Abstract

The study aimed to assess genetic diversity and genetic relationship of ulin wildlings randomly collected from a nursery and originated from Bukit Soeharto natural forest, East Kalimantan. DNA templates were extracted from leaf samples of 1.5 years old wildings. Five RAPD primers consisted 55 polymorphic loci were used for genetic studies. Genetic diversity and relationship were analyzed using GenAlex software. The results showed moderate mean value of genetic diversity (HE=0,345, SE 0,015) of the wildings. Forty eight wildings were clustered in only 3 groups; almost all wildings (65%) were clustered in one main cluster. Moreover, 4 wildlings were clones (8%). In conclusion, the 48 wildings of ulin consisted high genetic relationship and individual clones that reflects the low genetic diversity of this species.

Abstract

The quality of sandalwood (Santalum album Linn) oil primarily depends on the compound of two major sesquiterpenes (α-santalol and β-santalol). The aim of this research was to identify variation of chemical compounds of sandalwood oil from various provenances. Fifty four samples from 8 provenances and one land race taken from 13th years old sandalwood planted at Gunungkidul, Yogyakarta Indonesia. Sandalwood powder were obtained by drilling tree at 10 cm above the ground horizontally. The samples were extracted with n-hexane solvent. Identification of chemical compounds of this essential oils were conducted by gas chromatography mass spectrometry analysis (GCMS). The GCMS investigation of sandalwood oils showed that variation compounds both among individual within provenance and between provenances were found in 11 individual from 6 provenances. Compounds such as sesquiterpene, monoterpenes, and others, were found in variying amount. The results showed that α-santalol (32.38-78.29%), β-santalol (5.45%-37.83%) were the major compounds with varying quantities of compounds range of epi-β-santalol (5.97-17.16%), P-Menth-2-En-9-OL-Trans (2.87%-8.53%), and α-sinensal (3.83-9.39%).

Abstract

Selection is a fundamental process in the establishment of a progeny trial that will be converted into a seedling seed orchard. In present study, selection practiced in first-generation progeny trial of jabon merah (Anthocephalus macrophyllus Roxb. Havil.) was observed. The study was aimed to find out a criteria selection accompanied with the effective coefficient weight for predicting genetic gain in the first-generation progeny trial of jabon merah. The trial was established in Wonogiri which was laid out in randomized complete block design (RCBD) using 55 families, 4 replications, 4 tree-plot with a spacing of 4 x 2 meter. Measurement was conducted at 40 months after planting covering tree height, diameter at breast height, stem straightness, stem volume. First within-plot selection by culling 2 poorer trees within each plot was practiced at 46 months age. Selection differential from within-plot selection was used to derive coefficient weight which was then applied to predict genetic gain. Results of study showed that selection differential from first within-plot selection were positive for all measured traits. Stem volume was observed as highest priority trait with coefficient weight for each trait were -0.0863 (height), 0.1426 (diameter), 0.2195 (stem straightness), 2.3025 (stem volume). Prediction of genetic gain from within-plot selection were 3,50% (height), 3.58% (diamater), 3.41% (stem straightness), 7.40% (stem volume). While the gain from family selection were 2.80%, 3.75%, 3.54%, 7.10% for height, diamater, stem straightness, stem volume, respectively.

Abstract

Sustainable forest management recently calls for growth information concerning integrated functions of ingrowth, upgrowth and mortality. This study was conducted in logging concessions of PT. Tunas Timber Lestari (TTL), PT. Wapoga Mutiara Timber (WMT) dan PT. Manokwari Mandiri Lestari (MML) in Papua. Then, this research was intended to build growth stands models in logged over forest. The data were obtained from permanent sample plots (PSPs) in three logging concession in Papua forest. Results revealed that characteristics of stand namely basal area, stem density and diameter had significant coefficients to model of ingrowth, upgrowth and mortality in each logging concession. Specifically, PT WMT showed the highest value of coefficient of determination (>80%, P<0.05). For PT MML only had significant model namely ingrowth and upgrowth model while PT TTL only shwed ingrowth model as significant equation.

