Abstract

The qualitative and quantitative analysis of coumarin in cinnamon (Cinnamomum burmanii Bl.) which are qualified in to AA, KA and KB has been done by gas chromatography and gas chromatography-mass spectrometry. Extraction of quality KB coumarin was carried out by soxchletation in hexane, dichloromethane and methanol. Meanwhile, quality AA and KA coumarins are soxchletated in methanol. The qualitative analysis using gas chromatography-mass spectrometry with column BP-5 showed that hexane, dichloromethane and methanol extracts of qualified KB coumarin, as well as methanol extracts of qualified AA and KB only contain coumarin compounds, no other coumarin groups were found. From coumarin quantitative analysis by gas chromatography using BP-624 column and FID detector of methanol extracts obtained 1855.51 ppm (0.186%), 1915.75 ppm (0.192%), 9177.36 ppm (0,918%), for qualified KB, AA, and KA respectively.

Abstract

The aim of this study was to evaluate an antiinflamation effect of S.paniculata and Siabadicencis extracts at the dose of 300 mg/200 g BW by induction of rats paw edema. Edema condition was induced by administration of 0,2 ml aqueous solution of 0,1 % carrageenan. This was followed by the assay of antiinflamatory effect of fractionated S. paniculata extract with hexane, DCM, ethyl acetate and butanol respectively. A decrease in rats paw edema volume was measured after treatments. Results showed that percent inhibition of inflammation after administration of Siabadicencis, S. paniculata extracts and positive control were 35;48 and 63 % respectively, while that of the hexane, DCM, ethyl acetate and butanol fraction was 69, 27, 53, and 43 % respectively. The antiinflamation effect of the hexane fraction was then determined at various doses (10 ; 20 and 30 mg/20 g BW with positive control acetosal 1,3 mg/20 g BW, by induction of rat back edema. The edema was induced by administration of 1 % carrageen in sessame oil at day 0 (0,1 ml) and day 1 st (0,5 ml) respectively. Parameters such as edema volume, haemoglobin and haematocrit were observed. Results indicated that percent inhibition of inflammation diameter after administration of the fraction at doses  10, 20, 30 mg/20 g BW and acetosal 1,3 mg/20 g BW on day 5th was 11 ; 15 ; 17 ; and 20 % respectively. While percent inhibition of inflammation volume of those groups were 19, 38, 62 and 63 % respectively the haemoglobine (Hb) of dose group 10, 20, 30 mg/20 g BW and negative control was 14.808 ; 15.117 and 12.944 g/dL respectively and 45,25 that of control positive 44,2 respectively. The mean value of necrosis hepatocyte at 3 sites of dose group 30 mg/20 g BW, positive and negative control groups was 6,38 ; 13,82 and 3,55 respectively.

Abstract

Caffeine analysis from Luwak coffee in different types of coffees, Luwak species and processed by using TLC Scanner method had been done. 8 samples of Luwak coffee, that is R.Ab.K ; R.Ab.B ; R.Ph.K ; R.Ph.B ; A.Ab.K ; A.Ab.B ; A.Ph.K ; A.Ph.B which were extracted by dichloromethane (DCM). TLC separation had been done through stationary phase with silica gel GF254 and mobile phase are solute of dichloromethane (DCM) : methanol (9,5 : 0,5). Caffeine content had been counted by using regression equality y = 2715,112 + 4,389x and giving percentage in succession 1,91 ; 2,04 ; 1,30 ; 1,89 ; 1,29 ; 1,45 ; 1,25 ; 1,42 %. The highest of caffeine had been given by Robusta coffee which has been given before to A. binturong luwak and processed wetly 2,04 %. The lowest of caffeine content had been given to Arabica coffee which has been given before to P. hermophroditus luwak 1,25 %. Three way – Anova statistic SPSS 21 tabulation had showed the result that the kind of coffee, variety of Luwak and the way of processing gave significant value toward caffeine content in p < 0,05.

Abstract

Chili sauce sachet A, B and C containing synthetic red coloring agent  were taken from three fast food premises in Padang. Synthetic red coloring agent  were a food additives used by food manufacturers to enhance color of food products. The use of food coloring agent in Indonesia were regulated   in The Rule of Head of the Supervisory Food Board of the Republic of Indonesia Number 37 Year 2013 about the Limit Use of Food Coloring Agent. Thin-layer chromatography (TLC) -densitometry was  a precision method for the analysis of food additives. Development and validation of TLC method for chromatographic separation used GF254 silica plate eluted with a mixture of ethanol: butanol: distilled water (4: 5: 5) and spots were detected visually. A red spots on the sample B was identified as Ponceau 4R with Rf value of 0.76. It was then followed by quantitative analysis using densitometry. The linearity of the method  was found in the range of 2-10μg/ml with a correlation coefficient of 0.994. Precision intra-day and inter-day relative standard deviation was shown from 1.11% and 2.69% respectively. Accuracy of the method was shown on the percentage of recovery of the 3 different concentrations of solution which gave the average percentage of 108.17%. Limit of detection and limit quantitation was 0.8306μg/ml and 2.7687μg/ml.  The concentration of Ponceau 4R in the sample B was 11.9520mg/kg of material. This value was not exceeded the  maximum concentration of food coloring agent as stated in the rule which should not be above 70mg/kg of material.

Abstract

Agarosa merupakan salah satu biopolimer yang sering digunakan dalam bidang bioteknologi, diantaranya sebagai matriks pada kultur sel. Penelitian ini bertujuan untuk mengaplikasikan agarosa hasil isolasi dari tepung agar sebagai matriks kultur 3D pada sel kanker paru line A549. Hasil isolasi diperoleh Agarosa 1 (A1) dan Agarosa 2 (A2) hasil pemurnian dari A1. A1 dan A2 dengan konsentrasi 5% diaplikasikan sebagai matriks kultur 3D di dalam well plate kultur sel kanker paru line A549 dengan densitas 107, sebagai kontrol pertumbuhan digunakan kultur 2D sel (well plate tanpa matriks) dan kontrol positif kultur 3D sel pada matriks agarosa Top Vision®. Viabilitas sel diamati pada hari ke 2, 4 dan 6 menggunakan pewarna trypan blue. Proliferasi sel diamati pada hari ke 6 menggunakan reagen MTT. Hasil uji viabilitas terlihat agregat sel 3D pada matriks agarosa yang viable dan tidak menyerap warna trypan blue. Viabilitas sel dibuktikan dengan uji proliferasi dan diperoleh nilai rata-rata absorban pada kultur 2D, kultur 3D matriks kontrol, A1 dan A2 masing-masingnya 2.03; 1.21; 0.96 dan 1.22. Proliferasi kultur 2D lebih baik dari 3D, akan tetapi proliferasi kultur 3D pada matriks A2 lebih baik dari matriks kontrol.
