Abstract

This study aimed to compare the effects of simvastatin, rosuvastatin, and fenofibrate therapies on the body weight and lipid profiles of the female rats receiving a combination of oral contraceptives (COC) and a high-fat diet (HFD). Twenty female Wistar rats (130-200 g) were divided equally into four groups. The rats received a standard diet for seven days, and their baseline lipid profiles were analyzed. All rats also received COC containing 15.1 µg/kg levonorgestrel/3.1 µg/kg estradiol with HFD for 60 days. Starting on day 31, the rats were given the respected treatment. The control group received the vehicle, whereas the others were treated with either simvastatin (2.1 mg/kg), rosuvastatin (0.5 mg/kg), or fenofibrate (8.2 mg/kg). Blood samples were taken on days 30 and 60. The results showed that the administration of COC+HFD and vehicle for 30 days increased the rats’ body weight and dyslipidemia, characterized by a significant decrease in Low-Density Lipoprotein (HDL) and an increase in total cholesterol (TC) and triglycerides (TG) levels (P0.05). Among the treatments, only fenofibrate was found to be able to prevent a significant weight gain in rats. Both fenofibrate and rosuvastatin inhibited a significant rise in TG and TC levels. Meanwhile, the simvastatin administration failed to do the same. Although statistically insignificant, all treatments increased rats’ HDL levels. Thus, it can be concluded fenofibrate was the most effective treatment among all in reducing weight gain and improving the lipid profile of dyslipidemic rats induced by oral contraceptives and a high-fat diet.

Abstract

This study was conducted to determine the prevalence rate of mares’ GTCT in Indonesia. Ultrasound examination of ovaries were performed on mares in Java and Madura Island of Indonesia. Thirteen mares had been affected by GTCT from 2913 number of examinations. The prevalence rate of GTCT during year 2006 and 2007 were 4.71% and 5.56%, respectively followed by a decrease in prevalence rate of GTCT during 2008-2014. The reproductive cycle in a mare with GTCT had returned to normal 6 months after ovariectomy.

Abstract

This study aims to delineate the tissue structure of male cat external reproductive organs using ultrasound with different types of consoles and transducers frequencies through indirect method. External organ of 5 tom weighing 2-3 kg was evaluated using ultrasound with stationary console (linear transducer, 10 MHz) and portable console (linear transducer, 6.5 MHz), transcutaneously. The results showed that both console and transducer could not visualize the structure of organs sized less than 10 mm through direct method. However, the indirect method using stationary console with a 10 MHz transducer enable to visualize the organ and depicts clearly the internal organ structure such as penis and its parts, scrotum and its constituent layers, caput and cauda of epididymis, and testicles. Furthermore, portable console with 6.5 MHz transducer was still able to provide an adequate image of those organs. In general, fibrous tissues such as tunica vaginalis, tunica Dartos, funiculusspermaticus, and urethra were visible in ultrasonogram as hyperechoic structures, while soft tissues such as the parts of testicles and penis were visible as hypoechoic structures. Tissue containing fluid such as urethral lumen was visible as anechoic structure. In conclusion, indirect ultrasound imaging method was sufficient to visualize the organs sized less than 10 mm using 3-15 MHz transducer in which was currently widely used in animal health care clinics.

