Abstract

Telah dilakukan penelitian mengenai pengaruh jus daun Jati Belanda terhadap berat dan histologi ginjal mencit. Penelitian eksperimen ini menggunakan Rancangan Acak Lengkap (RAL) dengan empat kali pengulangan dan enam dosis perlakuan (0,00 g/BB/hari; 0,05 g/BB/hari; 0,10 g/BB/hari; 0,15 g/BB/hari; 0,20 g/BB/hari; 0,25 g/BB/hari) digunakan dalam penelitian ini. Mencit jantan usia empat bulan diberi perlakuan jus daun Jati Belanda dengan cara gavage selama 14 hari. Pada hari ke-15 mencit dimatikan dengan cara dislokasi leher kemudian dibedah dan ginjalnya diambil untuk kemudian ditimbang dan dibuat preparat histologinya. Pembuatan preparat dilakukan dengan metode parafin dengan pewarnaan Hematoxylin Erlich-Eosin untuk melihat struktur jaringan ginjal. Analisis data dilakukan baik secara kualitatif maupun kuantitatif. Hasil penelitian menunjukkan pemberian jus daun Jati Belanda mulai dosis 0,10 g/BB/hari hingga 0,25 g/BB/hari secara signifikan meningkatkan berat ginjal. Pengaruh jus jati belanda terhadap berat ginjal yaitu sebesar 86%. Rata-rata berat ginjal mencit semakin meningkat seiring dengan semakin tingginya dosis jus daun Jati Belanda yang diberikan. Adapun rata-rata diameter tubulus, diameter lumen, serta ketebalan sel epitel semakin meningkat seiring dengan semakin berat ukuran ginjal. Dengan demikian dapat disimpulkan bahwa  penggunaan jus daun Jati Belanda dalam rentang dosis antara 0,10 g/BB/hari hingga 0,25 g/BB/hari aman dikonsumsi bagi tubuh dalam jangka waktu singkat. Kata kunci : Guazuma ulmifolia Lamk., tubulus, lumen, Mus musculus L.

Abstract

The aim of this research was to reveal a relationship between students' concepts mastery on the matter of environmental pollution with the ability to assess the credibility of the source through problem-based learning. The type of research is pre-experiment which analyzed by correlation method. Then the study design using the post-test only design with a total sample of 39 students of grade 10 and for sampling technique uses simple-random sampling technique. Student’s concepts mastery on material environmental pollution are identified using the instrument in the form of multiple choice questions and essay question, while the level of ability to assess the credibility of the source is identified based on the accuracy of the students sort the level of credibility of source and the students' answers on the criteria that they use to sort the credibility of the source, which is scored with a rubric that has been made. The average score of students' mastery of concepts is 74.1 and quite good. In addition, through one sample t-test was also proven when problem-based learning have a real or significant influence on students' mastery of concepts in the matter of environmental pollution. Correlation test is then performed using the non parametric statistical using Spearman correlation, and got score 0.82 for correlation coefficient, which means having a high correlation with a contribution of 67.5%. Thus the conclusions that can be drawn is that there is a high correlation between students' concepts mastery on the matter of environmental pollution with the ability to assess the credibility of the source through problem-based learning.

