Abstract

The objectives of this in vitro study were to investigate the hatching rate, the outgrowth diameter andthe activity of mitochondria Nicotiamide Adenin Dinucleotide Dyhidrogenage (NADH)-CoQ reductase ofblastocysts trophoblast cells from aging mice. Blastocysts of aging (age >12 months) and young productive(age 2 months) mice were collected from the cornua utery at day-4 of pregnancy and were cultured inmDMEM medium supplemented with 10% New Born Calf Serum (NBCS), 10% ITS, and 50 ?g/mlgentamicine, in 5% CO2 incubator at 37°C for 10 days. The blastocysts hatching rate and the trophoblastsmonolayer were examined for their diameter outgrowth and the NADH-CoQ reductase activity. The resultsshowed that the hatching rate, the trophoblast outgrowth diameter and the activity of NADH-CoQ reductaseof blastocysts collected from productive mice were significantly higher than those collected from the agingmice (P<0,05). It can be concluded that the impairment of blastocysts implantation especially, in agingmice were caused by the low activity of the NADH-CoQ reductase that play important role in energyproduction needed for the hatching and trophoblast outgrowth.

Abstract

The aim of this study is to compare glycoconjugate expression in the lymphonodes between bali cattleexperimentally infected with jembrana virus disease and non infected cattle. The biotinylated lectinsused in this study were Concavalia ensiformis (ConA), Arachis hypogaea (WGA), Triticum vulgaris (PNA),Ulex europeaus (UEA-1), and Sambucus nigra (SNA), respectively. The expression of lectin bindingglycoconjugate exhibit differences between the lymphonodes of infected and non infected cattle. Con A,WGA, and SNA were found be bound to lymphocytes whereas in infected cattle it was less intensive,repectively. More over, PNA and UEA were found strongly bound to the either lymphocytes of eitherinfected and non infected cattle.

Abstract

Consensus sequence of hemagglutinin (HA) gene of avian influenza viruses of H5N1 subtype isolatedfrom fowl in Indonesia – hereafter named as H5N1_Indonesia – is compared with that of H5N2 and H5N9viruses. Sequence information were downloaded from the public database GeneBank. The genetic distancesand nucleotide as well as its deduced amino-acid sequence alignment were analysed. At nucleotide level,genetic distances of HA between H5N1_Indonesia to H5N2 and H5N9 are 16.2% and 9.6%, respectively.At amino-acid level, the distances are 9.7% and 6.8%. The genetic distances of HA1 fragments are 19.0%(H5N1_Indonesia – H5N2) and 10.9% (H5N1_Indonesia – H5N9). At amino-acid, level the genetic distancesof HA1 are 13.1% (H5N1_Indonesia-H5N2) and 8.8% (H5N1_Indonesia – H5N9). All three subtypes havedifferent glycosilation motive and variation of amino-acid sequence of four antigenic sites. The consequenceof those facts is discussed.

Abstract

Chicken taxonomy and pedigree investigation are important parts to understand the domesticationprocess occurred on chicken in Indonesia. The Indonesian native chickens have very different clade with thechickens from other Asian countries and others. This study was conducted to construct phylogeny junglefowls (red and green jungle fowls) and Indonesian native chickens, in order to know their relationship.Among the available DNA marker, the region D-loop on DNA mitochondria is the most effective markerused in the investigation. Mitochondrial DNA D-loop (hypervariable-1 segment) was PCR amplified andsubsequently sequenced for a total 33 individuals of green jungle fowls (Gallus varius), 9 individuals of redjungle fowls (Gallus g. gallus) from Indonesia, and 30 individuals of Indonesian native chickens (Lombok,Cemani, Kedu, Kedu Putih, Nunukan, Kate, Pelung, Gaok, Merawang, dan Sentul). Seventy two (72)sequences were used for analysis. Seven (7) published reference D-loop sequences of genus Gallus fromGenBank were also included in the analysis: Gallus varius (GenBank accession number D64163 danD82912), Gallus gallus (GenBank accession number AB098668), G. gallus spadiceus (GenBank accessionnumber AB007721), Gallus gallus bankiva (GenBank accession number AB007718), Gallus lafayetti(GenBank accession number D66893), and Gallus sonneratii (GenBank accession number D66892). Phylogenyanalysis indicates that Indonesia jungle fowls can be grouped into two clades (clades of red jungle fowls/Indonesia native chicken and green jungle fowls). Monophyletic phylogeny trees of jungle fowls from Indonesiawere shown in this study for pedigree investigation of domesticated chickens.

