Abstract

Fat reduction has been performed on crude gelatin extracted from chicken feet (shank) through curing process using sodium hydroxide and acetic acid in various proportions. This study was conducted to evaluate the best fat reduction condition by the addition of acetic acid at concentrations of 0, 2.5 and 5% (v/v) with decantation time 0, 6 and 12 hours on crude gelatine from curing process at a ratio of 1% acetic acid; 1% acetic acid with 1% NaOH; 1% acetic acid and 3% NaOH and without curing (initial gelatine). The analysis was carried out to determine the composition of gelatin and their byproducts to total solids, total fat and fat binding capacity. The results showed that the variation in the ratio of the addition of acetic acid and decantation time affect the composition of fat and fat-binding ability. Based on fat binding capacity (FBC), treatment of chicken feet gelatin with curing of 1% acetic acid and 3% NaOH and continued by the addition of 5% (v/v) acetic acid with a 12-hour decantation time resulted in gelatin with the best FBC of 7.4 g/g, fat content 1.0248% and total solids 2.7575%.DOI : http://dx.doi.org/10.15408/jkv.v0i0.3149

Abstract

Gelatin as the main ingredient of capsules is still a problem for a moslem. Most of gelatin production remains largely derived from non-halal materials. One of gelatin source is came from collagen of the skin and bones of bovine or pork. The main of study is determine the source of gelatin used in hard capsules by using SDS-PAGE (Sodium Dodecyl Sulphate Gel electrophoresis Poly Acrylamide) method. In the early stages, optimization of standards bovine and pork gelatin were hydrolyzed by pepsin at pH 4.5 and 60°C for 1 hour, 2 hours, and 3 hours. Gelatin hydrolyzateswere analyzed by SDS-PAGE to determine the optimal hydrolysis time. Identification of gelatin hydrolyzate fragments were carried by molecular weight. Hydrolysis time optimization throught applied to identify the source of hard gelatin capsules in the samples obtained from market and compared with the simulation of hard gelatin capsules. The results showed there were of specific bands of bovine gelatin with a molecular weight of 11,4 kDa; 34 kDa; 47kDa and specific bands of pork gelatin with a molecular weight of 24.7 kDa; 28 kDa; and 60 kDa. Similar results were obtained on a sample of hard capsules with bands of protein fragments that were identical to bovine gelatinstandard. Based on the results, each of the samples were tested contain of bovine gelatin respectively. DOI :http://dx.doi.org/10.15408/jkv.v0i0.3150

Abstract

Fraksi A mengandung steroid. Sesuai dengan uji KLT dengan bantuan sinar UV.DOI :http://dx.doi.org/10.15408/jkv.v0i0.4965

Abstract

High concentration of phosphates in the water can lead to eutrophication which leads to uncontrolled growth of algae (algae blooming). It underlies the need for determining the concentration of phosphate in the aquatic environment. However, the concentration of phosphate may change during storage of samples so that an accurate analysis difficult to achieve unless carried out in-situ. DGT (Diffusive Gradient in Thin Films) is an in-situ measurement method developed for measuring phosphate and metal. In this study the use of Fe-Al-Oxide as a binding gel that is expected to bind phosphate with a capacity greater than ferrihydrite. N, N'-methylenebisacrylamide is used as a substitute for commercial DGT Crosslinker as crosslinking for a cheaper price and selective for small molecule. Ferrihydrite-DGT and Fe-Al-Oxide-DGT are tested with a variety of concentrations, pH, and contact time. In both methods DGT found that the pH for phosphate measurements performed at pH 3. Capacity of Fe-Al-Oxide binding gel known to be higher than the ferrihydrite binding gel with result Cferrihydrite-DGT:Cstart is 76% and CFe-Al-Oxide-DGT:Cstart is 82%.DOI :http://dx.doi.org/10.15408/jkv.v0i0.3597
