Abstract

Isolasi dan karakterisasi alginat dari rumput laut Sargassum polycystum telah dilakukan dengan menggunakan metode jalur kalsium alginat. Proses ini dilakukan dengan cara pengendapan menggunakan CaCl2 dengan waktu 3, 5, dan 7 hari untuk mendapatkan alginat dengan kualitas yang baik. Beberapa parameter yang digunakan untuk pengujian kualitas alginat yaitu uji fisiko-kimia, uji proksimat, dan analisis FTIR, serta uji aktivitas antioksidan. Isolat Ca-alginat SAR-7 menunjukkan hasil bahwa variasi waktu pengendapan sangat berpengaruh terhadap rendemen, yaitu 3,52% dengan kemurnian 32,00%, viskositas 8,74 Cp, pH 7,00, kadar air 19,29%, kadar abu 30,00%, kadar protein 3,50%, kadar lemak 25,07%,dan kadar karbohidrat 22,14%. Analisis FTIR menunjukkan hasil bahwa kalsium alginat mempunyai gugus fungsi –OH, C=O, C-O, dan alkil. Hasil uji aktivitas antioksidan menunjukkan bahwa Ca-alginat dengan konsentrasi 50 ppm memiliki bioaktivitas antioksidan dengan kemampuan inhibisi DPPH sebesar 19,57%. Kata kunci: Sargassum polycystum, ektraksi, alginat, antioksidan

Abstract

Spons Haliclona sp. diketahui memiliki kandungan metabolit sekunder dengan aktivitas sitotoksik yang tinggi sehingga berpotensi untuk dimanfaatkan sebagai obat antikanker. Dalam penelitian ini, telah dilakukan uji toksisitas dan uji fitokimia terhadap ekstrak metanol spons Haliclona sp. asal Pulau Lemukutan Kabupaten Bengkayang Kalimantan Barat. Metode yang digunakan pada penelitian ini meliputi maserasi dengan pelarut metanol, uji fitokimia dan uji toksisitas. Uji toksisitas terhadap ekstrak metanol dilakukan dengan menggunakan metode Brine Shrimp Lethality Test (BSLT). Potensi ekstrak metanol spons sebagai antikanker dapat diketahui melalui tingkat toksisitasnya terhadap Artemia salina L. yang merupakan skrining pendahuluan menuju tahapan pengujian selanjutnya terhadap sel kanker yaitu sitotoksisitas. Ekstrak metanol dari spons Haliclona sp. bersifat toksik dengan nilai LC50 sebesar 70,1 ppm sehingga berpotensi dikembangkan dan diteliti lebih lanjut komponen aktifnya sebagai bahan baku obat antikanker. Kata Kunci: antikanker, Haliclona sp., toksisitas

Abstract

Marine organisms-associated bacteria have a potential source of antimicrobial compounds. The aim of the research was to find out characteristics of antimicrobial activity of E. cottonii Doty-associated bacteria extracts and their genus from Lemukutan Island, Kalimantan Barat. Five of 11 isolates showed antimicrobial activity namely Flavobacterium sp. K08, Alteromonas sp. K09, Bacillus sp. K10, Klebsiella sp. K11, and Pseudomonas sp. K12. Furthermore, their secondary metabolites were extracted with methanol. The characteristic of the best bacteria extracts minimum inhibitory concentrations (MICs) for Escherichia coli, Bacillus spaericus, Staphylococcus epidermidis, and Candida tropicalis were obtained from bacteria extract of Bacillus sp. K10 (250 μg/disc), Bacillus sp. K10 (750 μg/disc), Pseudomonas sp. K12, (500 μg/disc), and Alteromonas sp. Abstract

