Abstract

Obesity is defined as a condition with abnormal or excessive fat accumulation in adipose tissue so that it can interfere with health. The increasing prevalence of obesity was a major health problem worldwide. One effort to prevent obesity can be done by utilizing natural materials in Indonesia, including katuk leaves (Sauropus androgynus (L.) Merr). Katuk leaves contain kaempferol compounds which had the effect of reducing body fat or antiobesity. The nanoparticle formulation of katuk leaf ethanol extract made using a polymer of sodium alginate and calcium chloride (CaCl2) as a crosslinking agent. This study aims to determine the effect of the use of sodium alginate and calcium chloride (CaCl2) on physical characteristics in nanoparticle extracts of katuk leaf ethanol, and determine the nanoparticle formula with optimal physical characteristics. Optimized parameters were visual, transmittance, adsorption efficiency, particle size distribution, polydispersity index, zeta potential and particle morphology in the optimal formula of katuk leaf ethanol extract nanoparticles. Based on Design Expert 10.0.1 with the factorial design method, the optimal formula is obtained with a ratio of sodium alginate and calcium chloride (CaCl2) of 0.102%: 0.010%. Visual prediction results obtained 3 (clear); transmitting 90.66% and absorption efficiency 81.1359%. The t-test results showed that these observations differed insignificantly to the results of the predictions indicating that the equation of each optimization parameter was valid. The optimal formula had 66.4% units 54.59 nm of size nanoparticles and 33.6% units 440.1 nm, 0.465 of polydispersity index, -17.6 mV of zeta potential and particle morphology with a non-spherical shape in a loose aggregate.

Abstract

Alkaloids quinine is found in the quinine plant which is the raw material for the manufacture of quinine pills that are effective in the treatment of malaria. Selection of the method, solvent, identification technique and characterization of the alkaloid quinine compound in the quinine plant (Cinchona succirubra Pav. Ex Klotzsch) need to be done in an effort to produce the compound with the best separation.Extraction by maceration method, identification of groups using phytochemical screening methods. Fractionation using liquid-liquid extraction method and column chromatography. Isolation using preparative chromatography method and TLC-Densitometry method. Maceration extraction obtained the filtrate with a reddish brown color. Phytochemical screening was positive for triterpenoids and alkaloids. Liquid-liquid extraction resulted in the water fraction, ethyl acetate fraction I, and ethyl acetate fraction II. TLC and identification of positive chemical reactions containing quinine in ethyl acetate fraction II. Column chromatography obtained results in the form of 4 fractionations with different colors. Preparative chromatography and compound isolation by TLC-Densitometry showed the average quinine content of ethyl acetate fraction was 15.30%. The ethyl acetate fraction of quinine stem bark (Cinchona succirubra Pav. Ex Klotzsch) produced average quinine alkaloids of 15, 30% according to the cultivated kina plant which contains up to 15% quinine alkaloids.

Abstract

The novel coronavirus SARS-CoV-2 continues causing viral respiratory illness, known as Coronavirus Disease-2019 (COVID-19). COVID-19 is considered as the biggest global pandemic disease in 2020 and its antiviral therapies are still lacking worldwide. Current studies are still trying to find potential therapies of the disease, including from natural compounds. Therefore, it is important to acknowledge what the most effective herbs’ compounds can be used against the virus. Jamu is traditional medicine from herbs in Indonesia that possesses some medicinal properties. This research was done to find and explore the potential of Indonesian jamus as novel COVID-19 treatment. 283 bioactive compounds from Indonesian jamus were screened with ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) predictions and Lipinski’s Rule of Five parameters using ProTox-II and SwissADME. The 12 screened compounds then continued to be tested through molecular docking against SARS-CoV-2 Main Protease (Mpro) using AutoDock Vina. The result of our study showed that Curcuminol D has the lowest binding free energy of -6.9 kcal/mol and Ki (inhibition constant) of 2.99 µM compared to the other screened compounds. However, none of the screened compounds has lower binding free energy than the positive controls’, which valued -8.2 kcal/mol (Lopinavir) and -7.8 kcal/mol (Ritonavir). Despite their unfulfilled parameters, the other 271 compounds might have higher potential than the standard drugs. Therefore, the finding in the present study can be used as a starting point in the drug discovery process from natural compounds for treating COVID-19.

Abstract

Diabetes therapy with commonly used of anti-diabetes drugs (OAD) have unwanted side effects and the high price become problematic, so it is necessary to find or develop other safer, more effective, and inexpensive OAD alternatives. Based on in vitro and in vivo research models, flavonoids are proven to have efficacy in lowering blood glucose levels. In the citrus peel, flavonoid compounds are abundant. However their utilization is not optimal even though the mass production is very high. This study is to review current information regarding the effects of flavonoids on citrus peel in diabetes management and their molecular mechanisms. A literature review was conducted using the electronic databases of Scopus, ScienceDirect, and the American Chemical Society covering the most recent literature published from the years 2010 till present. Several keywords were combined to ensure all in vitro and in vivo studies were obtained. Key words used were “citrus peel”, “flavonoid”, “diabetes mellitus”, “mechanism”, and “blood glucose”.Based on the results of research that has been conducted by previous researchers, it was found that the flavonoids of citrus peel can improve glucose metabolism, hepatic enzyme activity, insulin signalling and lipogenesis regulation, repair damage to pancreatic islet cells and stimulate insulin secretion, and protect against complications of diabetes. Overall, citrus peel flavonoids as an antidiabetic can prevent an increase in blood sugar levels and reduce insulin resistance, the two hallmarks in diabetes melitus. Flavonoids play a role in enzyme inhibition, which is the main mechanism of medicine as well as inhibition at the level of gene expression and its antioxidant properties. Further research on the safety and efficacy of flavonoids is needed for the development of citrus fruit peel flavonoids as an alternative therapy for diabetes melitus.

Abstract

Natural ingredients are a good source of active compounds and drugs. The study of plant content is very interesting in the discovery of active compounds, especially for therapy in asthma. Asthma is a chronic disease with symptoms of wheezing, shortness of breath, chest tightness, and coughing. There are several mechanisms of action of anti-asthma compounds related to the types of asthma, namely H1 and H2 receptor inhibitors, reducing immunoglobin E levels, inflammation mediators, preventing secretion of cytokine Th2, and other specific mechanisms. The method used is literature study. The study was conducted by searching keywords such as herbs, anti-asthma, medicinal plants, and anti-asthmatic activity on electronic databases including ClinicalKey, Science Direct, PubMed, and Google Scholar. Therefore, data and explanations about potential medicinal plants found throughout Asia as alternative sources of asthma treatment were obtained.Thirty types of traditional medicinal plants in Asia that have the anti-asthma activity potential to have been obtained by various mechanisms of action. The plants activity data collected in this literature review indicate their use as an alternative source of asthma treatment.
