Abstract

The study about effect of media composting duration from industrial waste of tea production to the mycelium growth and production of black ear mushroom (Auricularia polytrica (Mont.) Sacc.) was conducted from December 2012 to February 2013 in the Laboratory of Microbiology, Department of Biology, Andalas University, Padang. This study aimed to determine the effects of media composting duration to the mycelium growth and production of black ear mushrooms. The research was designed by using completely randomized design (CRD) in five treatments of Composting duration, they were 0, 1, 3, 5 and 7 days in 5 replications. The result showed that the treatment of 5 days media composting was the optimum length of time for 32 days the growth of mycellium, the weight of fruiting bodies was 50.89 g, the widest diameter of the cap of fruiting body was 9.05 cm, the cellulase activity of the mycellium in the baglogs was 4,769 units/g and producing the highest content of glucose after composting was 153.526 µg/g. Keywords: composting duration, waste of tea, Auricularia polytrica, mycelium growth.

Abstract

Antibacterial activity of crude extracts of pisang kepok kuning plants (Musa paradisiaca Linn.) were tested against Staphylococcus aureus and Escherichia coli. Roots, corms, pseudoterms, flowers, and ripe fruits of pisang kepok kuning cultivar were macerated with ethanol 96% and evaporated in a rotary evaporator at 50C. All of crude extracts were screened for its antibacterial activity by using Agar Well Diffusion Method with concentration based on 200 grams of fresh weight. This research used Completed Randomized Design (CRD) with nested pattern, four replications, and consist of two factors. First factor was bacteria and the second factor was crude extracts of pisang kepok kuning such as roots, corms, pseudoterms, flowers, and ripe fruits. The results showed that all of crude extracts have significant antibacterial activity against S. aureus and E. coli. Crude extract of the corm has highest antibacterial activity against S. aureus with diameter of inhibition area was 20,391 mm with inhibition zone type isirradical and diameter inhibition area 18,962 mm for E. coli that has radical zone. Keywords : Musa paradisiaca Linn., S. aureus, E. coli, antibacterial

Abstract

A study about potential and characterization of native bacteria of intestinebroiler (Gallus gallus domesticus L.) as probiotic candidate of a broiler poultry product were conducted from December 2012 to March 2013 at Microbiology Laboratory, Department of Biology, Andalas University, Padang. This study aimed to obtainbacterial isolates and to describe morphological characters of each isolate of native bacteria as probiotic candidate for thebroiler. This study used an experimental method and analyzed descriptively. The results showed that rough material of intestine consisted of fermentative bacteria (57x107cfu/g), amylolitic (118x107cfu/g), cellulolytic (63x107cfu/g) and proteolytic bacteria (52x107cfu/g). Two isolates were potensial probioticcandidates; amylolytic bacteria and cellulolytic bacteria. The characteristics both probiotic candidates were gram negative, non-sporulating, positive catalase and motile. Keywords: Gallus gallus domesticus, intestine bacteria, characterization
