Abstract

issue cultures that have passed callus phase can induce somaclonal variation, of which the intensity was influenced by the addition of selective agents to culture media. Somaclonal variations of peanut plant obtained from in-vitro cultured and in-vitro selected somatic embryos using PEG was not yet understood. The objectives of this research were 1) to identify the qualitative variants of peanut plant var. Kelinci obtained from in-vitro cultured and in-vitro selected somatic embryos using PEG, 2) to estimate the factors that control the qualitative variants. The non-selected and the selected variant somatic embryos of peanut were germinated and cultured. From fertile R0 lines, sufficient number of R1 and R2 progenies were grown for evaluation. The results showed that phenotypic variations of qualitative characters can be observed among R0, R1 and R2 generations of somaclonal lines. Variant phenotypes of qualitative characters can be observed, these include wide branching, excessive branching, leaf variegation, leaflet number abnormality, leaf pointed tip, ‘rosette’ leaf, complete sterility and partial sterility. The data indicated that wide branching, excessive branching, leaflet number abnormality, partial sterility and complete sterility were genetically controlled, while variant phenotypes of ‘rosette‘ leaf, leaf variegation, and leaf pointed tip were epigenetically controlled. Abstract

In this research, we have studied synthesis of improvement of andrographolid bioactive compound on cell culture of sambiloto by addition of jasmonic acid. The essential problems in this research are firstly, the effects of addition of jasmonic acid either can induce or not andrographolide synthesis improvement of cell culture of sambiloto and secondly, to observe the largest content of andrographolide in jasmonic acid concentrations. Meanwhile, the purpose of this research are to observe the functions of jasmonic acid elicitor for induction of andrographolide synthesis improvement of cell culture of sambiloto and to optimize jasmonic acid concentrations which can produce the largest andrographolide content. The independent variable is concentration of addition of jasmonic acid on cell culture and the dependent variable are the growth of cell suspension culture and andrographolide bioactive content. Experiment result show that the optimum medium of sambiloto cell consist of Murashige & Skoog (1962) medium supplemented by 0,5 ppm kinetin and 2,4-D 5 ppm. The cell growth phases are the followings : lag phase at age of 0-5 days, exponential phase of 5-15 days, and stationary phase at age of longer than 15 days. The highest andrographolide was 4,66 x 10-2 reached in cell culture was supplemented with 10 µM jasmonic acid. Keywords : andrographolide, sambiloto cell suspension culture, jasmonic acid elicitor.

Abstract

Organs of many plants have been used as male contraception materials. One of the organs is papaya seed. This seed contains carpain, a molecule that could cause vacuole initiation on Sertoli cell. The research aimed to know whether papaya seed carpain affects mice spermatozoa quality. This research is an experimental one that makes use of 24 male mice. These mice are separated into 4 groups. Each group has its own papaya seed extract doses. The first group is the mice group that is given by doses of 0 mg/200 gr body weights. And the following groups are 10 mg/200 gr body weight for group II, 20 mg/200 gr body weight for group III, and 40 mg/200 gr body weight for group IV. Oral supplement of the papaya seed is 1 ml/mouse/day in the term of 40 days. These oral supplement is followed by reversibility time for 40 days. The data consist of spermatozoa viability, motility, and amount. These data are analyzed by 2 x 4 factorial analysis in the test scale of 5%. Statistical analysis shows significant differences among each treatment (8.27 > 2.76), spermatozoa taking time (5.81 > 4.60) and dose variation among groups (15.29 > 3.34). There are also significant differences among motility treatment (24.94 > 2.76), time (4.66 > 4.60) and doses (55.03 > 3.34). There is no significant difference among viability (3.22 < 3.74) nor treatment (2.30 < 2.76). The research concluded that papaya seed extract affects the spermatozoa motility and the decrease of spermatozoa numbers, but not to the reversibility and the viability. Abstract

