Abstract

Abstract: Companies need a strategy to determine the number of occupational accidents and occupational diseases with risk management. HIRADC (Hazard Identification Risk Assessment and Determining Control) is one of the key elements of a safe work plan that can minimize cases of occupational accidents and occupational diseases. This study aimed to determine the potential hazards and risk levels of the oilmill and maintenance work area of PT. X international crude oil companies in North Sulawesi, risk control and application of control methods in the field. This was a qualitative study for the application of HIRADC method conducted in South Minahasa Regency. Recruitment of informants based on purposive sampling as many as 11 employees at oilmill and maintenance area as the main informants, and EHS Supervisor as the key informant and the Head of Department (HOD) as additional informants. The instruments of this study were the in-depth interview matrix and the HIRADC matrix. Data were obtained by observation, in-depth interviews and company document, then were analyzed with HIRADC method. The results obtained 68 types of hazards including physical, chemical, biological, ergonomic, and psychological hazards. The risk levels of hazards were high risk (H 35.2%), medium risk (M 58.8%), and low risk (L 5,8%) hazards; no hazard at the extreme risk level (E). The identification of highlighted hazards was the potential hazard of noise, heatstress, welding and grinding dust, with the highest levels of risk were the M risk (58.8%) and the H risk (35.2%). In conclusion, it is necessary to take control of measures according to the hierarchy of control in the form of substitution, engineering control, administration and personal protective equipment (PPE). Keywords: potential hazard; risk level; Occupational Health and Safety; Hazard Identification, Risk Assessment and Determing Control (HIRADC)   Abstrak: Perusahaan memerlukan strategi untuk menentukan angka kecelakaan kerja dan penyakit akibat kerja dengan manajemen risiko. HIRADC (Hazard Identification Risk Assessment and Determining Control) ialah salah satu elemen kunci dari rencana kerja yang aman yang bisa meminimalisir kasus kecelakaan kerja dan penyakit akibat kerja. Penelitian ini bertujuan untuk mengetahui potensi bahaya dan tingkat risiko area kerja Oilmill dan Maintenance PT. X perusahaan minyak kelapa mentah bertaraf internasional di Sulawesi Utara, pengendalian risiko serta penerapan metode pengendalian di lapangan. Jenis penelitian ialah kualitatif untuk aplikasi metode HIRADC. Penelitian dilaksanakan pada salah satu perusahaan berskala internasional di Kabupaten Minahasa Selatan. Rekrutmen informan berdasarkan purposive sampling, sebanyak 11 orang yaitu karyawan area Oilmill dan Maintenance sebagai informan utama, serta EHS Supervisor sebagai informan kunci dan Head of Department (HOD) sebagai informan tambahan. Instrumen penelitian ini yaitu matriks wawancara mendalam dan matriks HIRADC. Data diperoleh dengan wawancara mendalam, observasi lapangan dan dokumen perusahaan, kemudian dianalisis dengan metode HIRADC. Hasil penelitian mendapatkan 68 jenis bahaya termasuk bahaya fisik, kimia, biologi, ergonomi dan psikologi. Tingkat risiko didapatkan rendah (L 5,8%), sedang (M 58,8%), dan tinggi (H 35,2%); tidak terdapat jenis bahaya tingkat risiko ekstrim (E). Identifikasi bahaya yang menjadi highlight ialah potensi bahaya kebisingan, heatstress, debu pengelasan dan gerinda dengan tingkat risiko terbanyak ialah tingkat M (58,8%) dan H (35,2%). Simpulan penelitian ini ialah diperlukan tindakan pengendalian sesuai hierarchy of control berupa substitusi, rekayasa engineering, administrasi dan APD (alat pelindung diri). Abstract

This research was conducted in Bulutui Village, West Likupang District, related to theproblems raised how is the solubility of Dissolved Oxygen (DO) and the pH of sediment-insertedwater at mangrove locations, and on the coast, then this research aims to find out how the solubility ofoxygen (DO), and acidity (pH) of sediment-inserted water in mangrove ecosystems and on the coastin Bulutui Village, West Likupang District. Based on the results of measurements that have beencarried out in Bulutui Village, West Likupang District, it was found that the value of the dissolvedoxygen content at the mangrove location was in the range of 0.36 – 1.98 ppm, with an average of0.98 ppm, the pH value ranged from 6.16 – 6.89 with an average value of 6.04. In coastal locations,the dissolved oxygen content ranges from 1.26 to 2.87 ppm, with an average of 2.09 ppm, the pHvalue ranges from 6.78 to 8.90, with an average value of 8.34. The statistical t test showed that therewas a difference between the dissolved oxygen values in the sedimentary water in the mangrovesand the coast, as well as the acidity (pH). Keywords: sediment; Inset Water, Dissolved Oxygen, pH Acidity ABSTRAK Penelitian ini dilakukan di Desa Bulutui Kecamatan Likupang Barat, berkaitan denganpermasalahan yang dikemukakan bagaimanakah kelarutan Oksigen Terlarut (DO) dan pH air sisipansedimen pada lokasi mangrove, dan di Pesisir pantai, selanjutnya penelitian ini bertujuan untukmengetahui bagaimana kelarutan oksigen (DO), dan keasaman (pH) air sisipan sedimen padaekosistem mangrove dan di pesisir di Desa Bulutui Kecamatan Likupang Barat. Berdasarkan hasilpengukuran yang telah dilakukan di Desa Bulutui Kecamatan Likupang Barat ditemukan nilai darikandungan oksigen terlarut pada lokasi mangrove adalah berkisar 0,36 – 1,98 ppm, dengan rataratanya 0,98 ppm, nilai pH berkisar 6,16 – 6,89 dengan nilai rata rata 6,04. Pada Lokasi pesisirdengan kandungan oksigen terlarut berkisar 1,26 – 2,87 ppm, dengan rata rata 2,09 ppm nilai pHberkisar 6,78 – 8,90, dengan nilai rata – rata 8,34. Uji statistika t test menunjukkan adanyaperbedaan antara nilai oksigen terlarut pada air sisipan sedimen di mangrove, dan pesisir, demikianjuga keasaman (pH). Abstract

