Abstract

Background: Physiologically, woman experiencing aging characterized by menopause. Gandar usa leaf contains isof lavon, a phytoesterogen expected to be used as esterogen replacement therapy.Objective: the aim of this research is to investigate the administrative ef fect of gandar usa leaf extract on the increase of estradiol hor mone level and the number of antral ovarium follicle of female mice.Methods: This is experimental study with posttest only control group research design. Samples are 24 balb-c female mice, aged 16-17 months, weighed 19-35 grams, divided into 4 groups. Control group (Ctrl-G) were given 0.48 ml aquadest; group JB10-G, JB20-G, and JB30-G, each were given gandar usa leaf extract with concentration 10%, 20%, dan 30% in volume 0.48 ml orally twice a day. Af ter treatment, dissection were conducted to make ovarium histology preparation with Hematoxylin Ehrlich-Eosin staining, obser vation were conducted using microscope.Results: Post hoc analysis indicated that the estradiol level and the number of antral ovarium follicle on administration of gandar usa leaf extract with concentration 10%, 20%, dan 30% are signif icantly higher, p < 0.05. The increase of estradiol level with extract concentration of 10% are signif icantly negatively cor related with the number of antral ovarium follicles p<0.05. negatively cor related with the number of antral ovarium follicles p<0.05.Conclusion: The administration of gandar usa leaf extract increase the estradiol hor mone level and the number of antral ovarium follicle in female mice.

Abstract

Background: Free radicals that enter the body due to consumption of reused cooking oil can cause liver cell damage. Mangosteen peel extract (Garcinia mangostana L) is known to contain mangostin as an ntioxidant. However, it is not known whether it can repair liver damage.Objective: To deter mine the ef fect of mangosteen peel extract on the levels of SGOT and SGPT of Wistar rats fed with reused cooking oil.the ef fect of mangosteen peel extract on the levels of SGOT and SGPT of Wistar rats fed with reused cooking oil.Methods: This study was a tr ue experimental study with post-test only controlled group design. Twenty four male Wistar rats were randomly divided into 4 groups randomly. The CN-G group was given the standard diet, the MJ-G group was given a standard diet and cooking oil, the MJM- 400 group was given standard diet, reused cooking oil, and mangosteen peel extract at a dose of 400 mg/KgBW, and the MJM-800 group was fed with a standard, reused cooking oil, and mangosteen peel extract at 800mg/KgBW. The treatment was car ried out for 28 days, and then continued with examination of reused cooking oil, and mangosteen peel extract at 800mg/KgBW. The treatment was car ried out for 28 days, and then continued with examination of SGOT and SGPT levels using the Inter national Federation of Clinical Chemistr y (IFCC) method without Pyridoxal Phosphate 37ºC.Results: Kr uskal Walis test showed that SGOT and SGPT levels showed no signif icant dif ferences between groups (p = 0.197 and 0.063, respectively).Conclusion: administration of mangosteen (Garcinia mangostana L) peel extract did not af fect SGOT levels, even tended to increase SGPT levels in ratsinduced by cooking oil.

Abstract

Background: Interleukin-6 (IL-6) has been known associated with oxidative stress and endothelial dysfunction, and also has important roles in pathogenesis of preeclampsia. Several studies have demonstrated that there was a significant increase of serum IL-6 levels in preeclamptic compared to normotensive pregnant women. However, study to evaluate serum IL-6 level in pregnant women with HELLP (hemolysis, elevated liver enzyme, and low platelet) syndrome is not well documented yet. This study aims to evaluate serum IL-6 levels between women with HELLP syndrome compared to normotensive pregnant.Methods: The research design was observational cross-sectional study. Samples were recruited consecutively using inclusion and exclusion criteria from emergency department and inpatient wards at Dr. Sardjito General Hospital, Yogyakarta, Indonesia. Serum IL-6 were collected from venous blood sample and measured by enzyme-linked immunoassay (ELISA) method. Independent-samples t-test or Mann-Whitney test was used to compare serum IL-6 values between women with HELLP syndrome and normotensive pregrnancy. A statistical measurement conducted using SPSS IBM Statistics 19® and considered significance when p<0,05.Results: There were 46 subjects, consisted of 23 women with HELLP syndrome and 23 women with normotensive pregnancy. Serum IL-6 level was a mean of 0,11±0,08 pg/mL and 0,15±0,20 pg/mL in women with HELLP syndrome compared to normotensive pregnancy respectively. There was no significant statistical difference between both groups (p=0,17).Conclusion:.The result of this study has shown that no significant difference of serum IL-6 level in HELLP syndrome women compared to normotensive pregnant women.

