Abstract

Introduction: One strategy for HIV-1 (retroviridae) infection therapy is using NNRTI (non-nucleoside reverse transcriptase inhibitor) to suppress viral replication by binding to the reverse transcriptase (RT-ase) enzyme. One drug in this class is Efavirenz (EFV). Based on the good effectivity of EFV in HIV-1, it is suspected that EFV also has activity against HIV-2 and HTLV. This study aims to determine the activity of EFV in the human-infecting retroviruses such as HIV-2, HTLV-1, and HTLV-2. Methods: This study uses Bioinformatics and Medicinal Chemistry tools such as, phylogenetic tree construction, pairwise sequence alignment, looking for RT-ase motifs and domains, predicting secondary structures, modelling 3D RT-ase structures, docking, and predicting hydrophobicity with Kyte-Doolittle Hydropathy Plot. Result: There is a common ancestor of the four viruses according to the constructed phylogenetic tree. There are also RT_like Superfamily domains at HIV-2 and HTLV-1. The similarity and identity value, and results of HTLV-1 and HTLV-2 RT-ase structural modeling do not give good results. Binding energy value of EFV to HIV-2 RT-ase is higher than HIV-1 RT-ase. Conclusion: EFV is not effective against HIV-2, HTLV-1 and HTLV-2

Abstract

Sebagai upaya untuk mewujudkan salah satu tujuan SDGs 2030 yang berhubungan dengan ekosistem lautan dan sebagai upaya untuk mengurangi segala jenis polusi kelautan, terutama dari aktivitas daratan, penulis menggunakan kemajuan Ilmu Pengetahuan dan Teknologi (IPTEK) untuk mengembangkan sedotan yang berbahan dasar gelatin. “Tari Latin untuk Mantanmu” dibuat menggunakan mesin pencetak sedotan dengan menggantikan bahan dasar yang tadinya plastik dengan campuran gelatin dan air seperti yang tertera pada formulasi di atas. Setelah dicetak, sedotan gelatin dipisahkan dan dibiarkan pada suhu ruangan untuk mempercepat pengeringan dan mencegah agar sedotan tersebut tidak menempel satu sama lain. Selanjutnya, sedotan gelatin dikemas dalam plastik biodegradable dan siap dipasarkan ke masyarakat. “Tari Latin untuk Mantanmu” merupakan inovasi paling tepat dan efektif untuk menghadapi permasalahan – permasalahan yang terjadi di Indonesia saat ini, yang terkait dengan penumpukan sampah sedotan plastik. Selain itu, “Tari Latin untuk Mantanmu” dapat mencegah timbulnya penyakit yang disebabkan oleh sedotan plastik. Dengan diwujudkannya inovasi ini, diharapkan dapat membantu Indonesia bebas dari sampah sedotan plastik dan menjaga keseimbangan ekosistem lautan.

Abstract

Luka kronis merupakan bentuk kerusakan jaringan pada kulit akibat infeksi, trauma berulang, dan sebab sistemik seperti diabetes mellitus serta membutuhkan waktu lama untuk sembuh. Luka akan memicu mekanisme regenerasi sel yang meliputi tiga fase utama: inflamasi, proliferasi, dan pembentukan jaringan, tetapi adanya luka kronis akan menghambat penyembuhan luka (wound healing). Saat ini penggunaan obat sintesis masih menjadi pilihan utama, sedangkan pemanfaatan bahan alam sebagai obat untuk luka masih jarang dioptimalkan menjadi sediaan farmasetis. Selain itu, sifat fisikokimia bahan alam menyebabkan bioaktifnya sulit untuk menembus jaringan kulit. Pengembangan teknologi formulasi sediaan perlu dilakukan untuk mengoptimalkan potensi dari bahan alam dalam menunjang efektivitas pengobatan luka kronis. Formulasi sediaan nano gel dapat menjadi solusi untuk mengatasi hal tersebut. Nano Gel memiliki keunggulan seperti ukuran partikel kecil, lebih stabil, tidak lengket, memiliki efek pendinginan pada kulit dan secara estetika lebih disukai. Kandungan flavonoid dari daun binahong (Anredera corifolia) diketahui memiliki aktivitas antiinflamasi dengan menghambat enzim siklooksigenase dan lipoksigenase. Disamping itu kandungan sinamaldehid dari kayu manis (Cinnamomum verum) dapat mempercepat regenerasi sel dengan meningkatkan proses angiogenesis melalui up-regulation sinyal jalur PI3K dan MAPK serta telah terbukti memiliki aktivitas melawan bakteri gram positif dan gram negatif. Kombinasi kedua tanaman tersebut berpotensi mempercepat regenerasi sel pada luka melalui aktivitas antimikroba, mengurangi inflamasi dan meningkatkan proses angiogenesis. Formulasi nano gel akan membantu meningkatkan penetrasi bioaktif dan bentuk gel berperan sebagai wound dressing yang dapat melindungi luka dari kontaminasi. Dengan demikian, formulasi nano gel kombinasi sinergis dari aktivitas kedua tanaman tersebut diharapkan mampu menunjang dan mempercepat proses wound healing.

