Abstract

Penduduk disekitar Gunung Kelud yang memiliki riwayat diabetes mellitus berisiko menghirup debu vulkanik yang bisa menyebabkan stres oksidatif melalui terbentuknya radikal bebas. Pada penderita diabetes melitus, keadaan hiperglikemia memicu terbentuknya radikal bebas. radikal bebas yang terbentuk karena kondisi hiperglikemia dan pengaruh paparan debu vulkanik ini menyebabkan kerusakan pada membran sel hepar dan meningkatkan kolesterol total darah. Penelitian ini bertujuan untuk membuktikan bahwa pemaparan debu vulkanik Gunung Kelud mempengaruhi peningkatan kadar kolesterol total pada tikus Wistar model diabetes melitus. studi eksperimental menggunakan post-test only control group design dilakukan pada 25 ekor tikus Wistar jantan. Sampel dibagi ke dalam 5 kelompok, yaitu kelompok kontrol negatif, kelompok kontrol positif, kelompok perlakuan 1, kelompok perlakuan 2, dan kelompok perlakuan 3. Variabel yang diukur adalah kadar kolesterol total. Hasil penelitian menunjukkan bahwa kenaikan kadar kolesterol total tikus setelah dipapar debu vulkanik berbeda bermakna (p = 0,049; α = 0,05). Terdapat hubungan yang kuat antara dosis paparan debu vulkanik dengan kadar kolesterol total serum tikus jantan Rattus norvegicus galur Wistar model diabetes mellitus (p = 0,016, r = 0,609). Kesimpulan dari penelitian ini adalah paparan debu vulkanik Gunung Kelud dapat meningkatkan kadar kolesterol total tikus putih model diabetes melitus.

Abstract

Tape merupakan hasil proses fermentasi yang akan menghasilkan alkohol. Pada beberapa daerah, banyak larangan ibu hamil untuk mengkonsumsi tape karena dipercaya dapat menyebabkan keguguran. Keguguran merupakan salah satu dampak dari anemia. Oleh karena itu penelitian ini bertujuan untuk mengetahui pengaruh pemberian air tape ketan putih terhadap kadar hemoglobin pada tikus Rattus novegicus bunting. Penelitian ini menggunakan desain experimental sejati dengan rancangan penelitian Post Test Only Control Group Design. Sampel yang digunakan sebanyak 20 tikus bunting yang dibagi menjadi 4 kelompok; kelompok kontrol (K), kelompok perlakuan dengan 3 dosis air tape ketan putih dengan kadar 2,79% (P1=20 ml/kgBB, P2=30 ml/kgBB, P3=40 ml/kgBB). Air tape ketan putih diberikan hari pertama sampai hari ke-19 kebuntingan. Kemudian pada hari ke-20 tikus dikorbankan dan diambil darahnya dari jantung untuk penghitungan kadar hemoglobin. Hasil pada penelitian ini menunjukkan bahwa kadar hemoglobin setelah perlakuan menunjukkan kecenderungan menurun tetapi tidak berbeda secara signifikan p = 0,1 (p>0,05). Namun berdasarkan hasil uji d-type effect size pada P3 menunjukkan pengaruh yang sangat besar dengan ES = 1.40540541 dalam menurunkan kadar hemoglobin. Hasil uji korelasi pearson p= 0,017 (p<0,05) menunjukkan ada hubungan yang cukup kuat dan berlawanan arah p=– 0,529 (p>0,5). Kadar hemoglobin setelah perlakuan menunjukkan kecenderungan menurun tetapi tidak berbeda secara signifikan. Kata kunci: hamil,air tape ketan putih, kadar hemoglobin

Abstract

This study aimed to evaluate the effects of ciplukan (Physalis angulata L.) leaf ethanol extract on L-NG-nitroarginine methyl ester (L-NAME)-induced hypertensive rats. We randomly divided twenty-five Wistar rats into five groups. The sham group was given a PBS injection. The hypertensive group was injected with L-NAME on days 1 to 28. Three groups of hypertensive rats were given the extract on days 4 to 28. Blood pressure was measured using the tail-cuff method on days 0, 4, 10, and 27. The endothelial progenitor cells (EPCs) in the blood were measured by flow cytometry as a percentage of circulating angiogenic cells (CACs, CD34+/CD309+/CD45+) and endothelial colony-forming cells (ECFCs, CD34+/CD309+/CD45-). Serum NO and MDA levels, as well as serum SOD activity, were measured colorimetrically. Serum TNF-α levels were measured by the ELISA method. The ciplukan leaf extract reduced systolic and diastolic blood pressure, reduced the percentage of EPCs in the blood, increased serum NO levels, reduced MDA levels, increased serum SOD activity, and reduced serum TNF-α levels in L-NAME-induced hypertensive rats. It is concluded that ciplukan ethanol leaf extract exerts protective effects on L-NAME-induced hypertensive rats. These study results can strengthen the scientific basis of using ciplukan leaf ethanol extract to treat hypertension.

Abstract

Background : Gestational Diabetes Mellitus is a complication of pregnancy in mothers without a previous diagnosis of diabetes, then chronic hyperglycemia is detected during pregnancy. Generally, this hyperglycemia is caused by impaired glucose tolerance due to pancreatic cell dysfunction, which begins with chronic insulin resistance conditions. The prevalence of GDM in Southeast Asia is 10%, while the overall diagnosis of diabetes in women in Indonesia is 1.8%. Nowadays, antioxidant therapy is being developed to treat DMG, one of which is the use of purple cassava plants.Objective : to determine the effect of purple cassava juice (Ipomea Batatas L.) on increasing insulin levels of wistar rats model of gestational diabetes mellitus. Methode : This study used an experimental method which was designed with the Post Test Only Group Design method. The total number of samples used in this study was 30 rats which were divided into 5 groups. Furthermore, the treatment group was given STZ injection of 45 mg/KgBB and then given purple cassava juice therapy for 14 days. Next, blood samples were tested for insulin levels using the ELISA Rat Kit. Result : There are no differences of insulin level between the treatment group and the control group because they had constant result. Conclusion : In this study, it was not found that there was an increase in insulin levels after treatment of purple yam juice for 14 days. So the research hypothesis was rejected.
