Abstract

Gynura procumbens has a potency to produce raw material for pharmaceutical industry. Liquid culture systems have significant effects on multiplication rates of organ, so the propagation technology to increase biomass of this plant or organ using the liquid culture system was necessary. This research was conducted to compare biomass production of adventitious roots of G. procumbens in different liquid culture (shake flask, temporary immersion bioreactor and balloon-type bubble bioreactor). Adventitious roots culture was maintained in Murashige and Skoog liquid medium supplemented with IBA (indole butyric acid) 5 mg/L and different concentrations of sucrose (10, 30, 50 g/L). Result showed that the highest biomass production (fresh weight) was in shake flask that was 3.9-fold higher than initial explant, while in temporary immersion system was 5.12-fold higher than initial explant, and in balloon- type bubble bioreactor was 13.1-fold higher than initial explant. The highest enhancement of adventitious roots was occurred at sucrose supplementation of 50 g/L. Based on this research we found that the best type of liquid culture to increase biomass of G. procumbens adventitious roots was balloon-type bubble bioreactor. Scaling-up of adventitious root culture are necessary to fulfill the need of raw material for pharmaceutical industry and ballon-type bubble bioreactor was the most suitable method to scale up the performance.

Abstract

ABSTRAK Cabai Jawa (Piper retrofractum Vahl.) dikenal sebagai tanaman hias dan tanaman obat. Metabolit sekunder pada tanaman ini adalah piperin, saponin, kavisin dan minyak atsiri. Metabolit sekunder  tersebut dapat diisolasi dari bahan tanaman atau kalus hasil kultur jaringan tanaman. Pada metode kultur jaringan  tanaman untuk menginduksi kalus diperlukan media dengan konsentrasi zat pengatur tumbuh yang tepat untuk mendapatkan hasil yang optimal. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi konsentrasi zat pengatur tumbuh Naphthalene Acetic Acid (NAA) dan 6-Benzyl Amino Purin (BAP) yang paling baik untuk induksi kalus P. retrofractum. Jenis penelitian ini adalah eksperimental laboratoris. Penelitian menggunakan rancangan acak lengkap (RAL) dengan 17 perlakuan yang terdiri atas 16 perlakuan kombinasi zat pengatur tumbuh dan 1 perlakuan kontrol, Setiap perlakuan terdiri atas  6 ulangan. Eksplan daun P. retrofractum ditumbuhkan pada medium Murashige dan Skoog padat ditambah  dengan zat pengatur tumbuh dengan konsentrasi masing-masing 0; 0,5; 1; 1,5; 2 mg/L. Data yang diperoleh  berupa data kuantitatif dan kualitatif. Data kuantitatif meliputi lama waktu induksi kalus, persentase eksplan membentuk kalus, berat segar kalus dan berat kering kalus, dianalisis secara statistik dengan SPSS. Data kualitatif meliputi warna dan tekstur kalus. Hasil penelitian menunjukkan bahwa zat pengatur tumbuh NAA dan BAP berpengaruh terhadap pertumbuhan eksplan daun P. retrofractum. Penambahan kombinasi konsentrasi NAA 0,5 mg/L dan BAP 0,5 mg/L menunjukkan respon terbentuknya kalus paling cepat yaitu 11,5 hari. Penambahan kombinasi konsentrasi NAA 1 mg/L dan BAP 0,5 mg/L menghasilkan berat segar terbaik  yaitu 70,6 mg, sedangkan pada kombinasi konsentrasi  NAA 1 mg/L dan BAP 2 mg/L menghasilkan berat kering terbaik  yaitu 18 mg. Warna kalus adalah putih dan putih kecokelatan dengan tekstur friabel dan kompak.Kata kunci: BAP, induksi kalus, NAA, Piper retrofractum Vahl.ABSTRACT Chili Java (Piper retrofractum Vahl.) is known as ornamental plants and medicinal plants. Secondary metabolites in this plant are piperin, saponin, kavisin and essential oils. Secondary metabolites can be isolated from plant material or callus from plant tissue culture. In plant tissue culture method to induce callus required media with the growth regulator concentration to get optimal result. The aim of this research is to know the effect of the combination of growth regulator of Naphthalene Acetic Acid (NAA) and 6-Benzyl Amino Purin (BAP) which is best for the induction of P. retrofractum callus. The type of this study was laboratory experimental. The study used a complete randomized design (RAL) with 17 treatments consisting of 16 treatment combinations of growth regulators and 1 control treatment. Each treatment consisted of 6 replications. P. retrofractum leaf eksplan grown on Murashige and Skoog solid medium coupled with growth regulator substances with respective concentrations of 0; 0.5; 1; 1.5; 2 mg / L. The data obtained in the form of quantitative and qualitative data. Quantitative data include duration of callus induction, percentage of callus form explants, fresh callus weight and dry weight of callus, analyzed statistically with SPSS. Qualitative data include callus color and texture. The results showed that NAA and BAP growth regulator effect on growth of P. retrofractum leaf eksplan. The addition of a combination of NAA concentration of 0.5 mg / L and BAP 0.5 mg / L showed the fastest callus formation response of 11.5 days. The combination of NAA concentration of 1 mg / L and BAP 0,5 mg / L resulted in the best fresh weight of 70.6 mg, while in combination NAA concentration 1 mg / L and BAP 2 mg / L yielded the best dry weight of 18 mg. Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh kombinasi konsentrasi zat pengatur tumbuh IBA (Indole Butyric Acid) dan BAP (6-Benzyl Amino Purine) yang paling baik untuk induksi kalus sirih hitam (Piper betle L.). Penelitian ini menggunakan rancangan acak lengkap dengan 25 perlakuan dan setiap perlakuan memiliki 6 ulangan sehingga terdapat 150 unit ekperimen. Pada tahap kultur kalus dilakukan dengan menambahkan zat pengatur tumbuh IBA dan BAP ke dalam medium Murashige and Skoog (MS). Hasil uji tersebut menunjukkan bahwa zat pengatur tumbuh IBA dan BAP dengan kombinasi konsentrasi berbeda berpengaruh terhadap waktu induksi kalus, berat segar dan berat kering kalus sirih hitam. Hasil penelitian menunjukkan waktu tercepat pembentukan kalus pada IBA 2,0 mg/L dan BAP 2,0 mg/L yaitu 10 hari. Berat segar dan berat kering tertinggi pada IBA 2,0 mg/L dan 2,0 mg/L yaitu 0,8507 gram untuk berat segar dan 0,0769 untuk berat kering. Warna kalus adalah putih kehijauan dengan tekstur kompak dan remah.Kata kunci: Induksi kalus, Piper betle L., Indole Butyric Acid, 6-Benzyl Amino Purine.ABSTRACTThe purpose of this research to determine the effect of the combination concentration of growth regulators IBA (Indole Butyric Acid) and BAP (6-Benzyl Amino Purine) was best for callus induction black betel (Piper betle L.). This research used completely randomized design with 25 treatments and 6 replicates of each treatment, hence there were 150 experimental units. At this stage of callus culture was done by adding the growth regulators IBA and BAP into Murashige and Skoog (MS). The test results showed that plant growth regulators IBA and BAP in combination with different concentrations of influence on callus induction time, fresh weight and dry weight callus Piper betle L. The results showed the fastest time of callus formation at IBA 2,0 mg/L and BAP 2,0 mg/L at 10 days. Fresh weight and dry weight of the highest in the IBA 2,0 mg/L and BAP 2,0 mg/L were 0,8507 grams and 0,0769 grams fresh weight to dry weight. The color of callus was white greenish with compact and friable texture.Keywords: Callus induction, Piper betle L., Indole Butyric Acid, 6-Benzyl Amino Purine.

