Abstract

In tree improvement program, vegetative propagules, including grafting, from improved materials will develop into individuals with the same genetic structures from its improved clonal sources. Vegetative propagules are also advantagous due to their earlier flowering and fruiting if compared to seedlings. Grafting success will be etermined by compatibilities between 2 different tissues from different genetic sources. In this study, grafting success of E. occidentalis between rootstocks and entries of different provenans and families were examined using scions collected from mature (6.5 year) trees to hasten deployment of improved genotypes. Reciprocal grafting between provenances with different provenance  rootstocks and scions were conducted with successvaried from none to complete survival. Abilities of two best provenans under waterlogging, Dumbleyung and Young River, to perform highest survival in grafting provide advantages for deployment of their genetic materials. Genotype differences between scions were more obvious to influence survival than between rootstocks. Small number of grafted plants initiated flowers 4 months after grafting. Scion diameter, length and initial leaves affect the growth of new shoots. The best E. occidentalis grafting success were determined by scion length of 2.5 - 4 cm, diameter of 6 - 7 mm with all leaves removed.

Abstract

Acacia mangium has been planted on large scale of industrial forest plantation in Indonesia, especially in Sumatera and Kalimantan islands. It has been reported that large area of mangium plantations have been infected rot root disease caused by Ganoderma sp. To date, there was no information of mangium which resist to Ganoderma sp. The study had by carried out with two aims : (1) isolate a compound with antifungal properties, the antifungal was identified as  p-Methoxybenzylidene p-aminophenol in the category of phenolic compounds. from the roots of healthy mangium, and (2) determine the concentration of antifungal compound from roots of healthy mangium. The roots of healthy mangium from the first generation seedling seed orchard in Wonogiri, Central Java, were used. Mangium roots which had had their external and internal parts separated were  macerated in a solvent of nhexane and methanol. Methods of the isolation of the antifungal compound were thin-layer chromatography (TLC), column chromatography and thin layer preparative chromatography. The antifungal was identified as p-Methoxybenzylidene p-aminophenol in the category of phenolic compounds. Determination of the concentration of the antifungal compound was done by a TLC densitometer on six different families of trees. The results revealed that the antifungal compound was successfully isolated in its from methanol extract from the interior of the root. Results of identification with the TLC densitometer method showed that among the six  families of trees, number 44 had the highest concentration at 40,52% w/w and number 67 showed the lowest concentration at 19,88% w/w.

Abstract

Flowering are influenced by internal factors, such as genetic and phytohormone, and environment factors, such as sunlight and nutrition intake. The flowering characteristics influence fruiting and genetic diversity seedlings through mating systems. This study aims to assess flowering and fruiting characteristics and to determine pattern of mating system of a Calophyllum inophyllum plantation at Watusipat, Gunung Kidul. Flowering and fruiting were observed at 4 locations, 3 parts of crown, and 4 main directions to know the effects of sunlight, nutrition intake and phytohormone in the flowering process. Mating system was assessed by comparing genetic diversity values between parent trees and offsprings. The values of genetic diversity were analyzed using 5 RAPD primers with 17 polymorphic loci. Analysis of variant showed that the locations, crown parts, directions and interaction between a location and direction significantly affected to differences number of flowers and fruits. Values of genetic diversity (h) of parent trees ranged between 0.1471 and 0.3056. The values increased at almost overall offsprings; it ranged between 0.2864 and 0.3750. Values of genetic distance (Da) between parent trees were high and very high (0.197 – 0.364), but the values was decreased between parent trees and their offspring, even between offspring populations. A dendrogram showed two main clusters; first cluster consisted parent trees at up edge with rare trees and second cluster consisted sub cluster parent trees at up edge; sub cluster parent trees at down middle; and sub cluster parent trees at down edge and overall offsprings. Flowering/ fruiting characteristics and pattern of mating systems of C. inophyllum were briefly discussed. Abstract

The first generation of Acacia hybrid between A. mangium and A. auriculiformis potential to be developed for plantation because of its superiority. The superiority of hibrid Acacia were fast growth, straight bole, light branching, more tolerant to pest/disease attack and better wood property than the parent trees. The only way to maintain their superiority is to propagate it vegetatively. The objective of the research was to observed characteristics of hibrid Acacia based on seed viability,sprouting and rooting ability. Materials used in this study were collected seed from control pollination at breeding garden which verified by molecular marker. The result showed that the character of seed viability on hybrid Acacia was still low (48.1%) meanwhile the sprouting and rooting ability were 16.5 ± 0.7 shoots and 78,4% respectively.

Abstract

Callophyllum inophyllum Linn. is one of potential tree species for commercial plantation as biofuel sources. This species was generally propagated by generative method and seldom vegetative method in mass planting stock production. A completely randomized design was arranged in factorial experiment to identify the effect of three grafting methods and four origins of scion in mother tree on the growth of grafted plants. The growth of grafted plants was observed at nursery and the  development of graft union anatomy was observed through microscopic method in laboratory. The result showed that grafting method significantly influenced to shoot number and shoot length, with the best result showed by veneer graft method. Position of scion in mother trees influenced shooting rate and the best result was gained through the scion from the lower part of the tree crown. The survival rate was 72-100%, shooting rate was 68-100% with 1,9 shoots per grafted and average of shoot length was 0,65 cm.The growth of graft union showed earlier by callus formation and then differentiated to a vascular tissue
