Abstract

Diabetes mellitus (DM) is a disease or chronic metabolic disorder that can cause long-term complications such as damage to the liver, heart, eyes, and kidneys. One of the diabetes drugs is glimepiride. Glimepiride has the main mechanism of action of lowering blood glucose by stimulating insulin secretion from the granules of Langerhans cells. This study aims to determine the effect of glimepiride on reducing blood glucose in diabetic-kidney dysfunction mice. The design of this research is Post-test Only Control Group Design. This study used 105 mice which were divided into 7 groups. The dose of glimepiride was made in three variations: 1 mg/kgBW, 2 mg/kgBW and 4 mg/kgBW. The results showed no significant effect on decreasing blood glucose levels in diabetic-kidney dysfunction mice at various doses of glimepiride (p>0.05). Furthermore, further tests were carried out which showed that there was a significant effect on the duration of administration of various doses of glimepiride in reducing blood glucose levels in diabetic-kidney dysfunction mice (p<0.05). From the results of this study, it can be concluded that the pathology of the disease does not significantly affect the mechanism of action of glimepiride. Still, the length of time glimepiride is administered to diabetic-renal dysfunction mice shows a significant effect.

Abstract

Flavonoids in soursop leaf ethanol extract have various pharmacological effects but are easily degraded due to the influence of pH, oxygen, and intense light exposure. Encapsulation using chitosan polymer in nanoparticles is able to overcome these limitations. The formulation of the nanoparticle system in lotion and cream preparations can increase the comfort of use by consumers. Therefore, this study was conducted to determine the effect of lotion and cream dosage forms on the antioxidant activity of soursop leaf ethanol extract nanoparticles. Soursop leaf extraction was done by soaking at room temperature for 3 days using 70% ethanol as solvent. The soursop leaf ethanol extract nanoparticle system was prepared with three graded Soursop leaf ethanol extract concentrations F1 100 mg, F2 200 mg, and F3 300 mg. The nanoparticle system is then formulated into lotion and cream preparations. Evaluation of antioxidant activity was carried out by reacting it with DPPH, and the IC50 value of lotion preparation was obtained and showed the result was (F1) 83,34±0,130; (F2) 74,69±0,131; and (F3) 64,26±0,129 ppm, while the IC50 value of cream was (F1) 84,32±0,133; (F2) 75,35±0,130; and (F3) 63,74±0,120  ppm. The antioxidant activity of soursop leaf ethanol extract nanoparticles in lotion and cream preparations is included in the category of strong antioxidants. Statistical analysis using an independent t-test of the antioxidant activity showed p>0.05, so it was concluded that the ability to scavenge free radicals from soursop leaf ethanol extract in the nanoparticle system was not affected by the dosage form.

Abstract

This study aims to explore the chemical compounds of the coffee pulp as an anti-covid-19 (anti-3C-like protease). In silico modeling was carried out by structure retrieval, protein preparation, and docking simulation. Specific Grid docking was used for ligands – protein interacting by using Molegro Virtual Docker 5, followed by visualizing Discovery studio version 21.1.1. Compounds contained in coffee pulp, quinic acid, gluconic acid, and ferulic acid separately bind to the same amino acid residues of 3CL Protease or each other. Furthermore, three compounds inhibited 3CLPro activity by catalytic site blocking of 3CLPro, indicating that coffee compounds have potential as molecular therapeutic materials for the coronavirus by inhibiting SARS-CoV-2 genome replication.

Abstract

A diabetic foot ulcer is a complication of diabetes that causes open sores that can develop into an infection. The selection of the right antibiotics is based on the pharmacokinetic profile of antibiotics that can show antibiotic activity as bactericidal or bacteriostatic, the main bacteria found in diabetic ulcer disease are gram-positive bacteria such as Methicillin-Resistant Staphylococcus aureus (MRSa). This study aims to determine the antibiotics acting on diabetic ulcer tissue and the pharmacokinetic profile and penetration of antibiotics into diabetic ulcer tissue based on a literature study using the PICO (patient/population, intervention, comparison, and outcomes) method. The results of the analysis of 8 articles that met the inclusion criteria showed that the pharmacokinetic profile (PK) of several antibiotics obtained showed bactericidal activity in the treatment of diabetic ulcers including the antibiotics ceftolozane/tazobactam, tedizolid, cefepime, daptomycin, trimethoprim/sulfamethoxazole. Several antibiotics have shown activity as bacteriostatic in the treatment of diabetic ulcers, including linezolid antibiotics and vancomycin antibiotics.

Abstract

Burns is a form of tissue damage caused by contact with sources that have high temperatures such as fire, electricity, and chemicals. Pangi leaves contain chemical flavonoids, alkaloids, sponges, tannins, and terpenoids that can be used as an alternative in the treatment of burns. This study aims to determine the effect of ethanol extract from Pangi leaves on the healing of second-degree burns. The research design used was an experimental method using white rats with Wistar strain as experimental animals. The treatments were made of 4 kinds with 6 repetitions each. Pangi leaf extraction was carried out by maceration method with 95% ethanol. Variations in treatment were positive control using silver sulfadiazine ointment, negative control without treatment, 2.5% ethanol extract of Pangi leaves and 5% ethanol extract of Pangi leaves. Burns were made by using an iron plate heated with Bunsen fire for 2 minutes and then affixed to the back of the rat for 5 seconds until a second burn was formed which was marked by a reddish color and there were bubbles filled with fluid in the wound. Observations were made by looking at and measuring the width of the wound on day 1, day 2, day 4, and day 7. The results showed that the ethanol extract of Pangi leaves accelerated the healing process of second-degree burns in white rats with the Wistar strain. Pangi leaf ethanol extract with a concentration of 5% was more effective than the concentration of 2.5%. It can be concluded that the ethanolic extract of Pangi leaves affects the speed of wound healing in white rats of the Wistar strain and can be considered one of the active ingredients for the treatment of burns.
