Abstract

Stevia leaves are harvested because of their sweetness. This special characteristic is contributed by steviosida, rebaudiosida and dulkosida. Steviosida and rebaudiosida are low calorie, not able to be fermented by bacteria in the mouth and non carcinogenic. These superiorities make steviosida able to act as sugar subtitute, especially for diabetics; obese and as sweetener for toothpaste. The aim of this research was to find planting medium of Stevia leaf explants which is capable of producing the highest level of stevioside. The explants were grown in Surakarta. The medium was New Phaleonopsis (NP) within the addition of growth regulator substance, the combination of 2,4 Dichlorophenoxyacetic Acid (2.4-D), and Furfuril Amino Purin (kinetin) each by kinetin 1 ppm; 2,4-D 0.25 ppm and kinetin 0,75 ppm; 2,4-D 0,5 ppm as well as kinetin 0,5 ppm; 2,4-D 0,75 ppm and kinetin 0,25 ppm 2,4-D 1 ppm. Callus levels were macerated by n-butanol, then were analyzed by TLC densitometry. Stevioside levels in callus were compared with the steviosid levels in the original plants. The highest stevioside level was found in the callus grown in NP medium by adding 1 ppm kinetin, amounting to 10,625 ppm. The steviosid level was higher than the original plants. Stevioside level in Stevia leaf was 0,65 ppm.

Abstract

Seligi (Phyllanthus buxifolius (BI.) M.A.) leaves contain flavonoid, polifenol, and saponin that have antioxidant activity and thought to potentially as antihiperlipidemic. This research was aimed to find out the effect of seligi leaves ethanol extract to increase HDL level and decrease LDL level in white rats. The subject of the research was 30 male white rats in 150-200 gram body weight and 2-3 months age. Rats were divided into 6 groups randomly then were given BR II and water everyday. Group I, II, and III were normal control group, the positive control (simvastatin), and negative control respectively. The treatment group was given seligi leaves extract in doses of 75; 150; and 300 mg/kg bw/day. The tested animal were given cow fat and yolk for two weeks up to hypercholesterolemia state, tested preparation was given on day-14 until the day-28. The HDL and LDL levels were measured on the day-0, 14, and 28 by CHOD-PAP method. The result of researchshowed that seligi leaves ethanol extract orally administration could increase the HDL and decrease the LDL levels of blood serum. Seligi leaf ethanol extract with dose 300 mg/kg bw/day had equal effectiveness to simvastatin.

Abstract

Hypertension is a major health problem most frequently found in Indonesia. This disease can affect anyone from various socio-economic groups. This study aimed to determine the drug utilization in terms of the drug classes, dosage form, drug administration route (pattern of antihypertensive drugs usage) in geriatric patients and its compliance with the Medical Care Standards (MCS) and the Hospital Formulary at Karanganyar Hospital in 2011 using the ATC / DDD methods, and determine antihypertensive drugs which include in DU90% segment. This study used secondary data which were taken from the patient's Medical Record. This research was quantitative descriptive research, the data were collected retrospectively. Data of antihypertensive utilization and inpatient visit obtained from Medical Record Installation. Entire data then were processed to determine the quantity of antihypertensive utilization in DDD/100 unit of inpatient day and Drug Utilization profile (DU90%). The results showed that the antihypertensive drugs utilization in geriatric patients (60 years and above) in Karanganyar Hospital during 2011 were captopril (60.69%), furosemide (11.26%), amlodipine (8.17%), nifedipine (9.45%), hirdroklorotiazid (5.94%), lisinopril (3.44%), bisoprolol (1.05%), and from the DU90% table, drugs which include in the DU90% segment were captopril, furosemide, and amlodipine. The compliance to Hospital Formulary in 2011 was 80%.

Abstract

Solanum torvum Swartz (Solanaceae), known as takokak is a medicinal plant which has phagocytosis activity. Takokak plant usage with intent to suppress or reduce viral and bacterial intracellular infections, to improve immunodeficiency condition or as growth stimulant of body's immune cells in the immune system. This study was aimed to determine ability of takokak extract to increase the phagocytosis activity. This study used eight groups of white male mice each group six mice. Mice were given the test preparation once daily for seven days sequently. Group I: negative control 1% CMC-Na; group II: positive control dexamethasone 2 mg/kg bw mice; group III until VIII were given takokak ethanol extract dose of 37.5 mg/kg bw, 75 mg/kg bw, 150 mg/kg bw, 400 mg/kg bw, 500 mg/kg bw, 600 mg/kg bw respectively. Phagocytosis activity test data were analyzed using one way ANOVA test. The results showed that ethanol extract of takokak could increasing the phagocytosis activity and capacity of macrophages in Balb/c mice induced carbon clearance with carbon dose of 5 mg/kg bw.

Abstract

One cause of tinea versicolor is a fungal Pityrosporum ovale which colonizes on the skin. Previous research has examined the red betel leaves, beluntas leaves and cabbage leaves as antifungal against Pityrosporum ovale. This study aims to determine the effectiveness of the above herbs in the form of single or various combinations to inhibit the growth of the fungus Pityrosporum ovale by in vitro method. The extraction method was performed by infudation on the red betel leaf powder, beluntas leaves and cabbage leaves. The results of infudation are then made in a combination of (1:1), (1:3) and (3:1). The antifungal test with dilution method was then performed on the single and the combination extract. Dilution method was performed by making a series of dilutions in concentrations of 50%, 25%, 12.5%, 6.25%, 3.13%, 1.56%, 0.78%, 0.39%, 0.19%, 0.09%. The results showed that the MKC (minimum killing concentration) of red betel leaves, beluntas leaves and cabbage leaves, combination of red betel leaves and beluntas leaves (1:1), (1:3) and (3:1), combination of red betel leaves and cabbage leaves (1:1), (1:3) and (3:1) were 50%, 25%, 25%, 25%, 12.5%, 50%, 50%, 12.5%, 25% respectively. The best combination were red betel leaves and beluntas leaves infusions with a ratio of (1:3) and the combination red betel leaves and cabbage leaves infusions with a ratio of (1:3).