Abstract

Penuaan sistem reproduksi (ovarian aging) telah diketahui pada beberapa spesies vertebrata dan manusia yang ditandai dengan kadar estrogen yang rendah. Hal ini berhubungan dengan peningkatan jumlah lipid (obesitas) dan sejumlah penyakit metabolik seperti hiperlipidemia, penyakit kardiovaskular, dll. Buah pare Momordica charantia L.) memiliki banyak manfaat yang menguntungkan bagi kesehatan manusia, seperti efek hipoglikemik, antioksidan, hipokolesterolemik, dan hipotrigliseridemik. Tujuan penelitian untuk mengetahui kadar lipid darah mencit pada usia middle-aged setelah diberikan jus buah pare. Dilakukan uji eksperimental pada Maret–April 2010 di Laboratorium Fisiologi Universitas Pendidikan Indonesia dan Kedokteran Hewan IPB terhadap 25 mencit galur Swiss Webster betina usia 10 bulan (middle-aged) yang terbagi dalam lima kelompok perlakuan berbeda (kontrol positif, kontrol negatif, 0,5 mL/40 g BB, 1,0 mL/40 g BB, dan 1,5 mL/40 g BB) yang diberikan jus buah pare selama 10 hari. Penelitian menggunakan rancangan acak lengkap. Hasil penelitian menunjukkan bahwa jus buah pare dapat menurunkan bobot badan mencit secara bermakna (p<0,05) pada semua dosis. Jus buah pare tidak dapat menurunkan kadar kolesterol total, tetapi meningkatkan kadar HDL (p<0,05) pada semua dosis perlakuan. Pada parameter lain, jus buah pare dapat menurunkan kadar trigliserida pada dosis 1,5 mL/40 g BB (p<0,05) dan menurunkan kadar LDL pada dosis 0,5 mL/40 g BB (136,49 mg/dL). Simpulan, jus buah pare dapat menstabilkan kadar trigliserida dan LDL kolesterol pada dosis 0,5 mL/40 g BB serta meningkatkan HDL kolesterol, tetapi tidak berpengaruh pada penurunan kadar kolesterol total. This is related to enhancement of lipid (obesity) and some metabolic diseases such as hyperlipidemia, cardiovascular disease, etc. Pare juice (Momordica charantia L.), contains of abundant advantageous bioactivities, such as hypoglycemic effect, antioxidant, hypocholesterolemic, and hypotrigliseridemic. This research purposed was to determine blood lipid levels of mice at the age of middle-aged after being given pare juice. The experimental animal used 25 females mice of 10 months old (middle-aged) on March–April 2010 at Physiology Laboratory Universitas Pendidikan Indonesia and Veteriner Medicine IPB, divided into five different treatment groups (controle positive, controle negative, 0.5 mL/40 g bw, 1.0 mL/40 g bw, and 1.5 mL/40 g bw) which were fed with pare juice for 10 days. Completely randomized design was used in this research. Results showed that pare juice reduced lower body weight of mice (p<0.05) at all doses. Pare juice did not reduce total cholesterol levels, but increased HDL (p<0.05) at all doses. Other parameters to pare juice reduced triglyceride levels at doses of 1.5 mL/40 g BW (p<0.05) and LDL levels at dose 0.5 mL/40 g BW (136.49 mg/dL). In conclusions, pare juice can stabilize triglyseride and LDL cholesterol level at dose 0.5 mL/40 g BW and increases HDL cholesterol, but has no influence to total cholesterol. Abstract

Permintaan terhadap produk makanan kesehatan seperti makanan bebas gula, rendah kalori, dan kaya serat semakin meningkat. Telah dibuat biskuit berbahan tepung kulit pisang yang memiliki kandungan serat pangan yang cukup tinggi. Tujuan penelitian yaitu mengidentifikasi jumlah mikroorganisme pada biskuit tepung kulit pisang. Metode pengujian mikrobiologi sampel makanan dilakukan yaitu uji TPC (Total Plate Count) untuk menghitung bakteri, Most Probability Number (MPN) untuk menghitung jumlah Escherchia coli, Salmonella sp., dan hitungan cawan untuk menghitung jumlah Staphylococcus sp. serta kapang/khamir. Analisis data mengacu pada persyaratan makanan yang sudah ditetapkan sesuai Standar Nasional Indonesia (SNI 01-2897-1992). Hasil penelitian menunjukkan bahwa pengujian mikroba biskuit tepung kulit pisang normal tidak melebihi ketentuan Standar Nasional Indonesia (SNI) yang mempersyaratkan maksimal sebesar 1,0x104 koloni/g bakteri. Uji Salmonella sp. diperoleh hasil negatif. Hasil uji kandungan E. coli kurang dari 3/g. Hasil uji kandungan Staphylococcus aureus sebanyak <1,0x101 koloni/g. Hasil uji jumlah kapang/khamir kurang dari 1,0x101 koloni/g. Kesimpulan hasil identifikasi mikrobiologi biskuit tepung kulit pisang dinyatakan dalam kategori normal sesuai Standar Nasional Indonesia (SNI, 2009) dan BPOM (2009), sehingga dapat dikonsumsi oleh masyarakat sebagai makanan kesehatan. Demand for health food products such as sugar-free foods, low in calories and fiber-rich foods is on the rise. It is known that made biscuits made from banana peel flour is known to have high dietary fiber content. The aim of this research was to identify the number of microorganisms on the biscuits with addition banana peel flour. Microbiological test method of food samples was done by TPC (Total Plate Count) test to calculate bacteria, Most Probability Number (MPN) to calculate the amount of Escherchia coli, Salmonella sp., and plate count to calculate the amount of Staphylococcus sp. and mold/yeasts. The results of data analysis refers to food requirements that have been established according to Indonesian National Standard (SNI 01-2897-1992). The results showed that the testing of microbial biscuits of banana peel flour did not exceed the provisions of Indonesian National Standard (SNI) which required a maximum of 1.0x104 colony/g bacteria. Salmonella sp. obtained negative results. The results of Escherchia coli content test less than 3/g. The results of the Staphylococcus aureus content test were <1.0x101 colony/g. The test result of the amount of mold/yeast is less than 1.0x101 colony/g. The conclusion of microbiology identification result stated that biscuit of banana peel flour was normal category according to Indonesian National Standard (SNI, 2009) and BPOM (2009), so it can be consumed by society as health food.