Abstract

Introduction: Good implementation of hand hygiene and the availability of cheap, affordable, and effective hand rub can prevent healthcare-associated infection. WHO has issued hand rub formulations that are easily self-produced, effective, and affordable. Objective: This study compares the difference between handrub’s effectiveness stored in National Hospital Diponegoro University ER, HCU, and storeroom for 2 and 10 weeks.Methods: This study was an experimental study with pretest posttest randomized group design. The effectiveness of hand rub measured by prEN12054 in cfu/ml. Results: There was no significant difference between efficacy of WHO formula A and B (p=0.458). In the Mann-Whitney test there was no difference between the effectiveness of WHO A and B formulas before storage (p = 0.567), after being stored for two weeks (p=1.000), and ten weeks (p=0.539). In the Kruskal-Wallis test, there was no difference in the effectiveness of the WHO A formula in three sites at weeks 0, 2 and 10 (p = 0.275, 0.584, 0.116), there was no difference in the effectiveness of the WHO B formula in three sites at week 0, the 2nd and 10th (p = 0.289.; p = 0.584, p = 1.000).Conclusion: No significant differences were found on the effectiveness of the WHO A and B formulas. There was no significant difference in the effectiveness of WHO A or B formulas stored in three sites. There were no significant differences in the effectiveness of WHO A or B formulas, before and after being stored for two and ten weeks in three sites.

Abstract

According to the free radical theory of aging, firstly proposed by Denham Harman, the most prominent cause of aging and degenerative diseases (DD) is continuously chemical reaction in cells and tissues. In this context aging and or DD is manifested as chemical composition, whilst aging process is reflected by chemical reaction affected by environment (Harman, 1981). Thus, both aging and DD have similar process, predisposed with oxidative stress and characterized by the progressive cellular alteration accumulation in line with time and account for increase in susceptibility to diseases and death. The most prevalent chemical reaction is between free radical reactive oxygen species (ROS) and reactive nitrogen species (RNS) with cellular biomolecules such as lipids, protein, carbohydrate, and DNA (Lamichhane et al., 2013). In order to prevent the deleterious effect of that chemical reaction, human’s body provides an antioxidant defense system to counterbalance the free radical dangerous effect. However, owing to inappropriate life style, eating style, physical activity, and poor environment, production of ROS and RNS may be increased and induce cellular damages. Numerous publications indicated that oxidative stress and cellular damages can be reduced even prevented by exogenous antioxidant, thus intake antioxidant from external sources is necessary. There are two type of exogenous antioxidant consisting of low molecular weight antioxidant (LMWA) and flavonoids. Unfortunately, the effect of LMWA on delaying aging and ameliorating DD remain inconsistent. On the other hand, numerous data indicate that flavonoids a botanical antioxidant was capable of improving oxidative stress and emerging as an alternative promising antioxidant (Sudhakaran et al., 2019). However, the effect of LMWA and flavonoids in combating oxidative stress remain unknown which one is better.