Nutmeg (Myristica fragrans Houtt) is a medicinal plant that produces essential oils and is often used as a flavoring of food and medicine. The process of extracting essential oils from the nutmeg fruit pulp using the steam-water distillation method. The yield of essential oil obtained at 0.07765%, with a pale yellow color, the distinctive smell of nutmeg oil with a refractive index of 1.485, and a specific gravity of 0.909 g / mL and soluble in ethanol. The results of GC-MS identification showed that the essential oil of nutmeg fruit flesh contains five components, the main compound is myristicin (18.44%), α-pinene (14.2%), limonene (9.37%), γ-terpinene (7.98%) and terpinolene (7.66%). Based on the test results of anti-inflammatory activity, the value of the percentage of red blood cell stability optimum mounting to 80.21% at a concentration of 10 ug / mL, where the standard aspirin has an activity of 72.26%. These results indicate that the meat nutmeg essential oil is potential as anti-inflammatory agents.

Abstract

Research on the adsorption of Mn (II) ions has been carried out using  biomass duckweed (Lemna minor linn) with the esterified by citric acid. This study aims to determine the maximum adsorption capacity of Mn (II) ions in using adsorbent biomass of duckweed esterified by citric acid. Water nail biomass adsorbents were characterized using FTIR to determine the functional groups of citric acid esterified adsorbents. The stages of this research include modification of water biomass using citric acid, followed by manganese (II) ion adsorption tests with pH variations in pH, contact time of adsorption and initial concentration variations under optimum conditions. The results showed that optimum adsorption occurred at pH  5, contact time for 10 minutes and the concentration of Mn (II) solution 15 mg / L was measured using atomic absorption spectrophotometry. The equation used in the study follows the Langmuir and Freundlich equations. Based on the Langmuir isotherm equation, the maximum adsorption capacity of Mn (II) ion using citric acid modified water nail biomass is  7,645 mg/ g, the equilibrium constant is 4,317 L/ mol and the adsorption energy is 36,84039 KJ / mol.

Abstract

Uji pendahuluan bioaktivitas dan identifikasi gugus fungsi metabolit sekunder ekstrak dalam metanol, n-heksan and metilen klorida dari spons Agelas nakamurai Hoshino, asal pulau Kapoposang, kepulauan Spermonde telah dilakukan. Juga test bioaktivitas dilakukan menggunakan Artemia salina (LC50) untuk mendapatkan  konsentrasi letalnya.  Hasilnya  LC50  adalah 187.932  ppm   yang  menunjukkan potensi ekstrak sebagai obat anti kanker. Identifikasi komponen dengan reagensia spesifik seperti Lieberman- Burchard, Mayer, Wagner, Dragendorff dan Salkowski ditambah hasil analisis FTIR menunjukkan kandungan komponen ekstrak seperti alkaloid, steroid and terpenoid. Karakteristik pita serapan alkaloid adalah for alkaloid adalah stretching NH at 3600 to 3400 cm-1 dan stretching pendukung C-N at 1250 to 1930 cm-1. Komponen spesifik untuk Agelas nakamurai adalah substitusi bromo dalam alkaloid yang ditandai oleh munculnya pita serapan  C-Br medium pada 760 to 745 cm-1.

Abstract

Isolation, identification and bioactivity test of nonpolar compunds has been conducted on n-hexane extract sponge Haliclona (Reniera) fascigera of the island Samalona, Makassar, South Sulawesi. Isolation techniques consist of maceration, partition, fractionation using 2 steps using vacuum column chromatography and flash column chromatography and also purification step. Identification of isolate was analyzed with TLC, FTIR, GC-MS, and 1H NMR. The research result was obtained isolate as much as 4 mg and clear solid form. FTIR analysis indicate functional group OH, CH­2, CH3, C=O, =CH and C-O. Identification of spot TLC under UV light and spotting solvent show that the spot was active on UV long and short waves and dark brown with cerium sulphate. The results show that isolate A contain aliphatic and alcohol compounds. Bioactivity test results indicate that these isolate A be able to inhibit the growth of bacteria Staphylococus aureus with inhibition zone diameter were 11.60 mm (100 ppm); 10.40 mm (50 ppm); 9.50 mm (25 ppm); 6.50 mm (10 ppm); 6,60 mm (negative control); 17.40 mm (positive control) and against bacteria Escherichia coli with inhibition zone diameter were 6.70 mm (100 ppm); 6,10 mm (50 ppm); 5,90 mm (25 ppm); 5.85 mm (10 ppm); 5,20 mm (negative control); 18.60 mm (positive control).

