Abstract

Differential display technique was used to identify differentially expressed gene ofswordtail fish (Xiphophorus spp.) that induced by 1 µg/ml copper for 24 hours. From eightprimers that have been used for differential display, seven of them showed the differentialgenes which were not present in the control. The reamplification of these fragments rangingfrom below 100 to 200 bp. But only two fragments were successfully cloned and sequenced,those are H-AP10 and H-AP11. Sequence analysis showed that both of the sequences revealedhigh similarity with Homo sapiens stress-associated endoplasmic reticulum protein, a signalgenerated that induces apoptosis.Keywords: differential display, swordtail fish, copper pollution, mRNA expression, biomarker

Abstract

The experiment was conducted to study the effect of ethylene glycol (EG) on disturbance of spermatogenesis process of the male mice. Sixteen mice at the age of 2 months were randomly into 4 groups of 4 each. The mice of control group were only given drinking water without EG. Group KPI, KP2, and KP3 were given EG 5, 10, and 15 ml/liter drinking water, respectively. On day 31 of post treatment, all mice were killed for their testis. The testis were collected and processed for microscopic examination and haematoxylin-eosin staining. The result showed that EG given orally for 30 days on male mice have  negative effect on the process of spermatogenesis as the numbers of spermatogonia, spermatocyte and Sertoli cells decreased significantly (P0.01)

Abstract

Vibriosis was a crucial problem and a primer infection causing mortality of humpback grouper.Virulence factor of vibrio, such as vili, is an adhesion molecule protein based on haemaglutination test toblood serum of humpback grouper. This research was aimed to characterize an adhesion molecule of viliprotein of V. alginolyticus on epithelial cells of Humpback grouper as haemaglutinin protein. Themethods are laboratory exploration by isolate epithelial cells of Humpback grouper, characterize a receptorand test vili protein to adhesion molecule that is epithelial cell of Humpback grouper. The result of proteinhaemaglutinin test to vili V. alginolyticus show that an adhesion molecule with a molecule weight of38.98 kDa is an adhesin vili V. alginolyticus and a haemaglutinin. The adhesion pattern of V.alginolyticus on epithelial cells of Humpback grouper exposed to protein haemaglutinin of vili show bothdiffuse and localize pattern. Research conclusion was the haemaglutinin protein of vili V. alginolyticuswas an adhesin having a role to both virulence process of bacteria and attachment process to receptor ofepithelial cells of Humpback grouper.Keywords: adhesion molecule, epithelial cell, vili, V. alginolyticus, C. altivelis

Abstract

This research was conducted to find out the mineral level status of Cu, Mg, and Fe in blood serum of local goats which are reared with different methods. Blood serum was taken from 15 male and 15 female goats, 15-18 month old. The goats were divided into three groups which consist of 5 males and 5 females. Three methods of caging in which most commonly conducted by local people were selected as treatments, i.e. 1) Goats are caged only at night, while in daytime (around 10.00-12.00 WIB) they are discharged freely therefore they could foraging; 2) Goat are caged at night, while in daytime (around 10.00-18.00 WIB) they are bound with rope at which they could foraging around and, the rope would be removed several times in that particular daytime; 3) Goats are released freely all day long, but they are provided with kind of place as a cage, under the house or under the tree. Blood serum was taken from every unit sample. This research revealed that there was no significant different among methods of caging on the level rates of Cu, Mg, and Fe in local goat serum. The level rates of Cu, Mg, and Fe in goats serum that was found from the research were in normal range.

Abstract

The present study was aimed to quantify the dynamics of early antigen specific proliferative responses of lymphocytes to (protein) antigens associated with experimentalMycobacterium avium subsp. paratuberculosis (Mp) infection cattle. The data were collected from20 experimentally infected calves, and 10 uninfected control animals, during the first 2 years oftheir lives. Several purified protein derivative antigens (Ppdp, Ppda, and Ppdb), tworecombinant Mp heatshock proteins (Hsp65 and Hsp70) and whole bacteria (sonicated Mpstrain 316F) were used to measure lymphocyte proliferation in a lymphocyte proliferationassay. Data were analyzed using a linear mixed effect (LME) model. The results showedsignificant group and timed effects for all antigens tested. At several time points, the responsesin the infected group were found significantly higher as compared to control group. The Ppdantigens induced similar lymphocyte proliferation patterns, as compared to whole bacteriaantigen and Hsp70. These results indicated that the antigen specific proliferative responses oflymphocytes differs for different antigens, probably related to differences in their availabilityduring different stages of infection. The application of LME model is a useful tool for analyzingthe quantitative longitudinal datasets. Keywords: dynamics, Mp, antigen, LME

Abstract

The objective of this research is to see the influence of weather on the prevalence of Newcastle Disease (ND) in chicken in Kabupaten Aceh Utara (North Aceh). Data used in this research were obtained from Dinas Peternakan North Aceh for the number of chicken suffered ND and from Badan Meteorologi dan Geofisika Lhokseumawe, North Aceh for the form of weather. Multiple Linear Regression Model with five independent variables (the average of rainfall per month, the average of maximum temperature, the average of minimum temperature, the velocity of the wind, and the average of humidity per month) was used to see the influence of wheather to the prevalence of Newcastle Disease. Proportion the number of chicken suffered from ND which is the ratio of the number of chicken suffered from ND to the total number of chicken was used as dependent variables. The result shows that the best model is Ŷ= 120.529278 – 1.33 x wind humidity + 1.907 x wind velocity.

Abstract

This research was aimed to find out a substance that was useful in early pregnancy diagnosis of Friesian Holstein cows. Pregnancy Specific Protein B (PSPB) was isolated, purified and partially characterized from cotyledon Friesian Holstein cows. Characterization of PSPB protein was conducted using SDS-PAGE and Western Blot. Antisera were developed against PSPB and by immunohistochemical techniques the protein was localized to the binucleated cells of the cotyledon. The estimated molecular size of Friesian Holstein cows PSPB was 59.88 kDa with the concentration of PSPB protein in cotyledon was 6480 ng/ml. PSPB protein could induce anti-PSPB antibody with the value of optical density (OD) were 0.179±0.0102 (before immunization); 1.466±0.3288 (3rd week after immunization); 1.936±0.4754 (1st week after booster;) and 2.256±0.4842 (2nd week after booster).