Abstract

White guava (Psidium guajava, L.) fruit extract has antioxidant activity, because it contains quercetin, a flavonoid compound. Lotion is cosmetic preparations that widely used. This research was aimed to find out the antioxidant activity of lotion of white guava fruit extract. The white guava extract was made lotion preparation in three different concentrations of 18,5%; 20,5%, dan 22,5%. The three formulas were tested for its dispersive, adhesion, and viscosity. Determination of antioxidant activity in lotion was conducted by DPPH (1,1-diphenyl-2 pikrilhidrazil) using rutin as control and the results were calculated by IC50 value by probit analysis. The results showed that guava fruit extract lotion had antioxidant activity. Formula with the highest antioxidant activity was formula contained white guava fruit extract concentration of 22.5% with IC50 value of 131.83 ppm.

Abstract

Actinomycetes is one type of microorganism that has been widely used for the production of antibiotics as well as a biological control for pests and diseases in plants. In general, many Actinomycetes distributed in soil, water, tissues endophyte, and other natural environments. The study aimed to determine the distribution and diversity of Actinomycetes found in the root zone at Solanaceae plants and to know the isolates of Actinomycetes isolated from the root zone of plants Soalaneceae could inhibiting the growth of bacteria. Actinomycetes isolated from seven soil samples of Solanaceae plants rhizosphere in Selo Boyolali, Colomadu Karanganyar, Gondangrejo Karanganyar, and Wonogiri Jatipurno. The isolates of Actinomycetes tested the antibacterial activity by well diffusion method on media Glycerol-Yeast Extract against four species of pathogenic bacteria Ralstonia solanacearum, Staphylococcus aureus, Bacillus subtilis and Escherichia coli. Results showed that there were 54 isolates of Actinomycetes isolated from the soil samples of Solanaceae plants rhizosphere. Isolates could inhibit the growth of R. solanacearum (29%), S. aureus (24%), B. subtillis (14%), and E. coli (7%). Isolates had the ability to degrade carbohydrate and protein.

Abstract

Pegagan (Centella asiatica, [L.] Urban) gives positive effect on the cerebral nerve stimulation, and to smooth the blood transportation to the cerebral vessels. Wild ginger (Curcuma xanthorrhiza, Roxb) potentially protects the cell from oxidative damage and increases appetite. This study was aimed to find out effect of the combination of pegagan herb and wild ginger rhizome extracts in instant preparation to increase the memory. The step through passive avoidance method was used by analysis pretest and posttest latency time interval parameter from the length of time the mice needed to enter the dark compartment from bright compartment. The dose of 0.195 mg Ginkgo biloba as positive control, and 5.2 mg aerosol as negative control, combination of dry extract preparation at 2.6 mg (0.91 mg pegagan extract and 0.98 wild ginger extract), 5.2 mg (1.82 mg pegagan extract and 0.98 wild ginger extract), and 7.8 mg (2.73 mg pegagan extract and 2.94 wild ginger extract) doses. The data was analyzed using two-way ANOVA, followed by Tamhan’s T2 test. The result showed that the combination of macerated pegagan herb and wild ginger rhizome in dry extract preparation can improce memory of the white mice.

Abstract

Leaves of Mirabilis jalapa L. can inhibit the growth of Staphylococcus aureus and Aspergillus sp. The study aimed to determine the antifungal activity of n-hexane, ethyl acetate, and water fractions from the methanolic extract of leaves of Mirabilis jalapa L. against Pityrosporum ovale fungi and to determine the most effective fraction in inhibiting the growth of Pityrosporum ovale. Antifungal activity assays using the diffusion method, in which the test solution of Mirabilis jalapa L. leaves made serial consentrations of 10%, 5%, 3% entered in wells that have been applied to SGA media fungal suspension test. The results was analyzed using two ways statistical analysis. The result showed that of n-hexane, ethyl acetate, and water fractions from methanolic extracts Mirabilis jalapa L. leaves have antifungal activity against Phytyrosporum ovale. Ethyl acetate fraction at a concentration of 10% had the highest antifungal activity was 25 mm.

Abstract

Buccal patch is a preparation used to put on the buccal mucosa so that issues such as first-pass metabolism, avoid pre-elimination systemic GI tract can be overcome. Isosorbide dinitrate is nitrate which used for anti angina, inside of administration through the oral only have bioavailability 29% and overhauled in the liver, time half-life 45-60 minutes. This study used isosorbide dinitrate in buccal patch with various concentrations of CMC-Na and PVP-K29 as a matrix, to determine the physical properties and release kinetics of isosorbide dinitrate. Physical properties test were weight uniformity, folding endurance, surface pH, swelling index and uniformity of content. In vitro release using USP Type II method of rotating paddle speed of 50 rpm with the dissolution medium aqua bidestilata 500 ml. High performance liquid chromatography used UV detector 220 nm, mobile phase methanol pro HPLC and aqua (50:50). The results showed that the ratio of 30:70 showed the results of swelling index minutes of 180 326.268%, dissolution percent minutes 480 90.625% follow the kinetics of zero order by diffusion.