Abstract

A study to find out the histological structures of small intestine (ie. duodenum, jejunum, and ileum) of bali cattle have been carried out. A total of 20 small intestines were collected from the abattoir at Pesanggaran, Denpasar Bali. Histological examinations were performed on Haematoxyline Eosin stained small intestines slides. The duodenum, jejunum and ileum of the small intestines have four mucosa membranes: i) mucosa membrane, ii) sub mucosa membrane, iii) muscular membrane and, iv) serous membrane. The average thickness of mucosa membrane of duodenum, jejunum and ileum was 36,3±13,5 mm; 29,9±3,1 mm; and 38,8±11,9 mm, respectively. Each part has villi with an average length of 27,5±8,3 mm; 20,1±3,7 mm; and 18,5±4,3 mm, respectively. This mucosa membranes consist of single columnar epithelium, muscularis mucosa, and lamina propria. The average thickness of sub mucosa membrane was 47,3±15,3 mm; 10,4±2,6 mm; and 16,9±5,6 mm, respectively. Sub mucosa membrane consisted of connective tissue. The average thickness of muscular membrane was 46,9±8,8 mm; 28,1±5,1 mm; and 62,4±11,3 mm, respectively, which consisted of circular and longitudinal smooth muscle. The average thickness of serous membrane was 19,9±3,1 mm; 11,9 ±3,0 mm; and 12,1±3,6 mm, respectively, which consisted of mainly connective tissue. Goblet cells were seen through all the epithel of small intestines with the highest number seen in the ileum. Specific structure of duodenum, jejunum, and ileum which is Brunner’s glands in the sub mucosa membranes of duodenum, circular plicae in the jejunum and mesenteric gland in the ileum were also observed.

Abstract

The sequence of the Non-Coding Region (NCR) and Coding Region (CR) of 5’-terminal cRNA of thepolymerase basic 1 (PB1) gene as a major factor for the species adaptation of avian influenza virussubtype H5N1 (AIV H5N1) has been analysed. The information could be a virological signal for theemergence of a new strain with pandemic potential. Total RNA from twenty six (26) avian influenzasubtype H5N1 isolates were amplified using reverse-transcriptase-polymerase chain reaction (RT-PCR)with a universal forward primer for influenza virus and specifically designed backward primers. Fifteen(15) PB1 gene fragments could be amplified. RT-PCR products were sequenced and analyzed using Mega4software. The length of NCR of PB1 gene was found to be 24 bases and mostly shows conserved sequence,with an exception of Dk/Badung/2006 isolate which has C-7T substitution. A/T composition of PB1 NCRwas 54,2%, while the Dk/Badung/2006 isolate was 58,3%. Abstract

The objective of this study is to observe the anatomical structure of horse hooves collected from the Laboratory of Anatomy FKH IPB. Twenty five hoof specimens, consisting of ten fore hooves and fifteen hind hooves were used as research materials. The external morphology such as color, angle, structure and condition of the hoof wall were described. The observation on external morphology showed that the hooves have two basic colors, black and white. Generally, all of the hoof specimens showed abnormalities in such aspect as angle, structure and condition of the wall. The structures of fore hoof and hind hoof from this study are classified into 8 categories, they are: flat foot, flared foot, knol hoef, fever rings, sand crack, club foot, contracted foot and bull nosed foot.

Abstract

Study on the morphometric characteristic of sambar deer (Rusa unicolor) which will be used as a baseline of growth trait selection had been conducted. The aim of this study is to set up criteria for better selection of sambar deer progeny. Morphometric characteristic observed in this study including : body weight, body length, chest width, chest girth, head length, head width, ear width, and ear length respectively. Result indicated that chest girth correlates significantly with the body weight (y=-108.004+1.875x). In conclusion chest girth can be used as a criteria for selection of growth trait of sambar deer.
