Abstract

A multiple genes family claudin encodes proteins which forms main structures of tight junction between epithelial cells. In intestinal epithelium claudin proteins are proposed to have roles in ion balances regulation and forming barrier against pathogenic invasion. In the present work, claudin genes were molecularly identified in common carp (Cyprinus carpio), one of the most important cultured fish in freshwater aquaculture. Carp claudin 1 and 2 were amplified with primers designed based on relevant known claudin genes in fish. On the basis of carp ESTs, other six carp claudin genes were amplified. The amplification products were cloned and sequenced. Phylogenetic and sequence analysis confirmed that the eight identified genes are claudin genes and could be designated as carp claudin 1, 2, 3b, 3c, 7, 11, 23, and 30, respectively. This finding could be used to develop further study of carp claudin especially roles of claudin during pathogen infection and strategy to modulate its expression in order to protect diseases in carp aquaculture.

Abstract

Aquaculture is growing rapidly in Southeast Asia (SEA) and five of the top ten aquaculture producers in the world are countries in SEA. The need to manage aquaculture responsibly is critical. Calculation of aquaculture carrying capacity (ACC) for shared water bodies is necessary for proper management. I review my observations of the needs and capabilities of SEA governments for ACC and then describe a new project in which teams of U.S., U.K. and Indonesian scientists will work on ACC for three Indonesian water bodies – Lake Cirata, Lake Toba and Lake Jatigede.

Abstract

The efficacy of organic zinc amino acid complex(AvailaZn®) on growth performance and immunity of Pangasius catfish (Pangasianodon hypophthalmus) was assigned in CRD with  4 treatments and 5 replicates. The treatments  were control with inorganic zinc 100 ppm (T1) and the other two treatments of zinc 100 ppm at different proportion of inorganic zinc : organic zinc  70:30 (T2) and 40:60(T3) and the treatment of only organic zinc 50 ppm (T4; 50% of control). Pangasius catfish with average weight of  209  g. were stocked in 1,000 liter fiber tanks at the density of 15 fishes per tank and fed treatment diets 3% of  body  weight  twice daily for three months. The results indicated that there were statistical difference (p<0.05) on growth performance in term of body weigh and specific growth rate of  Pangasius catfish fed diet supplemental organic zinc at different levels were higher (P<0.05) than group of inorganic control(T1). The specific growth rate were 0.89, 1.20, 1.16 and 1.27 %/ in T1, T2, T3 and T4, respectively. Feed  conversion ratio were not significantly different(p=0.08) and protein efficiency ratio also were not significantly different(p=0.09). White blood cell count, serum protein, immunoglobulin (IgM), hemoglobin, and super oxide anion in group of zinc amino acid complex were better (p<0.05) than group of inorganic control(T1).  Therefore, using zinc amino acid complex(AvailaZn®) in Pangasius catfish diet can enhance catfish growth performance (p<0.05) and decrease zinc level in the catfish diet by 50% of using inorganic zinc. Supplementation zinc amino acid complex(AvailaZn®) in Pangasius catfish diet is recommended at the level of 50 ppm for organic mineral and 100 ppm for the proportion of organic and inorganic 70:30 ppm.

Abstract

This research studied the effectiveness of Ascidian Didemnum molle extracts as curative for bacteria Vibrio harveyi infection in tiger shrimp (Penaeus monodon). The purpose of the research was to find out the effective concentration of antibacterial compound Ascidians Didemnum molle to curative the infection of bacteria Vibrio harveyi in tiger shrimp. The Ascidians Didemnum molle which were used in this research was originated from Kepulauan Seribu waters. Ascidians Didemnum molle collected by 3.5 kg with an average size of 5-15 cm per individual. The research was conducted from September to December 2013 at the Biotechnology Laboratory and Aquaculture Laboratory of the Faculty of Fisheries and Marine Sciences, Padjadjaran University. The method which was used in this research was a laboratory experimental method such as phytochemical test, extraction, anti-Vibrio harveyi activity test, LC50 test and chalenge test. Descriptive analysis was used in this research. Based on the anti-Vibrio harveyi activity test, it could be concluded that Ascidian Didemnum molle extract had potential as an antibacterial for Vibrio harveyi, which indicated by inhibition zones as 11.1 mm at a concentration of 1,000 mg/L. The result of EPA Probit analysis on LC50 test, with 48-hours of observation, determine 387 mg/L as a safe extract concentration for tiger shrimp. The chalenge test showed that treatment D with concentration of 290.25 mg/L was effective to be used as treatment for tiger shrimp that were infected by Vibrio harveyi bacteria solution of density 106 CFU/ mL, with the highest survival rate was 51.7%.

Abstract

Probiotics is a bacterial product that has beneficial for fish; both for growth and body's defenses from bacteria desease. The aim of the study is to determine the types of candidate of probiotic bacteria from the digestive tract of giant prawns that is found in Teratak Buluh Pekanbaru, Riau. This research was conducted at the Laboratory of Microbiology and Biotechnology of Fishery Faculty of Fisheries and Marine Sciences University of Riau, Pekanbaru to get a candidate of Probiotic Bacteria and do molecular examination that consists of purification of electrophoresis gel and DNA sequencing at the Institute for Biotechnology Serpong, Tangerang Banten. The method of this study is a survey method that used physical and biochemical test and molecular identification . The results showed that eleven isolates of bacterial are the species Bacillus BDU4 strain from A Bacteria isolates with 99 %  homology and 10 isolates of  bacterial are bacillus genus because the degree of homology below 97% and they are considered as the original indonesia’s new species probiotic bacteria.

Abstract

Hematological and biochemistry parameters are essential for the evaluation of fish health status; it helps provide essential information to diagnose fish diseases. This experiment was conducted to investigate the affect of Streptococcus agalactiae infection on hematological and biochemical parameters in red hybrid tilapia (Oreochromis sp.). The fish were inoculated intraperitoneally with 1 x104 cfu/mL bacteria then blood samples were collected and tested during the experiment. After seven days The red blood cell (RBC) counts decreased from 2.31 x 106to 1.68 x 106. The hemoglobin (Hb) and hematocrit (PCV) counts decreased to 7.17g/dl and 19.2% in the infected fish compared to the control group (9.71g/dl and 28.20%). In contrast, the white blood cell (WBC) counts increased from 4.20x103 in the control fish to 13.2x103. The biochemical results initially showed an increase in glucose levels of the infected fish compared to the control from 53.82 to 101.75 on the 72nd hour, respectively.  There were no significant differences in the levels of serum total protein (T. prot), albumin (ALB), and aspartate transminase (AST) between the infected and the control fish.The inoculated fish displayed marked clinical signs such as erratic swimming behavior, exophthalmia and lethargy during the experimental. The present study has established that the changes in hematological, biochemical and clinical signs of fish can be used as an early diagnosis of pathological and physiological status in red tilapia culture

Abstract

As the global scenario is now changing towards the development of sustainable and biosecure aquaculture by using of safe and eco-friendly products. This paper reviews the application of Moringa plant for development of sustainable aquaculture. The Moringa plant shows many applications in aquaculture such as antimicrobial activity, antibacterial efficacy, larvicidal activity, piscicide, anti-cyanobacterial activity, antifertility effect, food supplement and seafood preservative. The use of Moringa as antimicrobial activity of the seed coat and pod husks; as a medicinal plant for protection of Penaeus indicus from pathogenic Vibrio harveyi; as an evidence for cyanobactericidal activity of filtrate from crushed Moringa seeds and isolation of a protease inhibitor. In conclusion, these results suggest that Moringa oleifera showed potentially valuable plant for development of sustainable aquaculture
