Abstract

Telah dilaporkan bahwa probiotik dapat memelihara keseimbangan mikroflora usus dan meningkatkan sistem imun usus. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian probiotik indigenous Lactobacillus fermentum dan Lactobacillus plantarum pada profil kesehatan usus halus tikus yang diinfeksi Enteropathogenic E. coli (EPEC). Penelitian ini dilaksanakan pada bulan Agustus–November 2011 di Institut Pertanian Bogor. Sebanyak 90 ekor tikus jantan galur Sprague Dawley telah digunakan dan dibagi menjadi enam kelompok perlakuan, yaitu kelompok kontrol negatif (A), perlakuan Lactobacillus plantarum (B), perlakuan Lactobacillus fermentum (C), perlakuan Lactobacillus plantarum dan EPEC (D), perlakuan Lactobacillus fermentum dan EPEC (E), dan perlakuan EPEC (F). Perlakuan dilaksanakan selama 21 hari. Setelah tikus dikorbankan, usus halus diproses menggunakan metode embedding standar dengan parafin dan diwarnai memakai hematoksilin eosin. Data hasil penelitian dianalisis secara statistik dengan rancangan acak lengkap analysis of variance (ANOVA). Hasil penelitian menunjukkan bahwa perlakuan L. fermentum dan L. plantarum dapat menurunkan persentase kerusakan vili usus halus 14–62% dan meningkatkan tebal mukosa usus halus 14–29%. Simpulan, perlakuan probiotik indigenous L. fermentum dan L. plantarum dapat meningkatkan kesehatan usus halus. L. fermentum lebih baik dalam menghambat kerusakan usus halus tikus yang diinfeksi EPEC. The objective of this study was to analyze the influence of indigenous probiotic Lactobacillus fermentum and Lactobacillus plantarum on the morphological profile of small intestine of Enteropathogenic E. coli (EPEC)- infected rats. This study was done from August to November 2011 at Bogor Agricultural University. A total of 90 male rats Sprague Dawley were used in this study. They were devided into 6 groups; negative control (A), Lactobacillus plantarum treated (B), Lactobacillus fermentum treated (C), Lactobacillus plantarum and EPEC treated (D), Lactobacillus fermentum and EPEC treated (E), and EPEC infected group (F). The treatments were done for 21 days. Small intestine tissues were processed using paraffin embedding standard method and stained with haematoxylin eosin. The data were analyzed using analysis of variance (ANOVA). The results showed that L. fermentum and L. plantarum treatments decreased percentage of small intenstine villi damage (14 to 62%) and increased the thickness of small intestine mucosa (14 to 29%). In conclusions, treatment of indigenous probiotic L. fermentum and L. plantarum increase the health of small intestine. L. fermentum give better effect in inhibiting the damage of small intestine of rats, which under EPEC infection. Abstract

Aterosklerosis pada arteri iliak berkaitan dengan kejadian peripheral arterial disease (PAD) pada manusia. Penelitian ini bertujuan untuk menganalisis lesio aterosklerosis pada arteri iliak sebagai salah satu faktor penyebab PAD. Penelitian ini menggunakan sembilan monyet ekor panjang jantan dewasa dari 3 kelompok respons yang diberi pakan aterogenik IPB-1 selama 2 tahun. Arteri common iliak diambil pada saat nekropsi, diproses menjadi slaid histologi dan diwarnai dengan Verhoeff van Giesson (VVG). Keparahan lesio dikategorikan sebagai grade I sampai VI dan dianalisis secara deskriptif. Area lesio ditentukan melalui panjang lamina elastik interna (PLEI), area intima (AI), serta ketebalan maksimum intima (KIM) dan dianalisis dengan ANOVA. Hasil penelitian menunjukkan 55,56% monyet membentuk lesio plak aterosklerosis (grade IV sampai VI) yang mana mereka berasal dari kelompok intermediet dan hiper-responsif. Kelompok hiper-responsif menunjukkan AI dan KMI paling tinggi (0,979 mm2; 0,352 mm) diikuti kelompok intermediet (0,372 mm2; 0,237 mm) dan kelompok hipo-responsif (0,049 mm2; 0,052 mm) (P<0,05). Hasil ini menunjukkan semakin tinggi kolesterol plasma, semakin luas area lesio yang terbentuk. Keseluruhan hasil mendukung bahwa monyet yang diberi pakan aterogenik IPB-1 selama 2 tahun mampu membentuk lesio plak aterosklerosis pada arteri common iliak, dan berpotensi sebagai hewan model PAD, kecuali kelompok hipo-responsif.

