Abstract

Xylanase is extracellular enzyme that hydrolyzes xylan into xylose and xylooligosaccharides. The bacteria that produce the xylanase enzyme, namely SSA 2 isolate from the Sapan River Aro Solok Selatan hot spring, has a temperature of 75ºC and a pH of 8 or alkaline. Thermophilic bacteria can produce xylanase enzymes under optimum environmental conditions. Environmental optimization can be done by optimizing media composition, dissolved oxygen content, aeration, agitation, starter concentration, pH and temperature. This study aims to determine the specific activity of the xylanse enzyme at the starter concentration. The starter concentrations used were 2%, 4%, 6%, 8% and 10%. The highest specific activity of the xylanse enzyme was at a starter concentration of 4% (0.916 U/mL) this indicates that a low starter concentration can lead to good microbial growth.

Abstract

Abstrak. Enzim xilanase merupakan enzim extraseluler yang mampu menghidrolisis xilan menjadi xilosa. Isolat SSA 2 merupakan isolat dari bakteri termofilik hasil isolasi dari air panas sapan sungai aro, yang memiliki kemampuan menghasilkan enzim xilanase. penggunaan xilanase merupakan salah satu alternative mengurangi pemakain clorin dalam proses pemutihan pulp. xilanase diamobilisasi dengan menggunkan NA alginate. pulp difermentasi dengan suhu 50ºC, 55ºC, 60ºC, 65ºC, 70 ºC, 75 ºC, 80ºC, selama 6 jam, suhu optimum yang menghasilkan bilangan kappa terendah dan aktivitas enzim yang tertinggi pada suhu 60ºC.

Abstract

This study aims to determine the effect of the mindmap-assisted STEM approach on students' Higher Order Thinking Skills in learning biology class X SMA Negeri 4 Kerinci. This research is experimental research with a group pretest-posttest design model. The population in this study came from students of class X MIPA SMA Negeri 4 Kerinci in the academic year 2021/2022. The research sample was students of class X MIPA 2 and Class X MIPA 4. The sampling technique used was the random sampling technique. The data collection technique in this study was in the form of an objective test consisting of 20 HOTS questions that had been validated by experts. Data analysis is quantitative data analysis with tests using the SPSS 21 application. The results of the study can be concluded that the STEM approach has a significant effect on students' higher-order thinking skills in learning biology with the results of hypothesis testing 0.00 < 0.05. Furthermore, the STEM approach can increase students' HOTS in biology learning, the average score of the experimental class students is 85.76 and the control class is 70.31.

Abstract

This study aims to produce a valid and practical Mobile Learning (ML)-Based E-Module Assisted by the 3D Page Flip Professional Application. This type of research is development research using the Plomp model. The development stage of this model begins with the initial investigation stage, the development or prototyping stage, and the assessment stage. The instruments used in this development research were teacher interview guideline instrument sheets, student response questionnaire sheets, self-evaluation sheets, and E-Module validation sheets by experts, as well as e-module practicality assessment sheets filled in by teachers and students. The results showed that the 3D Page Flip Professional Application Assisted Mobile Learning (ML) Based E-Module based on expert judgment was very valid with a score of 89.10%. Practicality assessment of student assessment in field trials (large group evaluation) with a score of 88.95% with very practical criteria, and practicality assessment by the teacher obtained a score of 93.75% with very practical criteria. It can be concluded that the 3D Page Flip Professional Application Assisted Mobile Learning (ML) Based E-Module that has been developed is in the category of very valid, and very practical.

Abstract

This study aims to produce learning videos for Bacteria and Protists using Sparkol Videoscribe software for class X IPA students that are valid, practical, and effective. This research method is research and development (research and development) using the Plomp development model. This development stage consists of three stages, namely the preliminary research phase, the development or prototyping phase, and the assessment phase. The results of this study indicate that the developed learning videos have a validity value of 85.63% with a very valid category based on expert judgment. The practicality assessment by the teacher was 89.13% in the very practical category and the large group evaluation assessment by students obtained a practicality value of 87.69% in the very practical category. The results of the effectiveness test showed that the class that used learning videos had higher average competency values of knowledge, attitudes, and skills, compared to classes that did not use learning videos. It can be concluded that the learning videos that have been developed are in the category of very valid, very practical, and very effective for increasing students' learning competence

Abstract

Biofuels are fuels derived from biomass and consist of biogas, biodiesel and bioethanol. Bioethanol is a biofuel whose main ingredients are from plants and generally use microorganisms in the fermentation process. One way is to use thermophilic bacteria with the advantage of low contamination levels and better product quality. Bacteria that exist in nature are not only in a single form but also exist in the form of a consortium in which there are beneficial or detrimental bacterial interactions. Favorable bacterial interactions indicate that the bacteria are compatible. Using a consortium with compatible bacteria gives better results than using a single bacteria. This study aims to determine the compatibility and effect of a consortium of thermophilic bacteria from Mudiak Sapan hot springs on biofuel yields. This research is a descriptive research. To test the cooperation between isolates, a compatibility test was carried out using the disk diffusion method. Then the biculture consortium isolates of thermophilic bacteria were fermented in liquid TMM medium (Thermophilic Minimum Media) and the bioethanol content was measured using a distillation apparatus. The results of this study showed that the six pairs of compatible consortium and MS 9-12 consortium produced the highest bioethanol, namely 1.0003%. Then the biculture consortium isolates of thermophilic bacteria were fermented in liquid TMM medium (Thermophilic Minimum Media) and the bioethanol content was measured using a distillation apparatus. The results of this study showed that the six pairs of compatible consortium and MS 9-12 consortium produced the highest bioethanol, namely 1.0003%.
