Abstract

ABSTRACT The management of Type II Diabetes Mellitus with prolonged drug therapy might affect the life qualities of patients and lead to Drug Related Problems (DRPs). Several types of DRPs frequently occur and they result in sub-optimum treatment, increased side effects and expensive costs. Objective of this study was to know about the uses of anti-diabetic drugs, prevalence of DRPs, the numbers of DRPs cases and the factors causing DRPs having the potentials to affect the therapeutic effectiveness for Type-II Diabetes Mellitus treated at Semarang Tugurejo Hospitals during the period of 2007-2008. The possible factors that might lead to DRPs were inaccuracy in the drug selection and sub-therapeutic. Non experimental, retrospective and non analytical observational study design was done. The population for this study consisted of 43 Type-II Diabetes Mellitus patients hospitalized in Tugurejo Hospital, Semarang. Sample collected by retrospectively from Hospital’s medical records of the 2007-2008 period in the whole population. The results showed there were 23,3% of the patient experienced DRPs which had the potentials to affect the therapeutic effectiveness for Type-II Diabetes Mellitus. Eleven cases of inaccuracies in selecting the drugs affected the effectiveness of the therapies. These cases of inaccuracies consisted of 5 cases of ineffective drugs, 4 cases of effective but expensive drugs, 2 cases of unnecessary combinations of drugs and one case of sub-therapeutic dosages. Abstract

ABSTRACT Dengue haemorragic fever is a disease caused by dengue virus by mediator Aedes aegypti. The effort of overcoming this disease could be done by preventing the mosquito from bitting using repellent. One of natural repellents is neem (Azadiracta indica A.Juss). This study aimed to determine the repellent activity of chloroform fraction of ethanolic extract neem leaf against Aedes aegypti mosquito. Extraction neem leaf done using ethanol 96% by maceration methode and followed fractionation with chloroform. This study was conducted in 5 groups, positive control (DEET 13%), negative control (liquid paraffin) and three series concentrations of chloroform fraction of ethanol extract neem leaf; they were 5%, 10% and 20%. Repellent activity was carried out by counting number of mosquitoes alighted. The amount of repellent activity was obtained from the protection value which was then analyzed using two-ways Anova. The EC90 value gained from probit analysis at significance of 95%. Identification of chemical compound of chloroform fraction of ethanolic extract neem leaf was done using thin layer chromatography (TLC). The results showed that chloroform fraction of ethanolic extract neem leaf had repellent activity against Aedes aegypti mosquito. The average protection value for 6 hours in positive control group and chloroform fraction of ethanolic extract neem leaf at 5%, 10% and 20% concentration were 92,43%, 66,76%, 76,90% and 88,35%. The 6-hours treatment had caused decrease in protection value in all of treatment groups. The effective concentration of chloroform fraction ethanolic extract neem leaf to repel 90% population of Aedes aegypti mosquito for 6 hours treatment was 26,1%. Abstract

ABSTRACTBreast and cervical cancers were the high prevalency, both in the world and Indonesia. This study aimed to determine the cytotoxic activities and its potentcy of n-hexane fraction of ethanol extract alfalfa herb (Medicago sativa L.) in T47D cells and HeLa cells, also identificated its chemical compounds.Extraction was done using ethanol by soxhletation method and then followed fractionation with n-hexane. Cytotoxic assay to get the value of IC50 was carried out by using MTT assay with a series of concentration of fraction 62,5; 125; 250; 500; 1000 μg/ ml. The cytotoxic activities was determined with viability cells from absorbance ELISA reader. Identification of chemical compounds of n-hexane fraction of ethanol extract alfalfa herb was done using chemical reactan and paper chromatography and then was evaluated qualitatively.The results showed that the n-hexane fraction of ethanol extract of alfalfa herb had cytotoxic activities in T47D cells and HeLa cells. The IC50 values were 523,9 μg/ml in T47D cells and 503,5 μg/ml in HeLa cells. Abstract

