Pelita Perkebunan, Coffee and Cocoa Research Journal (CCRJ): ISSN:0215-0212 Since its establishment in 1911, Indonesian Coffee and Cocoa Research Institute (ICCRI) formerly Besoekisch Proefstation, had published its research findings through a journal call Mededelingen van het Besoekisch Proefstation. Between 1948-1981 the research institute was under the supervision of Bogor Research Institute for Estate Crops, and published its research findings through De Bergcultures which was later changed to Menara Perkebunan. Since the institute held the national mandate for coffee and cocoa commodities, and due to rapid increase in the research findings, ICCRI published its first issue of Pelita Perkebunanjournal in April 1985. Pelita Perkebunanis an international journal providing rapid publication of peer-reviewed articles concerned with coffee and cocoa commodities based on the aspects of agronomy, plant breeding, soil science, crop protection, postharvest technology and social economy. Papers dealing with result of original research on the above aspects are welcome, with no page charge. Pelita Perkebunan is managed by Indonesian Coffee and Cocoa Research Institute (ICCRI), which publish the research findings not only for coffee and cocoa but also other commodities relevant with coffee and cocoa, i.e. shade trees, intercrops and wind breakers.

Abstract

Coffee development in Indomesia by means of optimalizing local resources needs to be done for increasing national coffee production as well as for expanding domestic and international markets. These opportunities must be used to gain benefit as a strategic action for raising farmer’s prosperity. This study was aimed to observe physical quality and flavor profile of Robusta coffee from Tambora mountainside, and to identify agribusiness coffee system applied by the farmers, including problem identification at farmer’s level. This research was carried out at Pekat Subdistrict (Dompu District) and Tambora Subdistrict (Bima District), West Nusa Tenggara Province. Direct observation and in-depth interviews were conducted in this study. Data collected consisted of primary and secondary data, as well as 11 green coffee samples from farmers to be analysed its physical quality and flavor profile. The number of respondents were nine stakeholders consisted of three farmers, two farmer group leaders, one field officer, one duty officer, one trader, and one large planter official. Respondents selection were based on convenience sampling method. The results showed that physical quality of coffee bean was belonged to Grade 4—6 (fair to poor quality), while broken beans shared the highest number of physical defects. Robusta coffee from Tambora mountainside performed good taste profile, that the coffee can be promoted to be fine Robusta by improving post harvest handling. Robusta coffee farming at Tambora mountainside was characterized by monoculture cropping system, average of land ownerships about 1 ha/household, and average productivity about 900—1,000 kg green coffee/ha/year. Major problems on Robusta coffee farming at Tambora mountainside consisted of lack of coffee plant maintenance as well as limited accessibility to financing and technology. Key words: agribusiness, physical quality, flavor, Robusta coffee, Tambora mountainside

Abstract

Paecilomyces fumosoroseuswas known as one of the effective biological agents of cocoa pod borer and Helopeltis antonii. To find out the side effect of application of P. fumosoroseuson black ant, Dolichoderus thoracicus, a series of observations were carried out at the Laboratory of Pest and Diseases Indonesian Coffee and Cocoa Research Institute (ICCRI) and in a cocoa plantation of Glenmore, Banyuwangi district, since June until October, 2004. Laboratory research used four concentrations of P. fumosoroseusnamely 105, 106, 107 and 108 spores/ml, while in the field used concentration 2, 4, 6, 8 g dry spores/ml. Each trial as compared with spraying of carbamate and synthetic pyrethroid insecticides as control and untreated This research was designed by randomized block design and four replications. The results showed that in the laboratory, direct spraying suspension of P. fumosoroseuskilled black ant between 20—39% which infected fungi about 2.5—12.5%. The relationship between log of spores concentration of P. fumosoroseus and probit of ant mortality followed the regression equation Y = 3.653 + 0.097 X with LC 50 was 8 x 10 13 spore/ml. The period needed to kill a half of ant population at the laboratory (LT 50) at concentration 107 spores/ml followed the regression equation Y = 1.851 + 1.522 X, with LT50 is 12,01 days. The effect of pyrethroid and carbamate insecticide on ants mortality were 91.25% and 98.75% respectively. In the field, the effect of P. fumosoroseusspray on black ant population was very low, with the percentage of ant mortality at cocoa leaf nest were 0.25–0.46% and at cocoa leaf nest in plastic bag were 0.06–0.21%, while carbamate and pyrethroid synthetic effect were 37.35% and 52.37% at cocoa leaf nest, and 19.15% and 46.67% at cocoa leaf nest in plastic bags. Abstract