Abstract

Gyrinops versteegii (Gilg.) Domke is including one of superior agarwood-producing plants and naturally growing in Eastern Indonesia as Nusa Tenggara and Papua. Indonesia has been trading agarwood products both domestically and overseas which one of them is agarwood produced by G.versteegii. This study purpose was to develop an in vitro culture method for mass propagation of G. versteegii. Shoot induction conducted on MS medium supplemented with Benzyl Amino Purine (BAP) 0.7; 1.0; 1.5; and 2.0 mg/l. The shoots multiplication conducted on MS medium supplemented with the best concentration of BAP from shoot induction phase. The rooting of shoots conducted on half strength MS medium supplemented with interaction of Napthalene Acetic Acid (NAA) 0.01 mg/l with concentration of Indole-3-Butyric Acid (IBA) 1.0; 2.0; 3.0; and 4.0 mg/l. Epicotyl explant with a given concentration level of BAP 0.7 mg/l produce the highest rates number of shoots and shoot length compared to other explant respectively 4.8 shoots and 0.41 cm within 6 weeks. The best explant developments in the best medium able to promote the growth of the length and number of shoots are 0.28 shoots and 0.3 cm within 4 weeks. Explants easiest, quickest and most high sprouting ability as a factor of success in terms of multiplication is epicotyl. The combination treatment of material explant with concentration of BAP only affect to growth of shoots length. The combination treatment of NAA with concentration of IBA has no effect against root formation and growth rootlength.

Abstract

One of the purposes of seedling seed orchard (SSO) establishment is to produce genetically improved seeds. The amount of genetic gain in the SSO is determined from a series of selection involving within and between family selection in which the gain is generally achieved from the accumulated increases of gain in successive advanced generation breeding cycles. In present study, the amount of genetic gain was predicted in third-generation SSO of Acacia mangium after first within-plot selection using a retrospective selection index’s method. The SSO was established in  Wonogiri, Central Java which was laid out in a randomized complete block design, 65 families, six replications, four trees-line plot and spacing of 2 m x 4 m. Actual within-plot selection was subjectively practiced in 40 months after planting. The observed traits for genetic gain prediction was height, diameter at breast height (dbh), and stem straightness  using data collected at 24 months after planting. For calculating genetic gain based on multiple trait selection index, coefficient weight for each trait was then derived from the selection differential. The results of study showed that the first within plot selection was practiced mostly for imposing dbh rather than height or stem straightness with the relative gain was 6% for dbh, and 3% for both height and stem straightness. Using the derived coefficient weight, the expected genetic gain by family selection was found to be slightly lower than those predicted by within-plot selection at 3% for dbh, and 2% for both height and stem straightness.

Abstract

Forest timber plantation of nyawai (Ficus variegate Blume) established by using vegetative propagated seedling from superior individu is very potential to develop for fulfilling the raw material demand of wood processing industries. The purpose of research was to get the optimum rooting medium and growth regulator concentration for rooting shoot cuttings of nyawai. Materials cuttings were provided from seedlings at 12 months age. The experiment was performed as factorial in a completely randomized design with two factors, i.e. rooting media factors (sand, mixed coconut coir dust + rice husk (2: 1, v/v), and mixed coconut coir dust+ husk rice + husk rice charcoal (6: 3: 1, v/v)) and IBA growth regulator concentrations (0, 50, 100,200,400, 600, and 800 mg/l), with three replications so that twenty experimented unit were used at each replication. Rooting percentage, root number, root length, shoot length, root biomass, shoot biomass, and leaf number were evaluated. Based on the result, sand media produced the highest rooted cuttings (88.10%), while the use of IBA showed the trend to increase the number of root with increasing the concentrations, but it influenced to reduce the shoot growth. Combination treatment of sand media and IBA 800mg/l provided the highest rooted cuttings (93.33%) with the number of root 21. Therefore, for the successful propagation of nyawai, cuttings are planted in sand media with IBA 800 mg/l.

Abstract

Kayu merah ( Pterocarpus indicus Willd) which listed as one of the IUCN Red List of Threatened Species in the category Vulnerable (VU A1D) is naturally distributed throughout Indonesia included Java, Sulawesi, Maluku, Bali, East Nusa Tenggara, West Nusa Tenggara and Papua. Random Amplified Polymorphic DNA (RAPD) molecular markers have been used to analyze the genetic diversity of six (6) natural populations of kayu merah from Seram Island, Manggarai Timur-Flores, Ngada-Flores, Soe-Timor Tengah Selatan, Kefamenanu-Timor Tengah Utara and Kupang. Ninety-nine (99) polymorphic loci were obtained from twelve RAPD primers. The mean of genetic diversity was 0.2024. Population with the highest genetic diversity was Soe (0.2925) and the lowest genetic diversity was Ngada (0.1212). The highest genetic distance among the populations was between Ngada and Kefamenanu (0.376), while the nearest genetic distance was between Ngada and Manggarai Timur (0.060). The informations resulted from this study are important to support the tree improvement and conservation programme of kayu merah.
