Abstract

Caffeine analysis from Luwak coffee in different types of coffees, Luwak species and processed by using TLC Scanner method had been done. 8 samples of Luwak coffee, that is R.Ab.K ; R.Ab.B ; R.Ph.K ; R.Ph.B ; A.Ab.K ; A.Ab.B ; A.Ph.K ; A.Ph.B which were extracted by dichloromethane (DCM). TLC separation had been done through stationary phase with silica gel GF254 and mobile phase are solute of dichloromethane (DCM) : methanol (9,5 : 0,5). Caffeine content had been counted by using regression equality y = 2715,112 + 4,389x and giving percentage in succession 1,91 ; 2,04 ; 1,30 ; 1,89 ; 1,29 ; 1,45 ; 1,25 ; 1,42 %. The highest of caffeine had been given by Robusta coffee which has been given before to A. binturong luwak and processed wetly 2,04 %. The lowest of caffeine content had been given to Arabica coffee which has been given before to P. hermophroditus luwak 1,25 %. Three way – Anova statistic SPSS 21 tabulation had showed the result that the kind of coffee, variety of Luwak and the way of processing gave significant value toward caffeine content in p < 0,05.

Abstract

The influence of different species of civet on caffeine content of Robusta civet coffee ( Coffea Robusta) had been done. The Civet species used are Paradoxurus Hermophroditus (Pandan civet) and Arctictis Binturong ( Bulan civet). The Coffee used is Robusta varieties ( Coffea Robusta). Caffeine’s level of robusta civet coffee has been given to two species of civet and after that they are analyzed using TLC Scanner Method quantitatively. The process of robusta civet coffee extraction was done by using Dichloromethane solvent and sonicated for 15 minutes on  themperature 40 º C, then the filtrate obtained was filtered with whatmant filter paper, then concentrated by using rotary evaporator. The quantitative analysis measured from the caffeine was measured using Thin Layer Chromatograpy ( TLC ) then Scanned by Densitometry ( TLC Scanner ) to obtain the content of caffeine from the civet coffee. Robusta coffee beans given to the civet Bulan (Arctictis Binturong) species have 1,91 % caffeine content where the caffeine content is higher when compared to robusta  coffee given to Pandan civet (Paradoxurus Hermophroditus) which has only 1,30 % caffeine. Based on the SPSS 21 anne-ment using anava Three Roads, it is known that differences in civet species affect the levels of robusta civet coffee caffeine.Telah dilakukan Penelitian Pengaruh Perbedaan Spesies Luwak Terhadap Kadar Kofein dari Kopi Luwak Jenis Robusta (Coffea Robusta). Spesies Luwak yang digunakan adalah Paradoxurus Hermophroditus ( Luwak Pandan) dan Arctictis Binturong ( Luwak Bulan). Kopi yang dipakai adalah spesies Robusta ( Coffea Robusta). Kadar kofein kopi luwak robusta yang diberikan kepada dua spesies luwak tersebut kemudian dianalisis menggunakan metode TLC Scanner secara kuantitatif. Proses Ekstraksi Kopi luwak robusta dilakukan dengan  menggunakan pelarut diklorometan dan disonikasi selama 15 menit kemudian filtrate yang diperoleh disaring dengan kertas saring Whatman kemudian dipekatkan dengan rotary evaporator. Analisis kuantitatif dari kofein di ukur dengan menggunakan Kromatografi Lapis Tipis ( KLT ) kemudian di Scanning dengan alat  Densitometri (TLC Scanner) sehingga diperoleh kadar kofein dari kopi luwak tersebut. Biji Kopi Robusta yang diberikan pada Spesies luwak Bulan ( Arctictis Binturong ) mempunyai kadar kofein 1, 91 % dimana kadar kofein ini lebih tinggi jika dibandingkan dengan kopi robusta yang diberikan kepada luwak pandan (Paradoxurus Hermophroditus) yang hanya memiliki kadar kofein 1, 30 %. Berdasarkan analisis SPSS 21 menggunakan Anava tiga Jalan diketahui bahwa perbedaan spesies luwak mempengaruhi kadar kofein kopi luwak robusta.

Abstract

Telah dilakukan penelitian “Analisis Merkuri (Hg) pada Beberapa Merek Krim Pemutih Wajah dengan Metode Spektrofotometri Serapan Atom (SSA)”. Tujuan penelitian yang dilakukan untuk mengidentifikasi dan mengukur kadar merkuri dalam beberapa merek krim pemutih wajah yang dijual di pasaran Kota Bukittinggi. Sampel diekstraksi dengan cara destruksi basah selama 3 jam pada suhu ±100°C sampai diperoleh larutan bening. Analisa kualitatif merkuri menggunakan KI 0,5 N dan NaOH 2 N. Kemudian, penentuan merkuri dalam sampel menggunakan Spektrofotometri Serapan Atom Uap Dingin. Dari hasil validasi metode diperoleh persamaan regresi Y = 0,0072x – 0,0089. Hasil penelitian menunjukkan bahwa dari kelima sampel yang diuji mengandung merkuri dengan rata-rata kadar sampel A = 0,00087 ppm, B = 0,00075 ppm, C = 0,00066 ppm, D = 1,66794 ppm, dan E = 0,00189 ppm. Sampel D mengandung merkuri di atas batas aman yang diperbolehkan BPOM RI yaitu tidak melebihi 1 mg/L atau 1 ppm. This research has been done Analysis of Mercury on Several Brands of Whitening Creams by using Atomic Absorption Spectrophotometry Method. The purpose of this research was to identify and measure mercury consentrations on five samples of whitening cream in the market Bukittinggi City. The samples was extracted by wet destruction for 3 hours at ± 100 °C until the solution clear. Qualitative analysis was performed using color test KI 0.5 N and NaOH 2 N. Then, the determination of mercury in the samples by using Cold Vapour Atomic Absorption Spectrophotometry. The regression equation obtained from the calibration curve is Y = 0,0072x – 0,0089. The results of the five samples showed that all samples contained mercury with average level samples A = 0,00087 ppm, B = 0,00075 ppm, C = 0,00066 ppm, D = 1,66794 ppm, dan E = 0,00189 ppm. Sample D contained mercury above the safe limit allowed by BPOM RI that is not more than 1 mg/L or 1 ppm.
