Abstract

BSTRAKFitofarmaka di Indonesia sudah tidak asing lagi dan banyak dimanfaatkan untuk pengobatan-pengobatan tradisional. Tanaman sambung nyawa merupakan salah satu tanaman yang sudah banyak dimanfaatkan untuk pengobatan manusia karena memiliki kandungan senyawa metabolit sekunder yang berkhasiat obat. Kandungan-kandungan tersebut seperti flavonoid, tannin, dan saponin. Selain untuk pengobatan manusia, daun sambung nyawa juga berpotensi untuk digunakan sebagai obat ikan dalam rangka pencegahan penyakit. Vibriosis merupakan salah satu penyakit bakterial yang disebabkan oleh Vibrio alginolyticus dan rentan menyerang ikan kerapu macan. Penggunaan antibiotik sintetik telah banyak digunakan tetapi menimbulkan banyak dampak buruk, sehingga perlu alternatif baru untuk pencegahan vibriosis. Salah satunya yaitu dengan penggunaan ekstrak daun sambung nyawa, hal ini didukung karena potensinya. Penelitian ini bertujuan untuk mengkaji pengaruh ekstrak daun sambung nyawa untuk meningkatkan ketahanan tubuh ikan kerapu macan sehingga dapat mencegah serangan bakteri Vibrio alginolyticus. Metode yang dilakukan meliputi ekstraksi bahan, uji fitofarmaka, uji in vitro, uji in vivo, uji hematologi, dan uji histopatologi. Dosis ekstrak daun sambung nyawa yang paling efektif untuk meningkatkan ketahanan tubuh ikan kerapu macan dan mencegah serangan Vibrio alginolyticus adalah dosis 700 ppm.Kata kunci : daun sambung nyawa, pencegahan, vibriosi

Abstract

Vanamei prawns have fast growth and can reared in high density. it has an impact on water quality deterioration and disruption of survival rates and growth. Various ways to cope has been done, one of them is with probiotic bacteria. The new strain of D2.2 bacteria is thought to be effective of ammonia utilization. Probiotics with local bacteria Bacillus sp. D2.2 is cultured on molasses technical medium to be applied semi-mass. The purpose of this study is to asses the efectivity of Bacillus sp. D2.2 inthe molasses technical medium on water quality and growth performance of vaname prawns (Litopenaeus vannamei). The research was used complete randomized design (RAL) with four treatments, A (Control), B (Application of 5 ppm Bacillus sp. D2.2 cultured in molasses technical medium), C (Application of 10 ppm Bacillus sp. D2.2 cultured in molasses technical medium), D (Application of 15 ppm Bacillus sp. D2.2 cultured in molasses technical medium) were repeated three times each. The results showed no effect on water quality and shrimp survival rate, but absolute growth (W), daily growth rate (GR) and feed conversion ratio (FCR) showed that B and C treatment had better than control. Abstract

This study aims to analyze the effect of the various concentration of guava leaf extract (Psidium guajava Linnaeus) on the prevention of Motile Aeromonas Septicemia disease in common carp (Cyprinus carpio Linnaeus). In a total of 150 common carp, length 9-11 cm with a density of 10 carps/aquarium. This study used a completely randomized design through 5 treatments, each treatment has 3 replications, namely (K+) consists of without guava Psidium guajava leaf extracts and without bacteria-infection Aeromonas hydrophila, (K-) consists of without guava Psidium guajava leaf extracts with bacteria-infection Aeromonas hydrophila, (A) the application of 125 ppm guava Psidium guajava leaf extracts, (B) the application of 250 ppm guava leaf extracts, (C) the application of 500 ppm guava Psidium guajava leaf extracts for 23 days. The parameters observed total of leukocytes, differensial leukocytes, hematocrit levels, erythrocyte, hemoglobin, phagocytic activity (AF), phagocytic index (IF), relative percent survival (RPS), survival rate (SR) and water quality. Data were analyzed with Anova 95% confidence interval and continued with Duncan test. The parameter also showed a significant result (P<0,05) with the application of guava Psidium guajava leaf extracts to the total leukocytes, differensial leukocytes, erythrocyte, hemoglobin, phagocytic activity, phagocytic index and relative percent survival. The results of this research indicate the concentration of 250 ppm guava Psidium guajava leaf extracts is the optimum application to increase common carp non-spesific immune responses.

The aims of the study were to examine the exact percentage of prebiotics in sinbiotics, administered through feed, so as to enhance nonspecific immune responses in white shrimp. The sweet potato extract was used as prebiotic, combined with local isolated Bacillus sp. D2.2 as a probiotic applied simultaneously as a synbiotic. The feed was treated with 0% sinbiotic treatment (treatment A / control), 0% prebiotic and 6% probiotics (B treatment), 2% prebiotic and 6% probiotics (treatment C), 4% prebiotics and 6% probiotics (treatment D). Examination of nonspecific immune responses to shrimp included total hemocyte count (THC), phagocytosis activity (AP), phagocytosis index (IP), phenoloxidase activity (PO), and superoxide dismutase activity (SOD). Observation of the non-specific immune response of vaname shrimp after treatment showed that treatment C was the best prebiotic  to increase non-specific immune response in vaname shrimp.Penelitian ini bertujuan untuk mempelajari persentase prebiotik yang tepat dalam sinbiotik, yang diberikan melalui pakan, sehingga dapat meningkatkan respon imun nonspesifik pada udang vaname. Dalam penelitian ini digunakan ekstrak tepung ubi jalar sebagai prebiotik, dikombinasikan dengan isolat lokal Bacillus sp. D2.2 sebagai probiotik yang diaplikasikan secara bersamaan sebagai sinbiotik. Pakan yang diberikan adalah pakan dengan perlakuan 0% sinbiotik (perlakuan A/kontrol), 0% prebiotik dan 6% probiotik (perlakuan B), 2% prebiotik dan 6% probiotik (perlakuan C), 4% prebiotik dan 6% probiotik (perlakuan D). Pemeriksaan respon imun nonspesifik pada udang meliputi total hemocyte count (THC), aktivitas fagositosis (AF), indeks fagositosis (IF), aktivitas phenoloxidase (PO), dan aktivitas superoxide dismutase (SOD). Pengamatan pada respon imun non spesifik udang vaname setelah diberi perlakuan menunjukan bahwa perlakuan C merupakan persentase prebiotik terbaik untuk meningkatkan respon imun non spesifik pada udang vaname.