Abstract

Exposure to cadmium (Cd) could increase of reactive oxygen species (ROS) and changes in expression of antioxidant genes. Gynura procumbens is a medicinal plant that is rich in phenolic and flavonoid compounds. The aimed of study to evaluate the hepatoprotective effect of G. procumbens adventitious root (GAR) extract against Cd toxicity, especially expression rate of hepatic antioxidant genes. Twenty-five male mice were treated as follows: P1 (control), P2 (Cd100mg/L), P3 (GAR100mg/L + Cd100mg/L), P4 (GAR200mg/L + Cd100mg/L), and P5 (GAR300mg/L + Cd100mg/L). The samples (blood and liver) were collected for analysis of malondialdehyde (MDA) levels, superoxide dismutase (SOD) and catalase (CAT) activities, and their relative gene expression were determined. The hematological assay showed Cd-treated administered with GAR extract increased the number of red blood cells (RBC), haemoglobin concentration (HGB), hematocrit (HCT), mean corpuscular volume (MCV), and mean corpuscular haemoglobin (MCH), but reduced the level of alanine aminotransferase (ALT) and aspartate aminotransferase (AST). In addition, the GAR extract decreased the MDA production, but increased the activities of SOD and CAT. These enzymatic activities were positively correlated with their respective gene transcripts. Our study revealed that GAR extract administration showed marked hepatoprotective effects against Cd-induced oxidative stress.