Abstract

Infection is the highest prevalence of disease in Indonesia. Efforts to increase the prevalence of infection is to improve the community's immune system. Kasumba turate is an endemic plant of South Sulawesi empirically used in the cure of smallpox and improve the nature of the body due to the nature of its imunomodulator. Traditional decoction of the Kasumba turate is less effective for consuming because it has a bitter taste, so the aim of this study is to modified decocta of kasumba turate becomes effervescent granules which is contain antioxidant to reducing free radicals in the body without reduce its usefulness. The method of making effervescent granule supplementation is by using infusa (decoction) from a lyophilized kasumba flower with Freeze Dryer and formulated with a combination of citric acid and sodium bicarbonate 1: 1.5. Effervescent granule preparationss were evaluated by LOD, MC, compressibility, species weight and pH test to obtain a qualified preparation and antioxidant analysis by DPPH method. Based on the evaluation of the preparation, it is found that the effervescent kasumba turate granule fulfill the applicable requirements: acid granules obtained% MC and% LOD 0.5% and 0.5%, basic granule% MC and% LOD 0.7%, 0.77%, flow properties break 27o, and flow rate 9.7 g/s),% compressibility 16% and weight type 1.66, pH of the preparation 7. The effervescent lyophilisate preparation of kasumba turate becomes an effective supplement in enhancing the immune system and as an antioxidant for all age.

Abstract

Introduction: Diphtheria is an upper respiratory tract infection caused by Corynebacterium diphtheria bacterium. The recent diphtheria outbreak caused a large number of deaths in rapid period. This is due to the absence of a fast and precise method of diphtheria diagnosis. Current diagnosis practices aredone by cultivating bacteria which takes a relatively long time, which is 3 to 5 days, while the patients may already develop severe infection by the time the results are accessible. Therefore, a sensitive, easy and fast diphtheria diagnostic method is needed so that patients suspected of being infected can betreated quicker.Methods: This advertorial is written based on literature study method.Result: The diphtheria diagnostic kit was designed based on the DNA hybridization method by using DNA microarray. There are probes on the microarray plate in the form of a single stranded DNA fragment that is complement to the unique area of gene that encodes diphtheria toxin. The test sample wasobtained from the preparation of the patient's throat smear and labeled with a fluorescence marker before dropped onto the plate. Thus, if there is diphtheria DNA in the patient's sample, hybridization will occur between diphtheria DNA and the complementary DNA probe. The occurrence of hybridization will be characterized by fluorescence as an indicator of whether patients have diphtheria or not. The application of this diagnostic kit can shorten the diagnosis of C. diphtheria to less than 24 hours .Conclusion: Diphtheria diagnostic kit based on DNA hybridization can be made and applied.

Abstract

Some components of cigarette smoke are gases, such as CO, CO2, HCN, and NOx which are the potential to cause free radicals. PAH contained in Cigarette smoke can cause testicular atrophy,inhibits spermatogenesis, and damage the morphology of spermatozoa, moreover nicotine exposure causes a decrease in testosterone levels through the mechanism of inhibition of Leydig cell function where functions is a testosterone hormone secretor. Pumpkin seeds has contain alkaloid compounds,saponins, kukurbitasin, lecithin, resins, stearin, phytosterol compounds, phenolic, fatty acids, squalen,tyrosol, vanylic acid, vanillin, luteolin and sinapat acids, vitamins (including vitamin β-carotene, vitaminA, vitamin B2, a-tocopherol, vitamin C and vitamin E). Vitamin C is a watersoluble vitamin that canprotect spermatozoa from damage by oxidative stress by neutralizing hydroxyl, superoxide and hydrogen peroxide radicals and prevent agglutination of spermatozoa. Vitamin E can reduce the levels of malondialdehyde in spermatozoa, increase spermatozoa motility and play a role in reducing DNAfragmentation of spermatozoa. The research has become the basis of writing health articles aimed at new contributions to science.

