Abstract

Sea cucumber (H.Scabra)is a species of marine biota providing high economicaland medical values. It has been hundreds years consumed for drug to cure various diseases in China. The aim of this research was to determine the level of the toxicity against brine shrimp larvae A.salina Leach in each dried sea cucumber (H.scabra) extract collected from Kenjeran Coast, Surabaya. The information would be beneficial for anticancer drug discovery .The research was conducted by extracting samples using both ethanol and n-hexane solvents.The concentrated extract obtained then were used for the toxicity test against brine shrimp larvae of A.salina Leach (BSLT). The mortality data of A.salina Leach was analysed using probit analysis to determine the value of LC50 on each extract. The phytochemical assay and thin layer chromatography (TLC) was also apllied for both extracts.The result showed that both ethanol and n-hexane exctracts performed toxicity level against brine shrimp larvae of A.salina Leach with LC50less than 1000 ppm.The n-hexane extract indicated LC50 of 189.093 ppm, higher than the ethanol extract, 286.031 ppm. The presence of steroids were detected in both extract whilst the alkaloids were only detected in the ethanol extract. Further separation of the n-hexane exctrat using TLC in n-hexane;ethyl acetate;amonia (66:33:0.8) yielded 5 spots with the Rf of 0.3;0.44;0.54;0.72 and 0.78, respectively.

Abstract

IIndonesia is the biggest country in the production of red seaweed of Eucheuma spinosum. The red seaweed has bioactive compounds that have a potential activity such as phenolic compounds as well as carrageenan and pigments. This paper reported phytochemical analysis of E. spinosum harvested by a local farmer in Sumenep Island, East Java and free-radical scavenging activity (FRSA) derived from 2,2’-diphenyl-1-picrylhdrazyl (DPPH) for several organic solvents. For extraction, dried powder is added with 5.0 mL of various solvents following by ultra-sonication assisted extraction for 30 minutes. The extract was separated by centrifugation for phytochemical analysis and radical scavenging evaluation. The prospecting of dichloromethane, ethyl acetate, and n-hexane extracts indicated the potency for radical scavengers. Alkaloids, terpenoids, and saponins were secondary metabolites that indicated the presence in the extracts. The best IC50 value was presented by ethyl acetate extracts (384,86 ppm) with 38.78% for 50 ppm, while IC50 values of n-hexane, methanolic, dicloromethane extracts were 410.12, 677.76 and 685.08 ppm, respectively. Keywords: Eucheuma spinosum, phytochemical analysis, radical scavenging activity

Abstract

Abstract River is a natural resource that has a major contribution to human life. Utilization of river water in daily life activities can cause a decrease in water quality. Water quality that is lower than the standard categorized as pollution that causes health risks. River water quality can be carried out by using BOD (Biological Oxygen Demand) and COD (Chemical Oxygen Demand) test parameters. The results of calculating the water quality of the Dhurbugen River obtained a BOD value of 6.448 mg/l and a COD value of 57.6 mg/l. The results of this study concluded that the water quality of the Sumenep Dhurbugan River included in the safe category for water parameters that are in class III and IV based on its utilization function.

Abstract

The Patrean River is one of the rivers that flows in several locations in Sumenep District including Manding, Batuputih, Dasuk, Gapura and Kota sub-districts. This river is heavily flowed by domestic waste and industrial waste. The presence of such waste can cause a decrease in water quality characterized by an increase in Biological Oxygen Demand (BOD) and Chemical Oxygen Demand (COD). The purpose of this study was to determine the BOD and COD content at three locations of the Patrean river, and to assess the pollution status using the pollution index method based on Government Regulation No. 82 of 2021. Sampling was carried out in three locations (upstream, middle, and downstream), this research method used survey and experimental methods. The results of the calculation of COD values in Patrean River waters at upstream, middle and downstream points are 12.75 mg/l, 19.12 mg/l, and 25.50 mg/l, respectively. BOD values at three points are 9.02 mg/l, 16.11 mg/l and 20.94 mg/l.

Abstract

AbstractEucheuma spinosum is a type of red algae (Eucheuma) which is used as a food which has a high source of carrageenan. Information on secondary metabolites in E. spinosum is known to have bioactive properties as an antioxidant and antibacterial. The purpose of this study was to isolate and characterize secondary metabolites in E.spinosum. The study was started by isolating secondary metabolites from E.spinosum using Soxhlet extraction followed by solvent partitioning, fractionation and characterization. The rendemen from methanol extract was 6.07% while the highest partition result was in the ethyl acetate fraction, which was 1.25%. Fractionation was carried out by column chromatography resulting in 2 single spots F401 and A402. Pure isolate F401 has an Rf value of 0.71 in the form of a white solid, A401 has an Rf value of 0.67 in the form of a brown gel. The results of characterization of isolates with a UV-Vis spectrophotometer obtained maximum absorption and absorbance values of F401 218.50 nm 4.79 M-1 cm-1, 273 nm 1.06 M-1 cm-1 respectively. Isolate A402 233.50 nm 4.1 M-1 cm-1, 407 nm 1.03 M-1 cm-1. Identification using FT-IR found that both isolates showed the presence of functional groups =C-H, C-C, C=O, C-H. According to the result of the UV and FT-IR spectra, two isolates can be identified as terpenoid compounds. Keywords: E.spinosum, Secondary Metabolites, Ethyl acetate fraction, UV-VIS, FT-IR