Abstract

Penelitian ini dilakukan untuk mengaji respons metafisis tulang femur distalis tikus ovariektomi yang mengonsumsi suplemen kalsitriol selama enam minggu. Lima belas tikus Wistar umur delapan minggu, secara acak dibagi tiga kelompok (normal/N, ovariektomi/Ov, dan ovariektomi + kalsitriol/OvD) masing-masing 5 tikus. Tikus kelompok N dan Ov diberi pakan standar, sedangkan tikus kelompok OvD diberi pakan standar + kalsitriol. Pada umur 14 minggu, tikus dimasukkan kandang metabolik individu untuk studi balan. Pada hari 4-7 studi balan, setiap hari sisa pakan, urin, dan feses dikoleksi untuk pemeriksaan Ca. Pada akhir penelitian, tikus dieutanasia, tulang femur kanan diambil pemeriksaan histopatologis dengan pengecatan hematoksilin dan eosin. Hasil penelitian menunjukkan bahwa konsumsi Ca dan retensi Ca pada tikus OvD tidak berbeda signifikan meskipun cenderung meningkat, sedangkan ekskresi Ca dalam feses dan urin meningkat sangat signifikan (P0,01). Metafisis tulang femur distalis tikus kelompok OvD terlihat zona spikulum tulang trabekula lebih pendek dan berbentuk irreguler, dan rongga sumsum tulang didominasi jaringan adiposit. Dari hasil penelitian dapat disimpulkan bahwa suplementasi kalsitriol selama enam minggu pada tikus ovariektomi menyebabkan osteoporosis pada metafisis tulang femur distalis.

Abstract

 This study was conducted to know the influence of cattle fat and coconut oil in affecting bloodLDL cholesterol. Eighteen male local chickens (Gallus gallus) at the age of 2 months were used. Thechickens were allotted into 3 groups of 6 each. The chickens in each groups were fed with 521 standardration (control groups); 9% cattle fat in standard ration 521 (group II); 9% coconut oil in standardration 521 (group III) for 45 days. All chickens were fed ration and drunk ad libitum every day. At theend of the experiment, the blood samples were collected for analysis blood LDL cholesterol withenzymatic methods using spectrophotometre. Analysis of variance with completely randomized designindicated that treatment significantly effect on blood LDL cholesterol (P0.01). From the results could beconcluded that cattle fat 9% supplementation could be increase blood LDL cholesterol levels. While coconut oil supplementation significantly effect on decrease blood LDL cholesterol levels. Keywords: cattle fat, coconut oil, LDL cholesterol

Abstract

Penelitian ini bertujuan mengkaji aktivitas enzim alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), dan creatinine kinase (CK) pada kambing peranakan Ettawa. Sebanyak 43 ekor kambing peranakan Ettawa dari berbagai kelompok status fisiologis (jantan muda, betina muda, jantan dewasa, betina dewasa, betina bunting, dan betina laktasi) yang secara klinis sehat dan tidak cacat digunakan dalam penelitian ini. Pemeriksaan klinis dan pengambilan sampel darah dilakukan sesuai dengan skedul pemeriksaan dan pengambilan sampel sesuai dengan standard klinik yang diuraikan Baumgartner (1999). Pengambilan sampel darah dilakukan pada jam 7.00-8.00 wib. Pemeriksaan enzim di dalam serum diperiksa dengan menggunakan metode standar sebagaimana diuraikan oleh Kraft dan Duer (1999). Aktivitas AST, ALT, ALP di dalam serum memiliki level yang sebanding dengan level pada ruminansia lain. Sementara itu, aktivitas CK lebih tinggi dibanding nilai yang dinyatakan di dalam literatur untuk ruminansia kecil lainnya.

Abstract

This research aims to explain the evaluation of the differences in four commercial kits for the detection of serological toxoplasmosis used in Indonesia.  The results of the study found that the toxoplasmosis seropositivity determined by the four commercial kits showed a significant difference (P0.05).  Seropositive toxoplasmosis obtained using Pastorex, Toxotest, IDScreen, and Toxo Ab were 35.12%, 60.12%, 26.19%, and 10.12% respectively.  IDScreen had a good agreement with Toxo Ab (Gwet's AC1= 0.623) and a moderate agreement with Pastorex (Gwet's AC1= 0.494-0.511).  Toxotest had a low agreement with three commercial kits (Gwet's AC1= 0.2) but had a moderate agreement with western blotting (WB) and modified agglutination test (MAT) (Gwet's AC1= 0.458-0.557).