Abstract

According to WHO data in 2012 showed that global male mortality was 52% and was caused by non-communicable diseases (NCD). Compared to women, men are more likely to die prematurely (under 70 years) due to NCD in almost every country. Then, there is a survey in Asia that shows that life expectancy for men is lower than for women and is a sign of the need for improvement in men's health status about preventable health problems and encourages early detection and treatment of diseases in men. With education, awareness about personal health will increase, and also minimalize the wrong understanding of the clinical condition in men. Because men generally avoid to meet health practitioners, then monitor and treat their own symptoms, and eliminate the perception of having a disease (despite experiencing symptoms). In this technological era, information will be easier to obtain if accessed in the digital media, such as: via an online platform MATE: Men's Assistant Tracker on Health that can be accessed anytime, anywhere. This application educates both general and specificabout men's health and focuses on early detection of diseases combined with lifestyle advice with three main contents, namely: Daily Tracker, Health Insights, and Community.

Abstract

Diabetes mellitus is the increase in blood glucose level by a lack of secretion (insulin resistance) or an absence of pancreatic insulin which is one of the Non-Communicable Diseases (NCDs). 90% of existing cases are of type 2 diabetes mellitus (IDF, 2017). Type 2 diabetes, known as non-dependent-insulin occurs due to tissue resistance to the effects of insulin and the relative deficiency of insulin secretion (WHO, 2016).Type 2 diabetes mellitus is often diagnosed at a younger age and in men with lower body mass index than women. Various risk factors of diabetes are age, physical activity, body mass index (BMI), blood pressure, stress, lifestyle, family history, HDL cholesterol, triglycerides, gestational diabetes, history of glucose abnormalities and other complications. Other risk factors that may play a part are smoking and consuming alcoholic beverages (Isnaini, 2018). According to Riskesdas data in 2013, the prevalence of male smokers was 66%, which is about 10 times higher than women, which is 6.7% (Data and Information Center of the Indonesian Ministry of Health, 2013).Patients with type 2 diabetes mellitus, not only require non-pharmacological therapies such as diet regulation, physical activity, quitting smoking and alcohol, but also require pharmacological therapy that can be carried out by injecting insulin and/or taking oral antidiabetic drugs (Neal, 2012). However, insulin treatment therapy has a problem in patients who are afraid of needles and syringes, besides that, treatment with oral antidiabetic may cause several problems such as weight loss, hypoglycemia, digestive system disorders, etc. The cost of diabetes treatment is quite expensive due to long-term treatment and which may even last a lifetime. The cost needed to provide therapy to patients who have been diagnosed with diabetes worldwide from 2011 to 2030 is 1.7 trillion US$ (WHO, 2016). Considering the high cost of diabetes treatments and the side effects caused by antidiabetic drugs, alternative therapy is needed at a relatively cheaper price, with stronger potency and minimal side effects. This can be done using waste yet it is efficacious for the treatment of diabetes mellitus, an example being rice bran (Sivamaruthi, Kesika, and Chaiyasut, 2018).

Abstract

Minister of Health, Nila F. Moeloek, said that Indonesian men’s life expectancy is only 69 years. This data is worsened by the fact 9 last years of men is sickly age, leaving only 60 years of life span.There are many factors affecting the short age of Indonesian men, one of which is cardiovascular disease.Based on WHO 2012, cardiovascular disease claimed 31% of total world deaths and ¾ of it occurred in developing countries such as Indonesia. This is caused by high in fat, sugar and salt diet. As a result, risk factor for high prevalence of cardiovascular disease in Indonesian men is lifestyle. Of course, public participation would be minimal if they are forced to abandon the famous Indonesian culinary, one of the most delicious in the world. Even so, the problem suffered by these men cannot be ignored.Acaraki, or Nusantara’s ancient pharmacists have found herbs to deal with heart disease and its symptoms using ginger and soursop leaves. Phytochemistry has revealed the efficacy of Gingerol in ginger as a blood thinner, antihyperlipidemia, and antioxidants agent as well as efficacy of soursop as an effective vasodilator. All these properties are packed into herbal candy namely Permen Jasa.
