Abstract

BMPR-1B gene closely related to the rate of ovulation and follicular maturation. In poultry, BMPR-1B gene is located on chromosome 14th consists of 13 exons. BMPR-1B was expressed in the granulosa cells of the ovary and teca interna poultry. BMPR-1B was able to accelerate the maturation of the follicles so that the ova produced increasingly. BMPR-1B gene diversity was associated with poultry production.  DNA isolation was performed using GeneJET Genomic DNA Purification Kit from blood samples derived from 28 native chickens, 20 layer chickens and 10 arab layer chickens. A total of 58 DNA were  used as a template in the process of BMPR-IB gene amplification using Polymerase Chain Reaction method. Primers used in this study according to Zhang et al. (2008), Forward primer 5 'ATG GCT GGG AAG TCT GGA TG 3'dan Reverse primer 5' TGC CTT CTG TGT TAA CCG C 3 'with a length of 581 bp. This primary amplify BMPR-1B gene segment from exon 6 to exon 7 including the entire intron 6 with an annealing temperature of 58 0 C. BMPR-IB gene diversity was detected using restriction enzymes HindIII were incubated for 2 hours. The results of the study was showed that Hind III restriction enzymes do not cut the BMPR-1B gene segments in the population of native chickens, arab layer chickens and layer chickens, they were monomorphic.

Abstract

The concentration of high fat content in buffalo embryo culture media is very influential on the increase in oxidative stress that occurs in conditions of in vitro. The occurrence of increased oxidative stress in the process of embryo culture in vitro, would result in lower percentage of embryo culture until the morula stage (32) cells. The purpose of this study was to determine the percentage of fertilized oocytes supplemented with glutathione. To know the development of the embryo until the morula stage (32) cells were cultured in the culture medium supplemented with glutathione (GSH). To determine the viability of morula stage embryo or 32 cells. The materials used in this study is ovarian buffaloes and methods used in this study is an experimental method in the laboratory. Based on these results it can be concluded that the addition of glutathione 1.5 mM in media oocyte maturation buffalo in vitro provide a significant influence on the percentage of maturation ie          (P <0:01) or 62.5% and in line with the high percentage of oocytes matured in treatment increase glutathione 1.5 mM, then the percentage of oocytes were successfully fertilized also higher, namely 88.98%. Furthermore, the addition of 3 mM glutathione in embryo culture media in vitro buffalo give a significant influence on the percentage of embryo development to the morula stage or cell 32, namely (P <0.01) in, or 40.73%.

Abstract

Harvesting bananas will produce stems and stump that can be used as an alternative feed. One processing techniques tomaintain the nutritional value is silage. This study aims to determine the content of the fiber fraction ie. Neutral Detergent Fiber (NDF), Acid Detergent Fiber (ADF), lignin, cellulose and hemicellulose from bananas waste silage with the composition of the substrate and the different levels of molasses.  Research using a Completely Randomized Design factorial pattern with two replications. Factor A : the composition of the substrate, A1. stumps 100% :  stems 0%; A2. 50% stems : 50% stumps; A3. 0% stumps : 100% stems.  Factor B : the addition of molasses  0%; 2.5% and 5%. Data were analyzed using a completely randomized factorial design, the difference between treatments tested Duncan's Multiple Range Test (DMRT). The results showed banana waste silage with the composition of the substrate and molasses with different levels have moderate physical qualities and fiber fractions are NDF 57.86-89.63%; ADF 34.44-46.20%; lignin 11.75-17.19%; cellulose 15.10-32.34%; hemicellulose 11.66-55.18%. Silage that contains 100% of banana stem with the addition of 5% molasses is the best result.

Abstract

Gelatin is a compound hydrolysis of collagen fibers. The purpose of this research was to determine the influence of concentration of hydrochloric acid (HCl). and soaking time on the bovine hide making toward physical quality gelatin. The materials were bovine hide, hydrochloric acid and  distilled water. Data were statistically analyzed by A Completely Random Design with factorial pattern consist two factors; hydrochloric acid concentration (3% and 5%) and soaking time (4, 8, 12 and 16 hours) with three times replications. Parameters measured were pH, gel strength, yield and viscosity. The result showed that pH,  bloom strogh, yield and viscosity, centipoise (cP) of gelatin was 2.70-4.12, 53,33-185,00 g, 18.04-37.93% and 1.49-1,79, respectively. An interaction between hydrochloric acid and soaking time significantly (P˂0.01) influence in following categories: T pH, gel strength, viscosity and yield. It is conclude that the best combination treatments is  HCl 3% and 12 hours of soaking soaking.

Abstract

The study aimed to know the physical quality of pellets for finisher period added different feces and adhesive matter. The experiment design was randomized complety block design  factorial  (RCBD) with two factors and three replications. The first factor was feces (goat feces, cattle feces, buffalo feces) and the second factor was adhesive matter (tapioca flour, eeg white, sago flour). Physical observation were water content, specific gravity, bulk density, compressed bulk density, angle of repose, and the durability of pellet. The result showed that addition type feces influenced water content, specific gravity, bulk density, compressed bulk density, angle of repose and the durability of pellet form. The result showed that addition adhesive matter influenced bulk density, compressed bulk density, the durability of pellet form.Interaction the addition of cattle feces and different adhesive matter in water content, specific gravity, compressed bulk density, angle of repose and the durability of pellet. Interaction buffalo feces and tapioca flour best quality.
