Abstract

Introduction: This study was aimed to determine and formulate antibacterial activity of Zingiber Officinale Roscoe (Z. officinale) extract against Enterococcus faecalis (E. faecalis) as a potential irrigating solution which is easily available and cost-effective. Methods:  Extract of Z. officinale Roscoe was yielded through soxhlet technique. Samples divided into two groups, i.e., the extract of Z. officinale Roscoe and chlorhexidine 2% as a control. The antimicrobial activity was observed using the diffuse agar method. Minimum inhibitory concentration (MIC) was determined by serial microdilution method, and minimum bactericidal concentration (MBC) was determined by cultured samples in blood agar. Data were analysed using ANOVA and Kruskal-Wallis test. Results: Extract of Zingiber officinale Roscoe has antimicrobial activity against E. faecalis with the MBC of 15.625 mg/mL. (p<0,05). Conclusion: Extract of Zingiber officinale Roscoe have shown antibacterial activity against E. faecalis and can be further developed as a potential root canal irrigation solution.

Abstract

Docosahexaenoic acid (DHA) is one of essential fatty acids that are beneficial to health. Nowadays, the source of docosahexaenoic acid (DHA) is mainly obtained from fish which are extracted into fish oil products. However, the fish oil products still have some drawbacks in term of purity, acceptable flavor for costumers, and also their not environmental friendly production process. As an alternative solution, heterotrophic microalgae can be used as a potential source for DHA due to their excellence compared to fish oil products. The aim of this study is to isolate the heterotropic microalgae that can produce DHA. The heterotrophic microalgae were isolated from mangrove fallen leaves (Rhizophora apiculata) by using direct planting method. The morphology of pure microalgae colony were observed through light microscope and subsequently fermented for 14 days. Fatty acids were extracted and methylated through direct transesterification method. Identification and quantification of DHA were conducted by using gas chromatography. The results were four isolates of heterotropic microalgae, namely MTKC1, MTKC2, MTKC3, and MTKC4. The extract of MTKC2 that only showed the content of DHA with value of 9.2 % w/w. Therefore MTKC2 is a potential source for DHA. The MTKC2 was further identified by using molecular biology method and confirmed as Thraustochytrium aureum.

Abstract

Marine microorganisms, especially actinomycetes bacteria and fungi produce a variety of secondary metabolites that biologically active and have a unique structure. This study aims to isolate some strains of marine fungi that grow on marine algae as a potential source of antimicrobial agent. Marine fungal strains isolated from the marine algae with antimicrobial activity screening is done by using the disc diffusion method. In this study, it was obtained 18 strains of fungal isolates. The screening results that give best antimicrobial activity shown by the activity of the extract liquid culture of fungal isolate strain AKT.C6 are 10.0 mm against the bacteria Escherichia coli and 10.8 mm against Bacillus subtilis. Mycelium extract of fungal isolates strain AKT.C4 is 16.9 mm against the bacteria Escherichia coli and 17.6 mm against Bacillus subtilis, while the liquid culture extract and mycelium extract did not show any activity against Candida albicans. The isolation of fungi from Kappaphycus alvarezii algaee has potential as an antibacterial agent.

Abstract

Telah dilakukan penelitian pembentukan zat warna Monascus purpureus hasil fermentasi padat dengan dua variasi sampel limbah ampas kelapa sebagai substrat. Dilakukan pengukuran pembentukan zat warna dengan cara mengekstraksi sampel menggunakan etanol 95% pada fermentasi hari ke-7 dan ke-14 dilanjutkan dengan pengujian kromatografi lapis tipis menggunakan pengembang etanol:etilasetat (7:3). Serapan zat warna diukur dengan spektrofotometer UV-Vis pada panjang gelombang 400, 406, 498, 500, 511 dan 512 nm. Hasil menunjukkan serapan zat warna mengalami peningkatan dengan rentang serapan antara 0,128–0,269 dan 0,212-1,019 dan Sampel A menunjukkan serapan lebih tinggi dibandingkan Sampel B. Pengujian aktivitas antimikroba ekstrak zat warna ke dua sampel dilakukan terhadap Escherichia coli dan Candida albicans menunjukkan adanya aktivitas antimikroba. Kesetaraan aktivitas berdasarkan kurva baku Tetrasiklin HCl terhadap Escherichia coli dan kurva baku Nistatin terhadap Candida albicans menunjukkan ekstrak zat warna Monascus purpureus dengan konsentrasi 100% pada Sampel A lebih besar dibandingkan Sampel B. Dengan dua variasi waktu yang berbeda hari ke-7 dan hari ke-14 Sampel A mempunyai aktivitas kesetaraan konsentrasi 1,41% dan 4,13% Tetrasiklin HCl serta 0,81% dan 1,38% Nistatin. Sampel B pada hari ke-7 dan hari ke-14 mempunyai aktivitas kesetaraan konsentrasi 1,02% dan 2,44% Tetrasiklin HCl serta 0,79% dan 1,20% Nistatin.
