Abstract

Amentoflavone (1), a biflavonoid was isolated from leaves of Calophyllum inophyllum Linn. Isolation and purification of the compound used maceration and chromatography methods using Si-gell and sephadex LH-20. The structure was determined by UV, IR, 1H and 13C NMR spectroscopy includes HMQC/HMBC analysis and comparison with references.

Abstract

A xanthone derivative namely trapezifolixanthone has been isolated and identified from EtOAc extract of stem bark ofCalophyllum soulattri. The maceration method was used to isolates the trapezifolixanthone then followed by fractionation method using vacuum liquid chromatography and purification process by flash chromatography. A 14 mg of yellowish oily solid was extracted. The compound was elucidated by 1H NMR, 13C NMR, and 2D NMR (HSQC, HMBC, COSY) supported by UV and IR spectroscopy. Base on spectral data and comparison with some references, it is  known that the compound is trapezifolixanthone.

Abstract

Satu senyawa biflavonoid yaitu amentoflavon (1) berhasil diisolasi dari daun tumbuhan nyamplung (Calophyllum inophyllum Linn). Ekstraksi menggunakan metode maserasi dan pemurnian dengan kromatografi kolom dengan fasa diam Si-gel dan sephadex LH-20. Elusidasi struktur menggunakan metode spektroskopi UV, IR, 1H dan 13 C NMR beserta HMQC dan HMBC. Abstract

Telah dilakukan uji toksisitas secara Brine Shrimp Lethality Test (BSLT) ekstrak buah merah (Pandanus Conoideus Lamk.). Uji toksisitas dilakukan pada ekstrak etanol dan n-heksana. Hasil uji BSLT menunjukkan rata-rata prosentase kematian ekstrak n-heksan lebih tinggi dibandingkan dengan ekstrak etanol, sebesar 54 % diukur pada konsentrasi 400 (μg/mL). Ekstrak n-heksan selanjutnya dilakukan pemisahan secara kromatografi kolom dengan elusi secara gradien menggunakan pelarut campuran n-heksana-etil asetat untuk memperoleh fraksi aktif. Uji toksisitas secara BSLT menunjukkan fraksi aktif mempunyai nilai LC50 sebesar 138,05 μg/mL. Identifikasi fraksi aktif diperoleh 3 komponen senyawa asam lemak yaitu asam palmitat, asam oleat dan asam miristat.Kata Kunci : Buah merah (Pandanus conoideus Lamk.), identifikasi, toksisitas, Brine Shrimp LethalityABSTRACT

Abstract

Chemical isolation and identification of root bark of Calophyllum soulattri Burm. f. have been conducted. Isolation was conducted by maceration with ethyl acetate as solvent. Ethyl acetate extract was separated and purified by vacuum liquid chromatography and flash chromatography which was guided by thin layer chromatography to obtain yellowish white solid. Isolated compounds were determined by spectroscopy methods such as UV, FTIR, 1H NMR, 13C NMR, DEPT, HSQC, HMBC and were compared to the references. Based on the analysis result it’s known that the compounds are the mixture of β-sitosterol and stigmasterol.

Abstract

A xanthone, named ananixanthone (1) has been isolated and identified from the ethyl acetate extract of the root barks of Calophyllum soulattri. Structure of the compound was determined based on spectroscopic data, including UV, IR, NMR 1D, NMR 2D and by comparison with references.

Abstract

The purpose of this research was to evaluate antibacterial activity of Neem (Azadirachta indica A. Juss) leaves fraction against some pathogenic bacterial and to identificate the most active fraction. Leaves powder of Neem was macerated with ethanol and factionated by vacuum liquid chromatography using hexane, ethyl acetate and ethanol as eluent, respectively. The antibacterial activity of the fraction was evaluated by diffusion method, then the most active fraction of antibacterial was evaluated by zone of inhibition. The most active fraction of antibacterial was evaluated for Minimum Inhibitory Concentration (MIC) and equivalent value, compared with amoxicillin and chloramphenicol, then identified using phytochemical screening and Gass Chromatography-Mass Spectroscopy (GC-MS).The fraction of Neem leaves fractionated by vacuum liquid chromatography had antibacterial activity againstStaphylococcus epidermidis, Bacillus cereus, and Shigella flexneri. The ethyl acetate fraction showed the most active fraction of antibacterial against all bacterial tested. Ethyl acetate fraction had MIC 0.075% againstStaphylococcus epidermidis 0.05% against Bacillus cereus and Shigella flexneri. The antibacterial activity of ethyl acetat fraction compared to amoxicillin was 0.01% for S. epidermidis, 0.02% for B. cereus, and 0.02% for S. flexneri. Then compared to chloramphenicol was 0.04% for S. epidermidis, 0.02% for B. cereus, and 0.06% for S. flexneri. The result of GC-MS showed that the ethyl acetate fraction contained palmitic acid, ethyl linoleolate, stearic acid, trans-phytol, DOP (di octyl phthalate), and suggested 1 class of compound fatty acid and 5 classes of compound triterpenoids.