Abstract

AbstrakPiper betle L. var Nigra (sirih hitam)  merupakan salah satu jenis tanaman obat yang berpotensi untuk dikembangkan dengan metode kultur jaringan. Penelitian ini bertujuan untuk mengetahui pengaruh variasi zat pengatur tumbuh Indole Acetic Acid (IAA), Benzyl Amino Purine (BAP), Kinetin terhadap senyawa metabolit sekunder  yang terdapat kalus sirih hitam. Eksplan daun dari sirih hitam ditanam pada medium Murashige dan Skoog dengan 4 perlakuan kombinasi (I1,0K1,0; I1,0K1,5; I1B1,5; I0,5B0,5). Kalus dipelihara selama 8 minggu. Simplisia kalus sirih hitam di maserasi menggunakan pelarut metanol dan diidentifikasi menggunakan Gas Chromatography Mass Spectra (GCMS). Berdasarkan analisis GCMS menunjukkan bahwa senyawa metabolit sekunder dengan presentase tertinggi terdapat pada masing- masing perlakuan yaitu senyawa gamma sitosterol (14,88%) pada perlakuan I1,0K1,0 ; senyawa 14-Beta H Pregna (15,94%) pada perlakuan I1,0K1,5 serta senyawa beta-d-glucopyranoside (15,54%) pada perlakuan I0,5B0,5; dan senyawa beta-d-glucopyranoside (5,63%) pada perlakuan I1,0 B1,5. Katakunci: Kalus, Piper betle L. var Nigra, senyawa metabolit sekunder  AbstractPiper betle L. Var Nigra (black betel) is one type of medicinal plant that has the potential to be developed by tissue culture method. This study was aims to determine the effect of variations in growth regulator Indole Acetic Acid (IAA), Benzyl Amino Purine (BAP), and Kinetin on secondary metabolite compounds contained in black betel callus. The explant of black betel’s leaf was cultured on Murashige and Skoog medium with four combination treatments (I1.0K1.0; I1.0K1.5; I1B1.5; I0.5B0.5). The callus was maintained for eight weeks. Simplisia of black betel’s callus was macerated using methanol solvent and identified using Gas Chromatography Mass Spectra (GCMS). GCMS analysis showed that secondary metabolites with the highest percentage in each treatment were gamma sitosterol compound (14.88%) in treatment I1.0K1.0; 14-Beta H Pregna compound (15.94%) in I1.0K1.5 treatment and beta-d-glucopyranoside compound (15.54%) in I0.5B0.5 treatment; and beta-d-glucopyranoside compound (5.63%) in I1.0 B1.5 treatment.Keywords: Callus, Piper betle L. Abstract

Background: Tentena City is one of the tourist centers with rapid economic growth located in Poso Regency because Poso Lake is very famous and able to change the economic level of the community. But there are still 20% of people who live very poor and 60% of the lower middle class and the remaining 20% of the middle to upper middle. Through the youth community Pukat Tentena is expected to foster and encourage millennials to develop vegetable and fruit entrepreneurs using more modern cultivation techniques. Objectives: empowering the youth community in Tentena city, Pamona Puselemba Subdistrict, Poso Regency, Central Sulawesi with entrepreneurial tomato fruit products and melons produced by hyrdoponik drip irrigation system. Methods: This community service activity was carried out in Tentena village, Pamona Puselemba Subdistrict, Poso Regency, Central Sulawesi Province with TENTENA PUKAT Youth Community partners starting from May to September 2021. Results: Mitra has successfully cultivated tomato and melon plants with hydroponic drip irrigation system techniques in green houses until they are obtained salesable results. The average number of tomatoes per stalk is 4.05 and the average fruit diameter is 5.1 cm; While the average weight of melons is 1.2 -1.8 kg. Conclusion: The results of this community service activity provide benefits for partners, namely providing additional knowledge about how to cultivate tomatoes and melons with hydroponic drip irrigation system techniques and increase partner income by selling tomatoes and melons from hydroponic cultivation.

Abstract

Lead induced oxidative stress contributes to increase the productivity of reactive oxygen species (ROS) and to disrupt the antioxidant balance. Gynura procumbens adventitious root (GPAR) methanol extract contains abundant phenolic and flavonoids compounds as antioxidants and can be used as traditional medicinal plants. The objective of this study is to evaluate the protective effect of GPAR against lead acetate toxicity in mice to haematological parameter, histological of hepatic cells, and activities of antioxidant enzymes. The data obtained from five groups of treatment: P1 (control), P2 (Pb acetate-100 mg/L), P3 (GPAR-100 mg/L + Pb acetate-100 mg/L), P4 (GPAR-200 mg/L + Pb acetate-100 mg/L), P5 (GPAR-300 mg/L + Pb acetate-100 mg/L). The results indicated that administration of methanol extract of GPAR can prevent the decreasing of haematological parameter, maintain the percentage of normal hepatic cells, activities of superoxide dismutase (SOD), and catalase (CAT) due to lead acetate treatment. The effective dose of GPAR extract was 300 mg/L. This study provides that methanol extract of G. procumbens adventitious root exerts protective effects against lead acetate toxicity in mice.

Abstract

Gynura procumbens has been widely used to treat several health disorders, such as hyperglycemia, hypertension, diabetes, and allergies. This study aimed to determine the flavonoid content of G. procumbens cultured in a balloon-type bubble bioreactor using various sucrose concentrations (10, 30, and 50 g/L) and explant densities (5, 10, and 15 explants). The plant parts used were stem nodes of G. procumbens, grown in a balloon-type bubble bioreactor using Murashige and Skoog (MS) liquid media with IAA 2 mg/L and BAP 4 mg/L. The total flavonoid content was measured using a UV-Vis spectrophotometer (λ= 510 nm), and the flavonoid profile was tested using thin-layer chromatography (TLC). The administration of 10 g/L sucrose produced the highest total flavonoid production. In the treatment of explant density, the most increasing total flavonoid production was obtained in the treatment of 5 explants. The TLC results showed that each treatment forms spots like standard. This study proved that sucrose and explant densities culture on balloon-type bubble bioreactor method had optimized the production of flavonoids of plant shoots of G. procumbens culture.
