µg/L. The treatment used a static system for 96 hours. The LC50 96 h value was analyzed by linear regression. Histopathological effects on the organs were analyzed using descriptive comparative. Histological preparation was done by the paraffin method, fixed in Bouin solution, and stained with Hematoxylin-Eosin. The results showed that LC50-96 h values of λ-cyhalothrin on tilapia was 6.519 µg/L. Gills damages were in the form of hyperplasia, necrosis, edema, secondary lamella fusion, removal of lamella epithelial cells, and primary lamella dilatation. Liver damages include hemorrhage, intravascular hemolysis on blood vessels, piknosis, and leukocytes infiltration. The intestine damages were fatty degeneration, edema, leukocyte infiltration, hemorhage, cloudy swelling, vacuolisation, necrosis, and arthropy. The gills, liver, and small intestine damages of tilapia range from slight to severe. It could be concluded that λ-cyhalothrin caused histopathological effects on the gills, liver, and small intestine of tilapia.

Abstract

Mangosteen (Garcinia mangostana L.) is one of Indonesia's tropical horticultural commodities with high economic value. Mangosteen also has a lot of functions such as a producer of a wide variety of functional compounds such as anticancer, antiviral, antibacterial and antifungal that have some bioactive component i.e., micotoxin, enzyme, and antibiotics. The research was carried out to investigate the endophyte fungi in mangosteen plant tissue. Endophyte fungal isolation from mangosteen plant tissue resultsed 11 isolates which have identified Phoma sp., Acremonium sp., 2 isolates of Penicillium sp., Geotrichum sp., Pestalotiopsis sp., Botryosphaeria sp., Colletotrichum sp., Chrysosporium sp., Aspergillus sp. and Blastomyces sp., and 2 isolates have not been identified.

Abstract

Planaria sp. in Bukit Tunggul (Lembang) is rare within 10 years. This animal is sensitive to polluted water. Beside that, the number of this animal is decreasing due to alteration of landuse and climate change. This animal plays an important role as a bioindicator for clean water and as a media in learning of biology in high school as well as in university. Due to these need, study on ecology and distribution of Planaria sp. is done in Panaruban water fall, Subang. The sampling location is limited only at Cimuja, Karembong, Sawer and Goa Badak, which have an environment characteristics variation. Observations are done including water quality chemically, land use, and climatic factors within 3 times monthly. Planaria sp. counting is done stratified random sampling using a 1x1 m quadran. The number and biomass of Planaria sp. related to climatic factors are discussed. In general, it can be concluded that, abiotic environmental factors contributes to the abundance and biomass of Planaria sp.

Abstract

Enzyme Linked Immunosorbent Assay (ELISA) is an immunological method can be used to analyze aflatoxin B1 (AFB1) in feed. ELISA technique must be done by using an instument (ELISA reader) which is not effective when used in the field. Therefore, the simple ELISA technique is needed such as indirect dipstick ELISA (d-ELISA). The aim of research is to develop AFB1 screening method using   d-ELISA. The research is focusing on  development and validation of indirect d-ELISA, and its application on sample of feed and corn. The results showed that the best coating time for antigen AFB1-BSA (0,4 ug/ml) is 24 hours, reaction time for antibody anti AFB1 (1/800) and AFB1 standard is 15 minutes, and reaction time for goat anti rabbit-HRPO conjugate 1/2500 and substrate of orthodiasianin (ODN) is 20 minutes. The results of indirect d-ELISA on 22 samples are found that 7 samples are contaminated by AFB1 with concentration above 20 ng/g and 7 samples are contaminated by AFB1 with concentration in the range of 0 – 20 ng/g. Finally, it is concluded that the indirect d-ELISA is applicable to be used in the fields.

Abstract

Protease activity and body proximate composition of eel, Anguilla bicolor McClelland., under starvation and refeeding conditions with three treatments and four replications in randomized completely design has been evaluated. The result showed that the protease activity of eel was no significant different (P>.05) between treatment, but body proximate composition has significant differences (P<.05), especially in protein and lipid contents at starvation condition. It can be concluded that the eel protease activity did not change under starvation and refeeding condition, but the lipid content decreased at starvation condition.