Roxb with in vitro macrofag phagocytosis method. . Suji is a plant that is often used as a food coloring and has long been used medicinally. This study aimed to test in vitro macrophage phagocytosis on ethanol extract of suji leaf. The suji leaves are extracted by maceration method. The obtained viscous extract was then tested for Thin Layer Chromatography (TLC) and in vitro macrophage phagocytosis test. The parameters used are Phagocytosis Index and Macrophage Capacity. The results of in vitro macrophage phagocytosis test with 4 concentrations of 10.25,50 and 100 microgram / ml, showed for the largest average Fagocytosis (IF) Index shown at concentration 10 mcg / ml, while concentrations 25 and 50 mcg / ml showed values The same phagocytic (KF) capacity. Keywords :  Suji leaf, Phagocytic Macrophage Activity,  ethanolic  extract   ABSTRAKUji aktivitas imunomodulator telah dilakukan terhadap ekstrak etanol  daun suji (Dracaena angustifolia (Medik.)Roxb..  Suji merupakan tanaman yang sering digunakan sebagai pewarna makanan dan telah lama digunakan untuk obat. Penelitian ini bertujuan untuk melakukan uji fagositosis makrofag in vitro terhadap ekstrak etanol daun suji. Daun suji diekstraksi dengan metode maserasi. Ekstrak kental yang diperoleh kemudian dilakukan uji Kromatografi Lapis Tipis (KLT) dan uji fagositosis makrofag in vitro. Parameter yang digunakan adalah Indeks fagositosis dan Kapasitas makrofag. Hasil uji fagositosis makrofag in vitro dengan 4 konsentrasi sebesar 10,25,50 dan 100 mikrogram/ml, menunjukkan untuk Indeks Fagositosis (IF) rata-rata terbesar ditunjukkan pada konsentrasi 10 mcg/ml, sedangkan konsentrasi 25 dan 50 mcg/ml menunjukkan nilai  Kapasitas Fagositosis (KF) yang sama besar. Kata kunci : Daun suji, fagositosis makrofag, ekstrak etanol

Abstract

Anak-anak termasuk ke dalam kelompok usia yang sangat rentan terhadap penularan COVID-19 dari lingkungan sekitarnya. SDIT Insan Mulia Surakarta merupakan sekolah yang berada di zona merah dan sedang mempersiapkan kegitan tatap muka. Pengetahuan tentang Perilaku Hidup Bersih dan Sehat (PHBS) merupakan prioritas yang harus diberikan dan difahamkan kepada siswa agar dapat menjadi salah satu upaya perlindungan siswa dari COVID-19. Selain itu, edukasi tentang obat sejak dini perlu dilakukan agar nantinya tidak timbul permasalahan dalam penggunaan obat. Kegiatan pengabdian berupa edukasi dengan media handout dan buku ajar berbasis aktivitas dilakukan sebagai salah satu upaya peningkatan pengetahuan siswa. Sasaran kegiatan adalah siswa kelas 3 di SDIT Insan Mulia Surakarta pada bulan April 2021. Dalam memahami materi edukasi, siswa dipandu melalui pertemuan virtual dengan platform zoom meetings. Evaluasi keberhasilan program dilakukan melalui penilaian pengetahuan dengan pretes dan postes. Perbedaan nilai pretest dan postest dianalisis menggunakan uji Wilcoxon. Pada akhir rangkaian kegiatan hanya ada 37 siswa yang mengisi postes dengan lengkap. Hasil evaluasi menunjukkan bahwa ada perbedaan pengetahuan yang signifikan sebelum dan sesudah edukasi menggunakan buku ajar berbasis aktivitas dengan nilai p=0,047. Akan tetapi, tidak ada perbedaan yang signifikan pada pengetahuan siswa yang diberikan edukasi melalui media handout dan buku ajar berbasis aktivitas ( p=0,777.; p0,05). Media edukasi buku ajar berbasis aktivitas lebih tepat digunakan dalam meningkatkan pengetahuan siswa sekolah dasar. Kendala yang dialami selama kegiatan adalah jaringan internet dan fasilitas yang dimiliki siswa. Secara keseluruhan, kegiatan ini telah berjalan dengan lancar  dan dapat meningkatkan pengetahuan siswa tentang obat dan PHBS. Melalui media edukasi yang telah disusun oleh tim pengabdian, diharapkan edukasi tentang obat dan PHBS terhadap siswa dapat terus dilakukan secara berkala. Â