Abstract

This study aimed to explore the histomorphology of the pancreatic tissue of medaka fish to develop medaka fish as a diabetic animal model. Adult medaka fish were obtained with a body length of 2.53±0.22 cm and a weight of 265.8±25 g. Medaka fish were fixed using Boiun's fixative solution. Macro anatomical observations of the pancreas were conducted using a stereomicroscope. Histological preparations were made according to the Hematoxylin-eosin staining standard procedure. Pancreatic histomorphology of medaka fish was carried out using a light microscope with 100x and 400x magnification. The macro anatomy observations of the pancreas showed the interconnection with several tissues, such as spleen, gallbladder, liver, fat, and intestines. The pancreas was located toward the liver, oversees the intestines, and covered with fat. Pancreatic histomorphological observations showed exocrine cells composed of serous pyramidal acinar cells and endocrine cells (islets of Langerhans). Intralobular and intercalated ducts (ducts of Boll) were composed of simple cuboidal epithelium. This study concluded that the pancreas organ in medaka fish is a single organ, and histomorphologically has both of exocrine and endocrine cells.

Abstract

Penelitian  ini  bertujuan  menganalisis  pengaruh  pemberian  probiotik  lokal  Lactobacillus  fermentum  (L.  fermentum)  dan  Lactobacillus plantarum  (L.  plantarum)  terhadap profil kandungan IgA usus halus tikus yang diinfeksi   enteropatogenik  E. coli  (EPEC)  menggunakan teknik imunohistokimia.  Sebanyak  90  ekor  tikus  jantan  galur  Sprague  Dawley  digunakan  dan  dibagi  menjadi  enam  kelompok  perlakuan,  yaknikelompok kontrol negatif (A), kelompok perlakuan  L. plantarum  (B), kelompok perlakuan  L. fermentum  (C), kelompok perlakuan  L. plantarumdan EPEC (D), kelompok perlakuan  L.  fermentum  dan EPEC (E),  dan  kelompok perlakuan EPEC (F). Perlakuan dilaksanakan selama 21 hari. Deteksi IgA dilakukan dengan teknik imunohistokimia pada jaringan usus halus. Hasil penelitian menunjukkan bahwa perlakuan probiotik lokalL. fermentum selama 2-3 minggu, dan perlakuan L. plantarum selama 2 minggu mampu meningkatkan kandungan IgA di usus halus tikus. Pada tikus yang dipapar EPEC, L. fermentum lebih baik dalam meningkatkan kandungan IgA dibandingkan L. plantarum.

Abstract

Orangutan is a great ape that has anatomical and physiological aspects similar to those of gorillas and chimpanzees, although orangutan is more arboreal than other great apes. This research aims to study the morphofunctional characteristics of the lower hindlimb and foot muscles of Bornean orangutan and its relation to the type of locomotion and daily behavior. This research was conducted on the right leg of a male orangutan by observing the morphology of the lower hindlimb and foot muscles and observing the behavior of the Bornean orangutan through video observations. The results showed that digital flexor muscles are more developed than plantar flexor muscles as an adaptation towards arboreal activities that require finger flexor movements such as gripping tree branches. Orangutans have rudimentary hallux, which is related by the less developed intrinsic muscle and indicated by the way orangutan grips tree branches dominantly using digits II-V. At a certain time, male Bornean orangutans are active on terrestrial that causes the digital extensor muscles to have more developed as an adaptation to terrestrial activity. It can be concluded the morphology of the lower hindlimb and foot muscles is in accordance to its function and behavior adaptation in arboreal and terrestrial.