For cocoa clonal propagation it need rootstock with good characteristics. Half sibs families is the most appropriate alternative for rootstock selection. This study was aimed to select and estimate genetic parameters of cocoa seedling on the half sibs families of DR 1, DR 2, ICS 60, DRC 15, BLC 4, DRC 16, KEE 2, ICS 13, KW 162, KW 163, PA 300, RCC 70, TSH 858, Sca 6 and Sca 12. Research was carried out in Kaliwining Experimental Station of Indonesian Coffee and Cocoa Research Institute. Genetic parameter were estimated based on variance and covariance component of the variables of the number of leaf, stem height, stem girth, leaf acreage, the volume of fresh root, the length of secondary root and the length of primary root which assessed along seed growth till 90 day after planting. The result showed that most of the recorded variables perform high category of narrow sense heritability (h 2>0.5). Stem girth, leaf acreage, volume of fresh root, length of secondary root and the length of primary root were assessed as the broad category of genetic variability (d2 A >2Sed 2 A ). Therefore, length of primary root and length of secondary root would be the main criterion for rootstock selection as their performance on broad category of genetic variability and high category of narrow sense heritability. Length of secondary root can be estimated based on the performance of stem girth (r = 0.5 * ) and leaf acreage (r = 0.23*) and the primary root can be stimated based on the performance of stem height (r = 0.81 * ) and leaf acreage (r = 0.72*). Based on positive selection of primary root lenght and negative selection of secondary root lenght it was identified that the families of KEE 2 and Sca 12 potentially would be good rootstock due to their long category of the primary root and short category of the secondary root that having characteristics of high water uptake and low vigor. Abstract

Genetic engineering of Robusta coffee resistant to fungal diseases might be done by introducing a chitinase-encoding gene into genome of this plant. This research was aimed to confirm transgenic plant of BP 308 clone Robusta coffee transformed by chi gene and to evaluate its ability for the somatic embryogenesis. Confirmation of transgenic was carried out by analysis the presence of NPTII gene as a selectable marker for Canamysin resistant using PCR technique. The somatic embryo initiation and reproduction were evaluated in 11 plant accessions. Three kinds of sucrose concentration, 20%, 30% and 40% were applied in initiation stage of somatic embryo germination. The suitability of 4 medium, namely M1 (without addition by liquid medium), M2 (addition by liquid medium contained 0.25 mg/l kinetin), M3 (addition by liquid medium contained 0.25 mg/l IAA) and M4 (addition by liquid medium contained 0.25 mg/l GA3 ) was evaluated for somatic embryo maturation. The result showed that 8 out of 10 plant accessions tested were transgenic and they could be propagated through somatic embryogenesis. The ability of transgenic plant for somatic embryo initiation, reproduction and regeneration were similar with that of nontransgenic one. Germination of somatic embryo could be improved by using 40% sucrose. Maturation of somatic embryo could be improved by addition of fresh liquid medium on the ancient gelled medium that used for somatic embryos reproduction. The best result was obtained on addition of fresh medium contained 0.25 mg/l GA 3 in which 65% of the somatic embryos developed to pre-germinate somatic embryo. Abstract

In Indonesia, cocoa reproduction by cuttings is undeveloped yet because the available technology is more expensive than the other clonal reproduction methods. The success of cocoa cuttings is influenced by genetic and environmental factors. The purpose of this research is to study effect of endogenous auxin content, effects of light intensity and exogenous auxin application, on the rooted cuttings. The 2 nd research purpose is to study effects of PVP (Polyvinylpyrrolidon) and IBA (β-indole-butyric acid), clones, and microclimate. The experiment was conducted in Kaliwining Experimental Station of Indonesian Coffee and Cocoa Research Institute in Jember (45 m a.s.l. and D rainfall type according to Schmidt Ferguson). The design for the 1 st experiment was splitsplit plot, replicated three times. The main plot was light intensity inside the roof, i.e.15%, 30%, and 45% to direct sun radiation. The sub plot was cocoa clones, i.e. DR 2 and ICS 13, and the sub-sub plot was IBA concentration, i.e. 0 ppm, 1500 ppm, 3000 ppm and 4500 ppm. The 2 nd experiment was designed factorial 3 x 3 of CRD, replicated 3 times. Clones of KW 163, KW 162 and KW 165, and IBA at 0, 3000, and 6000 ppm were the factors. In the same time, effect of IBA 6000 ppm, PVP 6000 ppm IBA+PVP 6000 ppm, and control were observed using KW 165 clone, and designed in complete randomized design (CRD), replicated 3 times. The result showed that auxin content of ICS 13 was higher than DR 2 (62.67 ppm vs 40.90 ppm) so that gave higher rooted cuttings and more root number. Exogenous application of IBA improved auxin content of the cutting materials and promoted root growth. The optimum IBA concentration for root number was 3500 ppm. Light intensity of 45% improved number of rooted cuttings three times compared to 15%, however compared to the 2 nd research, percentage of rooted cuttings was still very low. Cocoa cutting method to gain rooted cuttings 80—90% has been obtained. The method was using IBA 3000 ppm or 6000 ppm mixtured with or without PVP 6000 ppm, conducted during rainy season, the nursery using permanent shade trees of leucena sp., temperature of 24—27oC and relative humidity was 78—87%. PVP was supposed inhibit oxidation of IBA so that the effect of IBA was more optimum. The response of KW 162, KW 163 and KW 165 clones to root were similar. Rooted cuttings during dry season was low due to the less fresh of cutting materials and high temperature inside the bed roof. Abstract

