Abstract

Introduction: One strategy for HIV-1 (retroviridae) infection therapy is using NNRTI (non-nucleoside reverse transcriptase inhibitor) to suppress viral replication by binding to the reverse transcriptase (RT-ase) enzyme. One drug in this class is Efavirenz (EFV). Based on the good effectivity of EFV in HIV-1, it is suspected that EFV also has activity against HIV-2 and HTLV. This study aims to determine the activity of EFV in the human-infecting retroviruses such as HIV-2, HTLV-1, and HTLV-2. Methods: This study uses Bioinformatics and Medicinal Chemistry tools such as, phylogenetic tree construction, pairwise sequence alignment, looking for RT-ase motifs and domains, predicting secondary structures, modelling 3D RT-ase structures, docking, and predicting hydrophobicity with Kyte-Doolittle Hydropathy Plot. Result: There is a common ancestor of the four viruses according to the constructed phylogenetic tree. There are also RT_like Superfamily domains at HIV-2 and HTLV-1. The similarity and identity value, and results of HTLV-1 and HTLV-2 RT-ase structural modeling do not give good results. Binding energy value of EFV to HIV-2 RT-ase is higher than HIV-1 RT-ase. Conclusion: EFV is not effective against HIV-2, HTLV-1 and HTLV-2

Abstract

Sebagai upaya untuk mewujudkan salah satu tujuan SDGs 2030 yang berhubungan dengan ekosistem lautan dan sebagai upaya untuk mengurangi segala jenis polusi kelautan, terutama dari aktivitas daratan, penulis menggunakan kemajuan Ilmu Pengetahuan dan Teknologi (IPTEK) untuk mengembangkan sedotan yang berbahan dasar gelatin. “Tari Latin untuk Mantanmu” dibuat menggunakan mesin pencetak sedotan dengan menggantikan bahan dasar yang tadinya plastik dengan campuran gelatin dan air seperti yang tertera pada formulasi di atas. Setelah dicetak, sedotan gelatin dipisahkan dan dibiarkan pada suhu ruangan untuk mempercepat pengeringan dan mencegah agar sedotan tersebut tidak menempel satu sama lain. Selanjutnya, sedotan gelatin dikemas dalam plastik biodegradable dan siap dipasarkan ke masyarakat. “Tari Latin untuk Mantanmu” merupakan inovasi paling tepat dan efektif untuk menghadapi permasalahan – permasalahan yang terjadi di Indonesia saat ini, yang terkait dengan penumpukan sampah sedotan plastik. Selain itu, “Tari Latin untuk Mantanmu” dapat mencegah timbulnya penyakit yang disebabkan oleh sedotan plastik. Dengan diwujudkannya inovasi ini, diharapkan dapat membantu Indonesia bebas dari sampah sedotan plastik dan menjaga keseimbangan ekosistem lautan.

Abstract

Infection is the highest prevalence of disease in Indonesia. Efforts to increase the prevalence of infection is to improve the community's immune system. Kasumba turate is an endemic plant of South Sulawesi empirically used in the cure of smallpox and improve the nature of the body due to the nature of its imunomodulator. Traditional decoction of the Kasumba turate is less effective for consuming because it has a bitter taste, so the aim of this study is to modified decocta of kasumba turate becomes effervescent granules which is contain antioxidant to reducing free radicals in the body without reduce its usefulness. The method of making effervescent granule supplementation is by using infusa (decoction) from a lyophilized kasumba flower with Freeze Dryer and formulated with a combination of citric acid and sodium bicarbonate 1: 1.5. Effervescent granule preparationss were evaluated by LOD, MC, compressibility, species weight and pH test to obtain a qualified preparation and antioxidant analysis by DPPH method. Based on the evaluation of the preparation, it is found that the effervescent kasumba turate granule fulfill the applicable requirements: acid granules obtained% MC and% LOD 0.5% and 0.5%, basic granule% MC and% LOD 0.7%, 0.77%, flow properties break 27o, and flow rate 9.7 g/s),% compressibility 16% and weight type 1.66, pH of the preparation 7. The effervescent lyophilisate preparation of kasumba turate becomes an effective supplement in enhancing the immune system and as an antioxidant for all age.

Abstract

Introduction: Diphtheria is an upper respiratory tract infection caused by Corynebacterium diphtheria bacterium. The recent diphtheria outbreak caused a large number of deaths in rapid period. This is due to the absence of a fast and precise method of diphtheria diagnosis. Current diagnosis practices aredone by cultivating bacteria which takes a relatively long time, which is 3 to 5 days, while the patients may already develop severe infection by the time the results are accessible. Therefore, a sensitive, easy and fast diphtheria diagnostic method is needed so that patients suspected of being infected can betreated quicker.Methods: This advertorial is written based on literature study method.Result: The diphtheria diagnostic kit was designed based on the DNA hybridization method by using DNA microarray. There are probes on the microarray plate in the form of a single stranded DNA fragment that is complement to the unique area of gene that encodes diphtheria toxin. The test sample wasobtained from the preparation of the patient's throat smear and labeled with a fluorescence marker before dropped onto the plate. Thus, if there is diphtheria DNA in the patient's sample, hybridization will occur between diphtheria DNA and the complementary DNA probe. The occurrence of hybridization will be characterized by fluorescence as an indicator of whether patients have diphtheria or not. The application of this diagnostic kit can shorten the diagnosis of C. diphtheria to less than 24 hours .Conclusion: Diphtheria diagnostic kit based on DNA hybridization can be made and applied.