Tempe content of aglikon isoflavone which have potency to as anticancer compound. During processing of tempe, beside tempe mold (Rhizopus oligosporus) share other microbial namely bacterium. One of the bacterium residing at tempe ecosystem is Micrococcus luteus, having the character of gram positive and can conduct genistein biotransformation and daidzein become factor II compound (6,7,4-tri hydroksi isoflavone). Its problems how amount of R.oligosporus cell and M. luteus at processing of tempe associated is full scale of yielded tempe isoflavone. The study aimed to improve full scalely tempe aglikon isoflavone through inoculation of M.luteus bacteria. Used complete random designd, with M.luteus bacteria, as independent variable and variable depended is the amount of mold cell (R..oligosporus), amount of full scale M.luteus cell and total of isoflavone. Result of one way Anava indicate that inoculation of M.luteus bacteria do not degrade the amount of R.oligosporus cell with F call (53,28) > F tab (3,10) at significancy level 5%. Amount of M.luteus cell from 1,3 x 109 cell / gr [at] fermentation first becomes 3,4 x 106 cell/gr and decrease after day fermentation second becoming 1,75 x 106 cell/gr. Proportion of increasing isoflavone content at day fermentation 1-2 from 19,1 becoming 52,7 mg/100gr. Inoculation of M. luteus bacteria increased to count isoflavone aglicone and no trouble at growth of tempe mold and it relate to the increasing of is full scale of yielded tempe isoflavone.

Abstract

The morphology and isozyme marker was not enough to reveal banana genetic diversity. So, it needs molecular analysis. The purpose of this research is to know the genetic diversity of banana with B genome using microsatelite marker. Forty bananas with B genom from Yogyakarta Agriculture Service and Animal Husbandary Service and also from Tropical Fruit Centre of Bogor Agriculture Institute were amplified using 6 loci (MaCIR327b, MaCIR108, Ma-3-139, Ma-3-90, Ma-1-17, and Ma-1-27). The diversity were identified with allele amount, genotype, observe heterozigosity, and filogeny. There are eight allele per locus average from six microsatelites. MaCIR108 produced the highest allele amount, there are 13 alleles. The research identified 75 genotypes. MaCIR327b, MaCIR108, Ma-3-139, Ma-3-90, Ma-1-17, and Ma-1-27, respectively, produced 8, 20, 13, 14, 15, and 5 alleles. The Ho average is quite high, there are 0,77. The similarity coefficient is between 0,597 – 1,00. MaCIR108 and Ma-3-90 can be used to identify banana group genom. As a group genom characteristic, Ma-1-17 needs more research. Identification result between morphology and microsatelite marker were different. Based on morphology marker, Sri Wulan and Byar cultivar were ABB, and Ketip Gunung Sari were AAB, but based on microsatelite marker Sri Wulan and Byar cultivar were AAB, while Ketip Gunung Sari has not B on its genom. Abstract

The activities of tomato juice’s antioxidant for the prevention of lung tissues damage in mice cigarette smoke induces endogen antioxidant is no longer effective on oxidative stress control towards cigarette consumer lung, thus exogenous antioxidant is a must. A kind of exogenous antioxidant is lycopen, a content of tomato. 40 mice is divided into 5 groups with 8 of each. Group I is the controlling group, group II is the negative control, and group III, IV, V are the group of mice that is supplied by tomato juice on the 1,7 g/100 g weight/day ; 3,4 g/100 g weight/day; and 5,1 g/100 g weight/day dozes. Each group, except the control one, is forced by cigarette smoke for 15 minutes per day in 54 days. On the 55th day, all mice is sacrificed. Specimens of their lungs are made in the way of histological by haematoxylin – eosin method. The data is about the comparison of lung tissue failed on each group, that is analyzed by Anava. The observation of each tissue on the group II showed emfisema pattern, that is alveolus membrane epithelia cell with no nucleus nor endothelium, with wide alveolus lumen and loose alveolus junction. The identical situation also found on group III. The failed tissue cannot be found on group IV and V. Antioxidant of tomato is capable of controlling lung tissue failed of mice of cigarette smoke. Abstract

A research on the use of sucrose and lactose as carrier media to protect Acetobacter xylinum cell subjected to freeze drying has been done. The aim of the research was to know the number of the viable cells from dried culture and to know the concentration of the carrier medium that would give best result. The best result is sucrose at the concentration of 15% that produced 28.2 x 106 viable cells/ ml of rehidrated culture. The rehidrated culture used in the research was Schramm & Herstin medium.