Cocoa pod borer (CPB, Conopomorpha cramerella  nell.) is a dangerous pest of cocoa which seriously reduce cocoa production mainly in Southeast Asia and Pasific. Prevention of CPB attack can be done by pod sleeving to prevent CPBs lay eggs on pod, or reduction of source of CPB infestation by using pheromone or kairomone as attractant in an insect trap. A preliminary research using sex pheromone has been conducted at endemic cocoa area infested by CPB in East Java. The objective of this study was to evaluate the effectiveness of sex pheromonesin controlling CPB. Trial was arranged by randomized completely block design in four treatments and four blocks as replication. Four densities trap/ha (0, 4, 8, and 12 traps/ha) were used as a treatments. Sex pheromone trap consisted of synthetic pheromone (lure) and sticky liner was hanged on 0.5 m above the cocoa canopy. The results showed that the number of CPB captured during four months was significantly decreased. The number of CPB captured per trap during the first two months in the treatment of 0, 4, 8 and 12 traps/ha were 0, 6.5, 4.72, and 5.58 CPBs, respectively. Four months after treatment, the number of CPB captured in the respective treatments was reduced to 0, 0.25, 0.6, and 0.96 CPBs. Estimate calculation on yield loss due to CPB attack showed that before treatment the yield loss ranged 37.4—45.6%, however six months after treatment, the yield loss in treatment plots decreased to 9.4—21%, whereas on control 38.47%. Use of sex pheromones to attract CPB at a density of 4 traps/ha reduced yield losses due to CPB damage by 67.7%. The significant correlation betweenthe number of CPB captured with the damage intensity followed regression equation of Y = - 0,00044X + 0,32059. Use of sex pheromone for monitoring or masstrapping of CPB, as a component in IPM of CPB is promising, due to its nature for specific target, environmentally friendly, effectiveness, and economic values.Key words: cocoa, cocoa pod borer, Conopomorpha cramerella, integrated pest management, biological control, sex pheromone

Abstract

Because of social economic judgment, many coffee planters nowadays grow Cassia spectabilisand in the certain regions used Cinnamomum burmani, Macadamia integrifolia, Tectona grandisand Cassia siameaas shade trees or intercrops. Before being used in large scale, allelopathy study is appropriate to be done because this effect is much more difficult to be overcome than competiton as growing factor. Research on allelopathy of those species had been conducted in glasshouse of Indonesian Coffee and Cocoa Research Institute using Salisbury & Ross method. Leachate of Cassia spectabilis Cinnamomum burmani, Macadamia integrifolia, Tectona grandisand Cassia siamea, pure media (without plant) and control (well water) were used as treatments. Planting material of Cinnamomum burmani, Macadamia integrifolia, Tectona grandisand Cassia siameawere as seedlings of one year old, whereas C. spectabiliswas 3 months old. Those materials were planted in polybags 20 cm x 30 cm and replicated five times. The media was a mixture of top soil, manure and sand 1 : 1 : 1 (v/v). After those species were maintained for one months and Arabica seedlings for three month old, watering of coffee seedlings then using leachate from shade trees media. Every two days, each seedling was applied with 200 ml. Control was applied with well water. Pure media was used to study the effect of nutrient supply contained in the leachate. The experiment was stopped at seven month old of the coffee seedlings. The result showed that C. spectabilisreleased chemicals which showed allelopathic effect to Arabica coffee, their growth was inhibited 10% to control. The growth decreament from Cassia siameaand D. zibethinustreatment mainly caused by lower mineral content in the leachate and indicated by weak allelopathic. On the other hand M. integrifoliaand C. burmanidid not show allelopathic to Arabica coffee. Thus, based on allelopathy aspect, it can be included that C. spectabilisand C. siamea were not recommended as shade trees or intercrops with Arabica coffee and for D. zibethinusits cropping pattern must be arranged so the mineral competition could be maintained minimum. Abstract

