Abstract

Paradoxical sleep deprivation (PSD) and total sleep deprivation (TSD) cause disrupt of male infertilitybut sleep recovery (SR) can improve male reproduction function that connected with occupational works health. The aim of this study is to determine the difference of reproduction parameters in male albinorats  after exposed by various sleep deprivation models. This research was experimental post-test only with control group design. Rats were divided into 5 groups (6 animals each group) : negativecontrol, PSD (II), TSD (III), PSD with SR, TSD with SR.  The study results  showed that average of spermatogenic  group IV (8.35 ± 0.06) and V (8.27 ± 0.27) had higher scores. group IV had the highest number of Leydig cell (5,91 ± 1,43),  group I had the highest rates (40,02 ± 2,04)   number of Sertoli cell and there were no significant difference  average of epithelial diameter (p=0,598) and height (p=0,895). There were significant difference of spermatogenic score post-SR,  number of Sertoli and Leydig cells, but no significant difference of epithelial diameter and height of seminiferous tubule after exposed by various sleep deprivation stress model. Sleep recovery in occupational work can  repair the histological parameter of reproduction. Key word : male albino rats, reproduction parameters,  sleep deprivation models

Abstract

Paradoxical Sleep Deprivation (PSD) elevates glucocorticoid and Reactive Oxygen Species (ROS) levels that cause oxidative stress, trigger spermatogenic cells damage, and reduce the number of sertoli cells. Vitamin C and E are antioxidants that could prevent spermatogenesis damage by preventing free radical formation. The study aimed to determine the effect of single and combined doses of vitamin C and E in improving spermatogenesis of white male rats (Rattus norvegicus) model of PSD. This is experimental research with post-test only and control group design on 28 white male rats distributed into four groups, i.e. group I (control, PSD), group II (PSD+vitamin C), group III (PSD+vitamin E), and group IV (PSD+a combination of vitamin C and E). Testicular preparations were stained using Hematoxylin-Eosin staining, quantitative scores of spermatogenic cells were measured using the Johnsen method, and the number of sertoli cells was counted in 10 seminiferous tubules in each of the three testicular sections. Group I has the lowest mean of Johnsen score (5.27±0.28), and group IV has the highest mean score (8.95±0.62), while the Mann-Whitney test showed a significant difference (p<0.05) between group II, III and IV compared to group I (control group). Mean sertoli cells number of group I is the lowest (10.66±1.04), and group IV has the highest sertoli cells number. The post-hoc LSD test showed a significant difference (p<0.05) between group IV and the other groups. Thus, the combination of vitamins C and E improves Johnsen score and sertoli cells number of male rats (Rattus norvegicus) experiencing PSD.

Abstract

Paradoxical sleep deprivation (PSD) or sleep disorders can affect sexual function and potentially reduce fertility rates. The active substance of Purwoceng (Pimpinella pruatjan Molk.), namely flavonoid and tannin compounds can improve the quality of function. Drug nanoemulsion is expected to increase the therapeutic effect and reduce toxicity. This study aims to determine the effect of purwoceng nanoemulsion on serum testosterone levels, testicular volume and testicular histopathology of male Wistar rats after stress induction of paradoxical sleep deprivation for 96 hours. This research is an experimental study with a post-test only design with a control group. A total of 24 rats were divided into 6 treatment groups. Group A was given PSD, group B was given PSD and sleep recovery, group C was given PSD and purwoceng extract 25 mg/300 gBW/day, group D was given PSD and purwoceng nanoemulsion 25 mg/300 gBW/day, group E was given PSD and purwoceng nanoemulsion. 50 mg/300 gBW/day, and group F was given PSD and purwoceng nanoemulsion 75 mg/300 gBW/day. Serum testosterone levels were measured by the ELISA method, testicular volume and histopathology determined using Image J software and data were analyzed by One Way ANOVA test and followed by Post Hoc Bonferroni test. Statistical analysis showed that there were significant differences in serum testosterone levels, testicular volume and testicular histopathology (p<0.005). Administration of purwoceng nanoemulsion affects serum testosterone levels, testicular volume, and testicular histopathology after PSD induction