Abstract

Introduction: As various pterygium morphologies have been advocated as contributing factor on corneal astigmatism, little support in the literature available in establishing techniques in measuring pterygium thickness as clinical indicator.Objective: The aim of this study was to describe a quantitative method in determining pterygium thickness using anterior segment optical coherence tomography (AS-OCT).Methods: Anterior segment imaging was performed using enhanced high resolution cornea (EHRC) of Visante™ AS-OCT in 120 primary pterygium eyes. Prior to imaging, corneal topography assessment was performed on each pterygium eye in order to identify its topographic location. Based on topography mapping, three meridians (in degrees) were selected as close as possible to the pterygium border, which signify the demarcation of pterygium from the cornea. Reliability testing between intra and inter-observer of AS-OCT imaging modality was examined using intraclass correlation and scatter plot.Results: The overall (n = 120) mean and standard deviation of pterygium thickness EHRC of AS-OCT modality were 0.48 ± 0.10 mm (confidence interval: 0.45 – 0.50). EHRC of AS-OCT also showed excellent intra and intergrader reliability in measuring pterygium thickness with intraclass correlation of 0.997 (confidence interval: 0.994 – 0.998).Conclusions: EHRC of AS-OCT imaging modality is a better choice in assessing pterygium compared to traditional slit-lamp biomicroscopy. This tool is applicable for future work related to better understanding on the role thickness in pterygium morphology, its progression and prediction of induced corneal astigmatism and visual impairment due to pterygium.

Abstract

Introduction: Just like any other test, anatomical identification test can also cause high stress. Classical music intervention has proven effective in mitigating stress level in clinical context. However, whether or not it is effective as well to reduce stress level during anatomical identification test remains unknown.Objective: discovering whether listening to classical music can alleviate stress level in students when attending anatomical identification examination.Methods: Experimental research with post-test only control group design, involving 148 students as its sample obtained using proportionate stratified random sampling. The sample is divided into 2 groups: the group who listen to classical music and the one who did not listen to classical music. The stress level was measured using Visual Analogue Scale (VAS). The data are analyzed using Chi Square test.Results: Students who do not listen to classical music are mostly experiencing moderate (33 students or 22.29%), mild (29 students or 19.59%) and severe (12 students or 8.1%) stresses. On the other hand, students who listen to classical music are mostly experiencing mild (44 students or 29.72%), moderate (29 students 19.59%) and severe (1 student or 0.67%) stresses. The Chi Square test shows significant difference,  p= 0.002.Conclusion: Listening to classical music makes students have lower stress level when attending anatomical identification test.

Abstract

Background: Wound healing problems are often associated with TNF-α and VEGF expressions. However, the benefits and effects of ethanol extract cream from soursop leaves towards TNF-α and VEGF expression for burns healing process are yet to be known. Objective: To evaluate the benefits and influence of ethanol extract cream from soursop leaves in decreasing TNF-α expression and stimulate VEGF expression in BALB/c mice exposed to acute UVB radiation. Method: This study is a laboratory-based experimental study with post test only control group design. The subjects were 48 BALB/c female mice, divided into 24 subjects for TNF-α and 24 for the VEGF group, and were randomly distributed into 4 groups in each study, with each group containing 6 BALB/c mice. Group K was smeared with a base cream of 0.3 grams three times daily, while P1, P2 and P3 groups were each applied 0.3 grams of ethanol extract with consecutive consentrations of 2.5%, 5%, and 10% three times daily. On the sixth day, termination was performed on 24 mice to assess Tnf-α expression and on the eighth day also termination on 24 mice to assess VEGF expression. Preparation of substances was done using IHC method, analyzed using Kruskal Wallis test followed by Mann Whitney test.Results: The results of TNF-a and VEGF expression analysis based on Kruskal Wallis test obtained a p value of 0.0001 (p<0.05) showing that the mean expression of TNF-a and VEGF among the four groups ware significantly different. Mann Whitney test results revealed that the differences in expression of TNF-a and VEGF between the two groups were all significant differences (p<0.05). The mean TNF-a expression in the three treatment groups (PI, PII, PIII) ware significantly lower than the control group. The mean TNF-a expression in the PII and PIII treatment groups ware significantly lower than in the PI group, and the mean TNF-a expression in the PIII treatment group was significantly lower than the PII group. The mean VEGF expression in the three treatment groups (PI, PII, PIII) ware significantly higher than the control group. The mean VEGF expression in the PII and PIII treatment groups ware significantly higher than the PI group, and the mean VEGF expression in the PIII treatment group was significantly higher than in the PII group.Conclusion: Ethanol creams made of soursop leaves extract is proven to be useful and decreasing TNF-α expression and increase VEGF expression in BALB/c mice exposed to acute UVB rays.Keywords: Ethanol cream of soursop leaves extract, TNF-α expression, VEGF expression