Conversion of cocoa beans to chocolate product is, therefore, one of the promising alternatives to increase the value added of dried cocoa beans. On the other hand, the development of chocolate industry requires an appropriate technology that is not available yet for small or medium scale of business. Breaking and deshelling cocoa roasted beans is one important steps in cocoa processing to ascertain good chocolate quality. The aim of this research is to study performance of rotary cutter type breaking machine for breaking and deshelling cocoa roasted beans. Indonesian Coffee and Cocoa Research Institute has designed and tested a rotary cutter type breaking machine for breaking and deshelling cocoa roasted beans. Breaker unit has rotated by ½ HP power, single phase, 110/220 V and 1440 rpm. Transmission system that use for rotating breaker unit is pulley and single V belt. Centrifugal blower as separator unit between cotyledon and shell has specification 0.5 m 3 /min air flow, 780 Pa, 370 W, and 220 V. Field tests showed that the optimum capacity of the machine was 268 kg/h with 500 rpm speed of rotary cutter, 2,8 m/s separator air flow, and power require was 833 W. Percentage product in outlet 1 and 2 were 94.5% and 5.5%. Particle distribution from outlet 1 was 92% as cotyledon, 8% as shell in cotyledon and on outlet 2 was 97% as shell, 3% as cotyledon in shell. Key words:cocoa, breaking, rotary cutter, quality.

Abstract

Field trial of Bt insecticide formulation with a code of formulation-A +1WP against cocoa pod borer (CPB) (Conopomorpha cramerella) has been conducted at the heavily infested cocoa area in Holo village, Amahai sub district, Central Maluku regency. Formulation-B 2.5 WP as a commercial Bt insecticide which has been distributed widely in the market was used as a comparison Bt insecticide. The results of the trial revealed that compared with untreated treatment, Bt insecticide formulation-A +1WP significantly reduced infestation of CPB, CPB population, and yield losses of dry cocoa bean caused by CPB. At the same concentration (2.5 g/L), the effectiveness of formulation-A +1WP was not significantly different with formulation-B 2.5WP. In the case of production increase, at the same concentration (2.5 g/L) formulation-A +1WP caused higher yield than formulation-B 2.5WP, i.e. 39.9% compared with 35.1%, respectively. Both Bt insecticide formulations tried did not caused phytotoxic against cocoa plant. It is recommended that controling of CPB should use concentration of 2.5 g/L Bt insecticide formulation/L of water, and should be applied at around early morning until 09.00 AM and / or at the afternoon after 15.00 PM. Spraying targets should be cocoa pods at the length of less than 8.0 cm which is not laid eggs of CPB yet. Abstract

Vascular-streak dieback (VSD, Oncobasidium theobromae) is the most prevalent disease of Theobroma cacao L. in Indonesia. This study aims to analyze resistance mechanism to VSD based on terpene profile, leaf anatomy, chitinase, and peroxidase study. Resistant clones of Sulawesi 1 and Sca 6 and susceptible clones of ICS 60 and TSH 858 were used for terpene profile, leaf anatomy analysis, chitinase, peroxides, polyphenol, lignin, and cellulose analysis. Those clones and KEE 2, KKM 22 and ICS 13 were used for peroxides analysis. For trichome study, the resistant clones of Sulawesi 1, Sca 6, KEE 2, and KKM 22, and susceptible clones of ICS 60 and TSH 858 were used. GCMS analysis showed that chromatogram pattern of resistant and susceptible groups were quite similar, but resistant clones contained 22% more components than the susceptible ones. Resistant clones contained groups of pinene, decane, myrcene, and octadecanoic acid, while those substances on usceptible clones were absent. Trichome was thicker on younger leaf, and its density on the basal was higher than that on the middle and tip leaf parts. Trichome density of resistant clone was not always thicker than that of susceptible ones. On resistant clones, stomatal density was lower and width of stomate pits was narrower, while thickness of epidermis layer and pallisade parenchym were higher. Polyphenol content of resistant clones were higher but lignin and cellulose of both groups were similar. Chitinase activity which has a role in hydrolysis of mycelia cell wall was higher on the resistant clones, but peroxides which has a role in polymeration of lignin biosynthesis was similar between both groups. It is concluded that groups of terpene pinene, decane, myrcene, and octadecanoic acid, thickness of leaf epidermis, density and width of stomata pit, and chitinase activity plays important role in cocoa resistance to VSD.