Marine biota resources as a potential asset that can be used into various products including pharmaceutical products because they were a natural ingredient that very rich in biologically active compounds with a unique structure. Softcoral Lobophytum is one of the marine biota that has the secondary metabolites which can be useful in pharmacology field. The purpose of this study is to test the toxicity of the soft coral extract of Lobophytum sp as an anti-cancer biopotential. This research is using the Brine Shrime Lethality Test (BSLT) method, with laboratory experiments. Solfcoral samples were taken from Bunaken Island that using purposive sampling method. The test concentrations using 10, 100, 500 and 1000 ppm. The analysis toxicity using probit analysis for minitab program. The results showed that the increasing of concentration was followed by the increasing of the mortality number of the animal testing. The results of probit analysis obtained an LC50 value of 9.98 ppm. These results indicate that the bioactive substance of softcoral Lobophytum sp has the potential as an anti-cancer compound. Keywords: Soft coral, Lobophytum sp, anti cancer, Artemia salina ABSTRAK Sumber daya biota laut merupakan aset potensial yang dapat digunakan menjadi aneka produk termasuk di antaranya produk farmasi karena merupakan bahan alam yang sangat kaya senyawa aktif biologi dengan struktur yang unik. Softcoral Lobophytum salah satu biota laut yang memiliki metabloit sekunder yang dapat bermanfaat dalam bidang farmakologi. Tujuan dari penelitian ini bertujuan untuk menguji toksisitas ekstrak karang lunak Lobophytun sp sebegai biopotensi antikanker. Metode penelitiaan digunakan yaitu metode Brine Shrime Lethality Tes (BSLT), dengan percobaan laboratorium. Sampel solfcoral diambil dari pulau Bunaken, pengambilan sampel menggunakan metode purposive sampling. Konsentrasi uji menggunakan 10, 100, 500 dan 1000 ppm. Analisis Toksisitas menggunakan analisis probit dengan menggunakan program minitab. Hasil yang diperoleh menunjukan setiap bahwa setiap kenaikan konsentrasi diikuti dengan kenaikan jumlah mortalitas hewan uji. Hasil analisis probit diperoleh nilai LC50 9.98 mg/l. Hasil ini menunjukan bahwa susbtans bioaktif darri softcoral Lobophytum sp berpotensi sebagai senyawa antikanker. Kata kunci. Karang lunak, Lobophytum sp, anti kanker, Artemia salina

Abstract

Green Algae Halimeda sp. Brown algae Padina sp. are two types of marine biota that contain secondary metabolites and are beneficial in the health sector such as anticancer. Bioactive compounds suspected of having anti-cancer activity were tested for activity by means of a toxicity test. The purpose of this study is to test the biotoxicity of Green Algae Halimeda sp and Brown Algae Padina sp to shrimp larvae (Artemia salina Leach). Knowing the toxicity value (LC50) of Green Halimeda sp and Brown algae Padina sp. The research method used is the Brime Shrimp Lethality Test method. Algae samples were taken in the waters of this study Makupa Village. The activity test was carried out in the laboratory of the Biotechnology and Marine Farmasetical, Faculty of Fisheries and Marine Science Sam Ratulangi University. Probit and regression analysis taken to analyte the data. The results of the probit analysis on the biotoxicity of bioactive compounds from Halimeda sp had an LC50 value of 67.87 mg/l and Padina sp 12.49 mg/l. These data indicate that the bioactive compounds from Padina sp are more toxic than Halimeda sp. Bioactive compounds from Padina sp algae can be developed as anticancer raw materials. Keywords: anticancer, Halimeda, Padina, Artemia salina Leach, bioactive compound ABSTRAK Alga Hijau Halimeda sp dan Alga Coklat Padina sp merupakan biota laut yang memiliki metabolit sekunder yang bermanfaat dalam bidang farmasi terutama bahan baku obat anti kanker. Senyawa bioaktif yang diduga memiliki aktivitas anti kanker terlebih dahulu dilakukan pengujian aktivitas dengan cara uji toksisitas. Metode Brine Shrimp Lethality Test. Tujuan penelitian ini yaitu uji aktivitas anti kanker dari ekstrak kasar Halimeda sp dan Padina sp terhadap larva udang Artemia salina L dengan menggunakan metode Metode Brine Shrimp Lethality Test. Sampel alga di ambil di Perairan Desa Makupa. Uji aktivitas dilakukan di Laboratorium Bioteknologi dan Farmasetika Laut Fakultas Perikanan dan Ilmu Kelautan Unsrat. Konsentrasi uji menggunakan 10, 50, 100, 250 dan 500, ppm, dengan terlebih dahulu membuat larutan induk 1000 ppm. Analisis data menggunakan analisis probit untuk menentukan nilai toksisitas LC50. Hasil yang diperoleh menunjukan bahwa kenaikan konsentrasi diikuti dengan kenaikan jumlah mortalitas hewan uji dimana Halimeda sp 56% dan Padina sp 65%. Hasil analisis probit diperoleh nilai LC50 67,87 mg/l untuk Halimeda sp dan Padina sp 12.45 mg/l. Berdasarkan data tersebut senyawa bioaktif dari Padina lebih toksik dibandingkan dengan Halimeda sp, sehingga dapat disimpulkan kandungan senyawa bioaktif dari Padina sp berpotensi untuk dikembangkan menjadi bahan baku obat anti kanker.
