Abstract

Berdasarkan studi in vitro, in vivo dan uji klinis, sambiloto (Andrographis paniculata (Burm.f.) Nees) terbukti memiliki aktivitas sebagai antihiperglikemia. Namun, mekanisme aksi dan zat aktif yang bertanggung jawab terhadap aktivitas antihiperglikemia dari sambiloto belum diketahui secara pasti. Penelitian ini bertujuan untuk mengetahui interaksi kandungan kimia dari daun sambiloto dengan beberapa target molekuler yang terlibat dalam penyakit diabetes mellitus (DM) serta prediksi parameter farmakokinetik dan toksisitas dari senyawa-senyawa tersebut. Pada penelitian ini sebanyak 14 kandungan kimia daun sambiloto ditambatkan pada 6 target molekuler yaitu aldosa reduktase, α-glukosidase, protein tyrosine phosphatase 1B, peroxisome proliferator-activated receptor gamma, reseptor insulin dan glycogen sintase kinase 3B menggunakan metode docking molekuler yakni Autodock 4.2 yang sebelumnya telah divalidasi. Senyawa kimia dari daun sambiloto yang memiliki interaksi terbaik, selanjutnya diprediksi profil farmakokinetik dan toksisitasnya menggunakan SwissADME dan Toxtree. Berdasarkan hasil docking molekuler dapat diketahui bahwa senyawa yang diprediksi memiliki afinitas pengikatan terbaik dengan target molekuler DM adalah 19-O-asetilanhidroandrografolid, β-sitosterol, neoandrografolid, daukosterol dan asam oleanolat. Hasil prediksi SwissADME menunjukkan bahwa kelima senyawa ini memiliki peluang untuk menjadi obat oral, beberapa diantaranya merupakan substrat Pglikoprotein dan mampu menembus sawar darah otak. Sementara hasil prediksi Toxtree menunjukkan toksisitas yang rendah sampai tinggi dan semua senyawa tidak berpotensi dalam menyebabkan karsinogenisitas baik melalui mekanisme genotoksik maupun non genotoksik.

Abstract

Daun kersen (Muntingia calabura L.) merupakan tanaman yang dapat digunakan sebagai alternative pengobatan kanker. Penelitian ini bertujuan mengetahui aktivitas sitotoksik ekstrak etanol daun kersen, fraksi n-heksan, fraksi etil asetat dan fraksi air ekstrak daun kersen terhadap sel T47D dan mengetahui pengaruh ekspresi protein p53 dan Bcl-2 pada pemberian fraksi aktif daun kersen. Daun kersen diekstrak dengan metode maserasi menggunakan pelarut etanol 70%. Ekstrak etanol kemudian difraksinasi dengan partisi cair-cair. Uji sitotoksik dilakukan pada sel kanker payudara T47D dan sel vero dengan metode MTT assay dan dibaca absorbansinya menggunakan ELISA reader. Untuk mengetahui pengaruh ekspresi protein p53 dan Bcl-2 dilakukan uji imunositokimia. Hasil penelitian ini menunjukkan bahwa fraksi n-heksan daun kersen memiliki aktivitas sitotoksik potensial terhadap sel kanker payudara T47D, sedangkan pada ekstrak, fraksi etil asetat, dan fraksi air memiliki aktivitas sitotoksik moderat. Fraksi n-heksan daun kersen mampu meningkatkan ekspresi protein p53 dan Bcl-2 pada konsentrasi 6,79 - 27,16 µg/mL.