Abstract

Fusarium disease of tomato plant is caused by Fusarium oxysporum. For gaining high quality and stable tomato production but physically construction, natural management of environment balancing, consistency, and stability is a must. A kind effort that should be tried is using natural agent, Trichoderma. This research aims to know the effect of Trichoderma implementation towards Fusarium oxysporum growth, and Tomato fruit production and vitamin content. There are two steps of this research. The first is seeing Trichoderma perform towards Fusarium oxysporum growth in a variation of incubation periode (7, 14. 21 and 28 days). The second, is giving Trichoderma agent on the tomato planting in some dozez (g), such as 0,10,20,30,40 and 50. The parameter are Fusarium oxysporum growth limits area, fruit weight per plant, and fruit vitamin C content. The research shows that Trichoderma giving are effecting Fusarium oxysporum growth, tomato fruit weight and fruit vitamin C content in a significance way. The highest level of vitamin C content (84,78 mg/matterial) and fruit weight (550 g) is gained on Trichoderma implementation which has incubation day of 21 (19.08 % ) and dozez of 40 grams.   Keywords : biofungisida,Trichoderma, Fusarium oxysporum , tomat

Abstract

Ploceidae family’s birds in Indonesia consist of 41 species include 13 species as Java’s endemic. Some species of Ploceidae family were start to rarely oberved, because of new house developing and hunting as a pets, zoo collection and education kit. It’s need some efforts to conserve these rare species through improving the habitat, rehabilitation, nursery, controlling, law inforcement for hunters, and genetic conservation according to a knowledgement of genetic variation. The research aimed to knew the length fragmen of ND3 DNAmt Gen and genetic variation among species of Ploceidae family’s birds. 11 species were observe for morphology’s characteristic than blood sample were collected for DNA isolation with Dixit methode. ND3 gen were amplificated by DNA isolated with PCR used H11151 and L10755 primer. PCR’s gain were visualized with 2 % agarose gel. The length fragmen of ND3 DNAmt were 321bp for Bondol Jawa, 338 bp for Bondol Haji and Bondol Peking, 393 bp for Burung Gereja Erasia, 413 bp for Manyar Emas, 406 bp for Gelatik Jawa, 334 bp for Manyar Tempua and Manyar Jambul, 351 bp for Pipit Benggala, 333 bp for Bondol Hijau Binglis, 317 bp for Pipit Zebra. The conclusion of this research were : 1). the length variation of ND3 DNAmt Gen among 11 species range from 317 – 413 bp and 2). morphological variation and length variation of ND3 DNAmt Gen shows that there was genetic variation among species of Ploceidae family’s birds. Abstract

This research would like to show the effect of tomato juice in a variety processing method towards hypercholesterolemia albino rat blood plasma lipid fraction. The special purpose if to know and analyze tomato juice advantages towards total cholesterol content, LDL–cholesterol, HDL–cholesterol and triglyceride. The research design is a Post Test Randomized Control Design by means of Complete Randomized Design (RAL). There are 4 groups of treatment, each groups has 6 repeaters. The data is tested by One Way Varian Analyze. If there is a significance difference, there will be Duncan’s New Multiple Range Test followed. The research shows that there is a significance difference of total cholesterol content, LDL–cholesterol, HDL–cholesterol and triglyceride. The highest total cholesterol content, LDL–cholesterol and blood plasma triglyceride content are on the hypercholesterolemia control group. The lowest total cholesterol content, LDL–cholesterol and blood plasma triglyceride content are on the group that is given by a fried tomato juice. The lowest HDL content is on the hypercholesterolemia control group and the highest one are on the group that is given by a fried tomato juice. The research conclusion is that tomato juice supplementation able to fix hypercholesterolemia albino rat blood plasma lipid fraction, especially to decrease total cholesterol content, LDL–cholesterol and blood plasma triglyceride, and increase hypercholesterolemia albino rat blood plasma HDL-cholesterol content. Fried tomato juice give the best effect on decreasing total cholesterol content, LDL–cholesterol and triglyceride and increasing blood plasma HDL-cholesterol content.