Abstract

This study examined the correlation between the concentration of blood metabolites (triglyceride, albumin, phosphorous) on the reproductive performances (S/C and PPM) of lactating Holstein Friesian crossbred (HFC) cows. Blood samples were collected through the caudal vein 8 hours after feeding and the plasma was analyzed at the Integrated Research and Testing Laboratory of Universitas Gadjah Mada (LPPT UGM). Samples were stored at 5° C. Triglyceride, albumin, and phosphorous were measured using glucose oxidase-phenol 4-aminoantipyrine (GOD-PAP) method, photometric bromocresol green method, and cypress diagnostic, respectively. Blood metabolites data were analyzed using Pearson correlation model. The results showed that the correlation coefficients of triglycerides, albumin, and phosphorus levels on PPM and S/C were -0.521 and -0.650; -0.447 and -0.612; -0.513 and -0.700, respectively. In conclusion, there are significant negative correlation between triglyceride, albumin and phosphorus levels on the PPM, and very significant negative correlation between triglyceride, albumin and phosphorus levels on the S/C of lactating HFC cows.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak etanol daun pegagan terhadap gambaran darah dan peningkatan fungsi kognitif pada tikus. Penelitian ini menggunakan 16 ekor tikus Wistar jantan, dibagi secara acak ke dalam 4 kelompok dan masing-masing diberi ekstrak etanol daun pegagan setiap hari dengan level 0, 100, 300, dan 600 mg ekstrak/kg bobot badan selama 8 minggu. Hasil penelitian menunjukkan bahwa pemberian ekstrak pada level 300 mg/kg bobot badan meningkatkan nilai packed cell volume (PCV), sel darah merah (SDM), dan nilai hemoglobin (Hb). Pengamatan aktivitas pada kelompok dengan level 300 dan 600 mg/kg bobot badan menunjukkan adanya peningkatan dibandingkan kelompok kontrol (P0,05). Pemberian ekstrak etanol daun pegagan juga berperan dalam meningkatkan keberhasilan pengenalan lorong target. Hasil di atas mengindikasikan dengan kuat bahwa ekstrak pegagan mempunyai peran penting dalam perbaikan profil hematologi dan meningkatkan fungsi kognitif.

Abstract

This research was aimed to study the effect of administration of glycerol and dimetilacetamida (DMA) in BTS® and MIII®extender supplemented with sodium dodecyl sulphate (SDS) on boar frozen semen. A number of four boars were used in this study for semen collection (n=20). The collected semen was evaluated both macroscopically and microscopically. In this study, only the semen that demonstrated70% sperm motility, 200.106/mL sperm concentration, and20% sperm abnormalities were used and divided into eight tubes. A number of 4 tubes were diluted with 5 mL of BTS, and the rest with 5 mL MIII. The sampel was stored at 20-22° C for 2 hours, followed by centrifugation for 15 minutes (at 2000 rpm), and taken of pellet with 1 ml supernatant. The pellet that was resulted from centrifugation using BTS, then re-diluted with BTS-glycerol 5% (BTSG), BTS DMA 5% (BTSD), BTS-glycerol 5% and SDS (BTSG-S), BTS-DMA 5% and SDS (BTSD-S). Four other pellets that were centrifuged with MIII also re-diluted with MIII-glycerol 5% (MIIIG), MIII-DMA 5% (MIIID), MIII-glycerol 5% and SDS (MIIIG-S), MIII-DMA 5% and SDS (MIIID-S). Next, all of diluted semen were inserted into 0.5 mL straw and equilibrated for 2 hours (4° C), then frozen and stored in liquid nitrogen. The evaluation of frozen semen quality was conducted at 24 hours after frozen. The result of this study showed that post-thawing motility of spermatozoa in BTSD-S (40.17±0.2%) was found higher (P0.05) compared to seven other dilution processes. Therefore, it is concluded that the concentration of 5% DMA that supplemented with SDS in BTS dilution much better for maintaining boars frozen semen quality.
