Abstract

This study aims to analyze the parameters of physics, chemistry and metal content of mercury and determine the condition of water quality around the Bay of Manado. The tools used in this study consisted of a sample bottle, thermometer, pH meter, pen and camera. Research materials include water samples, aqua, ice cubes, tissue. The Sampling technique using sampling methods “purposive random” with 4 point sampling locations. Namely the location of the sampling site 1 (Manado Harbor), location 2 (region MTC), location 3 (region Prof. Dr. Kandouw Hospital) and location 4 (Malalayang coastal areas). Samples were analyzed in the Laboratory of Industrial Research Standardization Manado (BARISTAND). The results of the analysis of mercury in waters around Manado Bay using Atomic Absorption Spectrofotometer (AAS) obtained results undetectable (nil). The value of the analysis the temperature at the point I – IV range from 28 0C– 300C and pH values ranging from 7 – 9. The condition of water at four point research is still below threshold quality Standards Minister of Environment No. 51 2004 (0,001 mg/l). Abstract

The aim of this study was to know the  applied technology and the production value of the aquaculture activities in the waters of  Lake Tondano and to identify factors influencing aquaculture production in Tondano lake. Data collected  consisted of primary and  sekunder data.  Primary data  were collected by conducting a survey to fish-farmers as respondents around the lake Tondano.  Operational aquaculture data were obtained  by interviewing the fish farmers and by observation.  Secondary data were collected from various  institution  such as Minahasa  Fisheries  Office  ,  Office of  Village Head and Fish-farmer  Group  Administrative Centre.  Collected data were  descriptively analyzed  and presented in the form of tabulation and various model of diagram.  Fluctuated data in aquacultural production value  and the number of  operated  KJT  during the periood of  2001 to  2014 was analysed by using linear regression  and correlation. The aquacultral production value  of the lake Tondano in  2014 was  estimated only 1227,3 tons /year.  The number of operated  KJT  only remained 1203 units.  The production value decreased consistently  from 2003 to 2014.   Statistical analysis showed that during  2001-2014 there was a reduction about 1.236 ton every year in aquacultural production, and  607 units of operated KJT.  There was a  very strong correlation  between  time  (year) and the  the aquacultural production, and also between  time (year) and  the number  of  operated KJT.  There were 3 important  factors  influencing the aquacultural production value of lake Tondano, which were: environmental, technical and economic factor.   However, the primary factor  was  ‘economic’ factor.  The  ‘very high price’ of the fish formulated diet  and  the "very low price”  of  the produced fish were the main causes of the problem.   The fish farmers could not afford the invesment and operational cost.   Environmental degradation of  Lake Tondano waters  also significantly  influenced the operation of aquacultural activities.   Keywords:  Production, technology, aquaculture, Lake Tondano

Abstract

One type of enzyme whose application is very widespread is protease enzymes. Microorganisms that have not been widely explored as a source of protease enzymes are bacteria that are in conjunction with Nudibranch. The study aims to identify the morphological characteristics of bacteria, then determine the hydrolysis ability of protease enzymes from bacteria symbiont Nudibranch. Identification of morphological characteristics is done macroscopic, and microscopic with gram staining. Proteolytic activity tests are performed qualitatively by measuring the proteolytic index of bacteria grown on Skim Milk Agar medium. The results of macroscopic identification of symbiotic bacteria with Nudibranch are dominated by irregular colony shapes, undulated edges, growth of the entire colony has a flat height, milky white colony color, buttery texture and opaque bacterial colonies. Based on microscopic identification, two gram-negative isolates are obtained, namely RL.S14, S1.1.1, and gram positive isolates, namely AK.S00, AK.S01, AK.S02, AK.S03, S.PP1, RL.S6, S2.1.5, S2.2.1, S3.3.1, S3.3.4, RL.S10. Nudibranch symbiont Bacterial has colony cells were dominated by the diplobacillus form. The results of the protease activity test of Nudibranch symbiont bacteria showed that 12 positive isolates had proteolytic activity with the highest proteolytic index produced by isolate AK.S00, which was 2.5 mm. Keywords: Nudibranch, symbiotic bacteria, characteristics, protease

Abstract

This study was conducted to determine the level of aquaculture production Tondano and determine the impact of the production level of the waters of Lake Tondano nutrifikasi . The data collected in this study , consisting of primary data and secondary data . Production data obtained from the questionnaire and interviews to farmers fish there , as well as data obtained from the Department of Fisheries and Village Head Office . Water sampling to determine levels of phosphate in the waters of the lake . Water sampling conducted on six (6 ) locations: Village Ranomerut , Eris Village , Village Paslaten , Leleko Village , Village and Village Paleloan Toulour . Sampling was conducted at six (6 ) points to pay attention to the position of the inlet and outlet of the waters of Lake Tondano . Tondano aquaculture production in 2014 was estimated at 1222.12 tons / year . Number KJT operated as many as 978 units . There was a decrease in aquaculture production annually over a period of 2001 - 2014. Followed by the declining number of KJT on Lake Tondano . Phosphates were donated to the waters of fish farming activities from the rest of the feed pellets . The content of orthophosphate in the waters of Lake Tondano of 0.06 mg / l means the waters of the lake are in a safe condition Tondano not pass the specified limits ( PP RI No. 82 of 2001 ) . The decline in aquaculture production decline followed Tondano phosphorus content in Lake Tondano . Another factor that is expected to also affect the amount of water hyacinth around the lake so that the phosphorus content is also on the wane . keywords : Lake Tondano, Production, Nutrification