Abstract

Curcumin was reported can suppress the growth of blood tumor cells. This study aims to determine the physical and chemical properties of curcumin derivatives that give affect to antitumor activity of blood cells which were expressed as the best HKSA equation model. This   study   used   curcumin   and   twelve   derivatives   which   have   antitumor   activity parameters of blood cells (K562) as IC50. Parameters of physicochemical properties of curcumin compounds series and their derivatives as descriptors include net charge of atoms in the curcumin framework structure, EHOMO, ELUMO, Log P, E Hid, a, MR, Volume, Surface area (SA), Total Energy, dipole moment (u), Hf, and Mass which are the results of geometric optimization calculations using the PM3 semiempirical method with hyperchem application. HKSA equation model was built using the BuilQSAR statistical calculation analysis. The best HKSA equation model is then validated internally and externally. Calculation of statistical analysis with BuldQSAR, the descriptor that has the most influence is  obtained, namely net  atomic charge, EHOMO,  surface area, dipole moment and  Hf with  linear regression equation: Log 1/IC50 : + 1.7622 (± 0.3655) qC4 - 0.9457 (± 0.1047) E Homo + 0.0013 (0.0002) SA + 0.0575 (± 0.0216) u - 0.0068 (± 0.0004) Hf - 11.4161 (± 1.0440) , n = 10; R = 0.999; R2= 0,9990; SPress = 0.0232. as well as non-linear regression equation : Log 1/IC50 = + 15.1400 (±4.9008) qC7 + 7.4959 (± 5.6081) qC8 - 0.5839 (± 0.1397) qC15 + 0.0005 (± 0.0003) SA - 0.0054 (±0.0008) Hf - 5.7246 (±3.2476), n = 10; R = 0,999, R2 = 0,9977; SPress = 0.0341.

Abstract

The residents of Karangturi in Triyagan village, located in Mojolaban sub-district of Sukoharjo regency, require further training to enhance family health care using readily available traditional materials. They also need motivation to start home-based industries to boost entrepreneurship and the local economy. Hence, there is a necessity for educational and training programs focused on making Natural Herbal Soap.This community service initiative is divided into two segments:Firstly, providing education on the health benefits of traditional natural ingredients. Secondly, offering training on creating Natural Herbal Soap by combining oils (olive oil and coconut oil) with an alkaline solution (NaOH), which will produce a thick liquid known as trace. This mixture is then blended with essential oils (green tea, chamomile, lemongrass) and left to solidify.The objective of these activities is to equip the community with the skills to make Natural Herbal Soap easily and with low capital investment. This skill can be a viable option for enhancing family entrepreneurship and improving daily family health.

Abstract

Eyebrow pencil is a cosmetic that is used to correct less symmetrical eyebrows. The colors marketed are very diverse. Lead is also obtained from tools used during the production process. This study aims to determine the lead content in eyebrow pencils and to determine the lead levels in eyebrow pencils circulating in Tulungagung by atomic absorption spectrophotometry (AAS) still meets the requirements of BPOM No. 12 of 2019. Sampling using a purposive sampling technique. Preparation samples were carried out using the dry deconstruction method. The research began with chemical and physical qualitative analysis. The determination of lead levels using AAS at a wavelength of 283.3 nm was then determined. The AAS analytical method is suitable for lead based on the results of method verification meeting the requirements including linearity with a value of r = 0.9929, accuracy of 102%, LOD 0,1189 mg/L, but did not meet the LOQ of 0.3964 mg/L. The results showed that 3 out of 5 samples were positive for lead with levels ranging from 4.9339-5.4896 mg/kg. The results still meet the BPOM contamination limit requirements, which are not more than 20 mg/kg, thus all samples are still within safe limits according to the requirements set by BPOM.

Abstract

Diabetes mellitus is a metabolic disorder characterized by hyperglycemia. The development of diabetic complications plays a pathological role in increasing Reactive Oxygen Species (ROS) levels, which induce oxidative stress. The aim of this study was to examine the antidiabetic activities of kemuning and moringa leaves based on glucose, SOD, and GPx levels, and the protective activity of pancreatic cells in STZ-NA-induced rats. The kemuning and moringa leaves were dried, finely powdered, and extracted by the remaceration method using 96% ethanol. The test was carried out on 30 rats that were divided into 6 treatment groups, namely the normal control group, negative control, positive control (glibenclamide), extract dose of 76.5 mg. /200 g BW : 6.5 mg/200 g BW, 114.75 mg/200 g BW : 3.25 mg/200 g BW and 38.25 mg/200 g BW : 9.75 mg/ 200 g BW. The antidiabetic test was carried out using STZ-NA, glucose levels were measured, and SOD and GPx levels in the liver supernatants were measured. Cell protection was tested using the cell damage score method. The data obtained were analyzed using one-way ANOVA variance. The results showed that kemuning and moringa leaves at a dose of 38.25 mg/200 g BW: 9.75 mg/200 g BW were able to lower blood glucose levels, increase the activity of SOD and GPx, and protect pancreatic cell damage in STZ-NA rats.