ABSTRACTAlfalfa plant (Medicago sativa L.) contains of flavonoids compound, it has already known. One of the effects of flavonoids is anti-inflammatory effect of alfalfa herb (M. sativa L.). The purpose of this research to know about anti-inflammatory effect of alfalfa herb ethanolic extract on carragenan induced male rats and also to know the content of its active compound. Thirty male wistar rats were randomly divided into 5 group of 6. Group I, negative control, were treathed CMC-Na 0,5 %, group II, positive control, were treathed natrium diclofenac 6,3 mg/ kg BB. Group III, IV and V were treathed ethanolic extract of alfalfa herb orally dose 45 mg/ kg BW, 90 mg/ kg BW and 180 mg/ kg BW. Anti-inflammatory effect were measured of edema feet were done every 30 minutes for 6 hours using plestismometer. After edema feet data vs time got, then we can get AUC0-360 value and percentase anti-inflammatory effect. Statistic analytical used one way ANOVA (p<0,05) and continued with Tukey test then. Identification of active compound of alfalfa herb ethanolic extract was used Thiny Layer Chromatography (TLC). The result of the research shown that ethanolic extract of alfalfa herb by dosage 45 mg/ kg BW, 90 mg/ kg BW and 180 mg/ kg BW could reduce edema in carrageenan induced rats, significantly. The percentase of anti-inflammatory effect on dosage of 45 mg/ kg BW is 58,99 %, on 90 mg/ kg BW is 72,64 % and on 180 mg/ kg BW is 64,23 %. TLC result shown that active compound the ethanolic extract of alfalfa herbs was flavonoids. We could be concluded that ethanolic extract of alfalfa herb have anti-inflammatory effect. Abstract

ABSTRACT Breast cancer is the prime cause of women death in various parts of the world.  Many research  to get breast cancer drug has been widely applied. The aims of    this study were to determine the cytotoxic effect of ethyl acetate fraction of ethanolic extract of the beet root  (Beta vulgaris  L.  var. rubra  L.) on T47D cell line which was the breast  cancer cell and test their chemical compound. The  extract was made by macerare method, and than it was fractionated by using ethyl acetate. Cytotoxicity test using MTT assay method with concentration 500; 250; 125; 62,5; 31,25 µg/ml. Observation was done  to get the viability cell absorbance by using ELISA reader. Analytical data carried out  toward percentage viability T47D cell line  to determine a value IC50.  The chemical compound  tests done through a process of preliminary test with a chemical reagent and thin layer chromatografi. The chemical compound were qualitative analysis. The result showed that ethyl acetate fraction of ethanolic extract beet roots had cytotoxic effect against T47D cell line with IC50 253,86 µg/ml. The chemical compound of the ethyl acetate fraction of ethanolic extract of beet root were flavonoids. Abstract

ABSTRACTTherapy failure of breast cancer as a consequence of cell resistention has encouraged the effort to develop a cancer medicine including herbal medicine. Based on some research, it can be concluded that there is antioxydant and anticancer activity from beetroot involved with betasianin and betasantin substance that belong to flavonoid compound. The purpose of this research was to find the effect cytotoxic of methanolic extract of beetroot (Beta vulgaris L. var. rubra L.) on cell line T47D which was a breast cancer cell.This research was conducted on several experiment step namely plant determination, cell preparation, cell harvest, methanolic extraction, and cytotoxic test. The extraction of beetroot is conducted by maceration method using methanol 70 %. Cytotoxic test on cell line T47D is conducted by MTT assay using five concentration ceries were 500, 250, 125, 62,5 and 31,25 ug/ml. Data analysis uses probit analysis.Research result shows that methanolic extract of beetroot there was no cytotoxic activity on cell line T47D. It shows that five consentration series methanolic extraction of beetroot have % cell viability more than 100%.Keyword: Beetroot, Beta vulgaris L. var. rubra L, Cell line T47D, Cytotoxic, Flavonoid.

Abstract

ABSTRACTHaloperidol and chlorpromazine antipsycotic were used for broad and so long time. That drugs used could shown undesire side effect. Objective of this study was to investigate haloperidol and chlorpromazine side effect, especially was to know relative risk (RR) of extrapiramidal and hypotension orthostatic side effects of haloperidol and chlorpromazine antipsycotic used in Dr. Aminogondohutomo Mental Hospital durimg 2005. Non experimental, retrospective and Case Control study design was done. Population of this study was patient ward in Dr. Aminogondohutomo Mental Hospital durimg 2005. Data was taken retrospectively from medical record. Analytical of data used Chi Kuadrat and Odds Ratio (OR) was to know relative risk (RR) of extrapiramidal and hypotension orthostatic side effects of haloperidol and chlorpromazine antipsycotic used. The results showed there were 27,17% extrapiramidal side effects of haloperidol used and 15,24 % hypotension orthostatic side effects of chlorpromazine used. That were the first rank of side effect in every that antipsycotic.The value of relative risk (RR) of extrapiramidal side effects of haloperidol used was 5 and 3 for hypotension orthostatic side effects of chlorpromazine used. That meaned patient who used haloperidol could 5 more time got extrapiramidal side effects than haloperidol unused and patient who used chlorpromazine could 3 more time got hypotension orthostatic side effects than chlorpromazine unused. Abstract