Abstract

Toxicity assessment have been conducted on nine sponges species collected from Spermonde Archipelago namely Kapoposang Island, Samalona Island and Lanjukang Island. Toxicity assessment was using Brine Shrimp lethality Test (BST) method againts sponge extracts. Theresults test were expressed as 50% lethal concentration (LC50) to Artemia salina and correlated with prospecting extract potential as anticancer. All extracts obtained from smallscale extraction by methanol. Test results as follows: Acanthostrongylophora ingens (24.830 μg/mL), Hytios erectus (46.140 μg/mL), Stylissa massa (>1000 μg/mL), Theonella sp (35.010 μg/mL), Niphates olemda (87.500 ppm), Haliclona (Reniera) fascigera (78.160 μg/mL), Petrosia (Petrosia) sp (3.008), Callyspongia (Euplacella) biru (0.515 μg/mL). The test results showed that seven species are potential as anticancer with LC50 value ≤ 500 ppm and only Stylissa massa extracts was not potential as anticancer.

Abstract

The isolation of lipophilic component of CC_7 on n-hexane fraction of Agelas nakamurai sponge has been conducted. The isolates are liquid oil colorless in 5.1 mg from 53,7 g sample. The isolates were analyzed by GC-MS and FTIR. The results of the analysis isolate CC_7 by FTIR showed the typical vibration of stretching at 2956,87 (asym. –CH3), 2924,09 (asym. –CH2), 2854,65 (sym. -CH3), 1730,15 (C=O), 1662,64 (C=C), 1595,13 (C-C(O)), 1269,16 (C-O), 1186,22 (C-C(O)-O), 1111,00 (O-C-C), 1020,34 (C-O-C-C), 962,48 (=C-H), 678,94 and 657,73 (cis –HC=CH–), bending rocking (-CH2-) at 731,02, bending (-CH2) dan (-CH3) at 1460,11 dan 1373,32. The results of GC-MS analysis showed the lipophilic component such as methyl palmitate (tR 19,77; m/z = 270), ethyl palmitate (tR 21,07; m/z = 284), palmitic acid (tR 21,18; m/z = 256, ethyl oleate (tR 24,09; m/z 310), tributyl asonitate (tR 24,38; m/z = 342), tributyl citrate (tR 24,95), tributyl acetyl (tR 26,41; m/z = 402), dioctyl adipate (tR 28,11; m/z =m 370, and squalene (tR 30,30; m/z 410).

Abstract

The increase of issues on the antibiotics resistant pathogenic bacteria has triggered high exploration for new antimicrobial compounds. One of the potential sources is sponge-associated bacteria. The aim of this study was to get sponge-associated bacteria extract containing antimicrobial activities. On the basis screening of antimicrobial activity using by streaking on agar medium, there were two potential isolates with antimicrobial activities namely LCS1 and LCS2. The two isolates were cultivated,then secondary metabolite product were extracted using methanol as a solvent. Minimum inhibitory concentrations (MICs) of extract LCS 1 were 1,000 μg/well for S. aureus, 950 μg/well for Salmonella sp.and 800 μg/well for Bacillus subtilis. Minimum inhibitory concentrations of extract LCS 2 were 500 μg/well for S. aureus, 1,050 μg/well for Salmonella sp., 750 μg/well for Bacillus subtilis, 350 μg/well for P. aeruginosa, 750 μg/sumur terhadap B. subtilis. Based on the MIC values, the two assay extracts have a relatively low antimicrobial activity. Keywords:Antimicrobial,Sponges associated bacteria,MICs