ABSTRACTDrug formularium is a method used by hospital medical staff to evaluate and choose a drug for patient treatments from various available drug trade name preparations within hospital. Objective of this study was to evaluate prescription compliance on drug cards of Kutilang in-patients ward towards Dr Kariadi Hospital Formularium during 2007. Non experimental, retrospective and non analytical observational study design was done. Sample collected by a proportional stratified random sampling from Kutilang in-patient drug card during 2007. Data were compared to Dr Kariadi Hospital Formularium. When prescription of drugs trade name different with formularium but generic name available, it was considered as non-formularium A and non-formularium B if either trade name or generic name drugs were not present. The results showed there were 56.1% female patients and 43.9% male patient hospitalized in Kutilang ward during 2007, in which 30.4% was belong to Internal medicine patients. Total number of drugs used were 613 items consist of 46.8% generic drugs and 53.2% trade name (patent) drugs. Overall agreement of drug used to the formularium was 77.7%. The rank of drug prescription agreement were as follows, anesthesiologist and psychiatry both were 100% agree to formularium, cardiologist was 14.8% agree to non-formularium A and dermato-venerologist was 40.9% belong to non-formularium B. The higest rank of prescription based on treatment classes were immunosuppressant (92.9%) among formularium, anti diabetics (32.2%) among non-formularium A and ENT drugs (83.3%) among non-formularium B. Abstract

ABSTRACT Soursop leaves ethanol extract (SLEE) has been shown to lower blood levels of total cholesterol. However, it not yet investigated the effects on triglyceride and HDL levels. There is also no research on combining related soursop leaves with gemfibrozil. The purpose of this study is to compare the efficacy of combination therapy SLEE and gemfibrozil to SLEE or gemfibrozil single treatment on decreasing triglyceride levels and increase HDL level in rats induced by high-fat feeding. Thirty male Wistar rats were divided into six groups (normal control, hyperlipidemia control, single gemfibrozil, single SLEE, 1st combination, and 2nd combination groups). Each group treated a high-fat feed two times a day for 14 days, except for the normal control. The test material was treated one time a day for five days. Measurement levels of triglycerides and HDL used GPO-PAP reagent and Direct Enzymatic Colorimetric Test. The data was analyzed by One-Way ANOVA test followed by Tukey's test. The results showed that SLEE can decrease triglyceride’s level and increase HDL levels of male Wistar rats. The combination of SLEE and gemfibrozil also can decrease triglyceride’s level and increase HDL level. However, there are no significances different between the efficacy of combination SLEE and gemfibrozil than SLEE or gemfibrozil single treatment in decreasing triglyceride's level and increase HDL levels of male Wistar rats. Abstract

ABSTRACT Soursop leaves can be used as a drug hypercholesterolemia. Simvastatin is one of the drugs that can be used to lower total cholesterol and LDL. This study aims to determine the effectiveness of combination therapy EEDS and simvastatin in inhibiting elevated levels of total cholesterol and LDL induced rats a high-fat feed when compared with simvastatin alone or single EEDS. This study is experimental with post-test only control group design. Using the test animals were 30 male rats Wistar strain were divided into 6 groups. Test animals fed a high-fat twice daily for 14 days except for the normal control group, the test preparation administration once a day for 5 days. Measurement of total cholesterol using  the reagent kit CHOD-PAP and LDL measurement method  Direct Enzymatic Colorimetric Test with a spectrophotometer UV-Visibel. Data were statistically analyzed by One Way Anova followed by Tukey test. The results showed that administration of EEDS can inhibit LDL levels induced rats a high-fat feed. eeds combination and simvastatin can inhibit the increase in total and LDL cholesterol levels did not differ significan (p> 0.05), which means it has the same ability in lowering total cholesterol and LDL induced rats a high-fat feed simvastatin compared with administration of single and single EEDS.
