Abstract

ABSTRACTThe purpose of this research is to find out the profile of secondary metabolite contained in the extract of seeds (Crescentia cujete Linn.) And cytotoxic test against shrimp larvae Artemia salina Leach by BSLT method. Phytochemical tests showed that the extract be expected contained the terpenoid, alkaloid, phenolic, secondary metabolite compounds. The results of qualitative analysis with positive TLC contains phenolics. The result of identification with Spectrophotometer UV-visfor determination of α max from berenuk seeds extract consisted of 3 peaks with α max 326 nm, 258 nm, 222 nm be expected terpenoid compound, alkaloid, and phenolic. The results of toxicity test using shrimp larvae (Artemia salina leach) were divided into 4 groups: 1 control group and 3 groups of concentration series. The concentration of test solution used was 1,000, 100 and 10 μg / ml. The shrimp larvae mortality was observed after 24 hours of treatment. Based on the analysis with probit method, LC50 value of seed extract berenuk to shrimp larvae Artemia salina Leach is equal to 82,30 μg/ml. This shows that the extract of berenuk berries is toxic to shrimp larvae Artemia salina Leach, presence to terpenoid content, alkaloids and phenolics in the extract of seeds berenuk.Keywords : Secondary metabolite; Cytotoxic; BSLT.

Abstract

ABSTRACT The chemicals contained in an extract must be maintained its quality. The aim of this study is to prove both that the ethanol extract of kecapi (Sandoricum koetjape, Merr.) and keluwih (Artocarpus communis) leaves meet the non-specific parameters on the standardization of the extract. The ethanol extract of kecapi and keluwih leaves were extracted using the ultrasonic method. The extract was tested with non-specific parameters including water content, total ash content and metal contamination level. The test results are compared with standard parameter references. Ethanol extract of kecapi leaf have water content of 27.75% ± 1.708, total ash content of 4.856% ± 0.040 and levels of As metal contamination < 0.030 mg/Kg, Hg < 0.002 mg/Kg, Pb < 0.400 mg/Kg, while ethanol extract of keluwih leaf have water content of 23.206 ± 0.091%, total ash content of 9,678 ± 0.306% and levels of heavy metal contamination of As < 0.03 mg/Kg, Hg < 0.006 mg/Kg, Pb < 0.02 mg/Kg. The both extract meet the standard parameters of water content, total ash content and levels heavy metal contamination. Abstract

Bay leaves that it contains flavonoid compounds can inhibit the growth of Escherichia coli bacteria causes diarrhea. Hot extraction method can extract more compounds in the plants, particularly are heat stable. This study aimed to determine the different of total flavonoid content and antibacterial activity against Escherichia coli from bay leaves extracts that extracted with some solvent by reflux. The bay leaves extract was extracted with 70% ethanol, 96% ethanol, and methanol solvent. The spectrophotometric method was used to determine of total flavonoid contents at λmax 431,5 nm and operating time 30 minutes with AlCl₃ reagent. The disc diffusion method was used antibacterial activity test with chloramphenicol as a positive control and dimethyl sulfoxide as negative control. Analysis data of total flavonoid content with One Way Anova statistics, meanwhile the zone of inhibition from antibacterial activity test analysed by Two Way Anova continued with Tukey. The total flavonoid content of bay leaves extract (ethanol 70%, ethanol 96%, and methanol) were 6,089; 5,028; 4,052 mgQE/gram extract. There was significant difference of total flavonoid content between variation solvent and the most highest at 70% ethanol. Different extraction solvent and concentration serial of bay leaves extract showed significant difference and the most highest zone of inhibition at extract with 96% ethanol solvent.

Abstract

INTISARI Senyawa flavonoid dalam daun kelor diduga mampu meningkatkan kelarutan kalsium batu ginjal. Penelitian ini bertujuan untuk membuktikan kemampuan ekstrak metanol (EMDK) dan ekstrak etanol daun kelor (EEDK) dalam meningkatkan kelarutan kalsium  batu ginjal secara  in vitro. Daun kelor diekstrak dengan cara maserasi menggunakan cairan penyari metanol dan etanol 70%. Kandungan senyawa aktif golongan flavonoid diidentifikasi secara kualitatif dengan KLT. Serbuk batu ginjal direndam dalam seri konsentrasi EMDK dan EEDK (2%, 4%, 6%, 8%, dan 10%) dan diinkubasi selama 6 jam pada suhu 37 0C. Kadar kalsium terlarut dianalisis dengan SSA pada panjang gelombang 422,7 nm. Hasil penelitian membuktikan bahwa EMDK (2-10)% dan EEDK (4-10)% mampu meningkatkan kelarutan kalsium batu ginjal secara  in vitro  (p≤0,05). Kelarutan kalsium batu ginjal dalam EMDK dan EEDK mengikuti pola tergantung konsentrasi. Kadar kalsium batu ginjal terlarut dalam EMDK dan EEDK berturut-turut berkisar antara (86,27-185,87) ppm dan (95,31-177,29) ppm. Hasil analisis dengan KLT membuktikan bahwa senyawa aktif golongan flavonoid berhasil terdentifikasi dalam EMDK dan EEDK.  Kata Kunci: Batu ginjal kalsium, daun kelor (Moringa oleifera Lam.), flavonoid, maserasi   ABSTRACT Flavonoids  in  kelor leaves  (Moringa oleifera Lam.)  suspected  had  an  important  role  on dissolving  calcium  kidney  stones.  The  purpose  of  this  research  is  to  reveal  the  effect  of  kelor leave's methanol extract (KLME) and ethanol extract (KLEE) on the in vitro solubility of calcium kidney  stones. The kelor leave's  simplisia extracted with  maceration  method.  Methanol and ethanol 70% were  used  as  solvent. An  active  flavonoid  compound  in  KLME  and KLEE has identified  with  TLC  method. Kidney  stone's  powder  soaked  in  KLME  and KLEE  series concentration  (2%, 4%, 6%, 8% and 10%) and  incubated  for  six hours at 37 °C. The dissolved calcium levels from kidney stones in KLME and KLEE were analyzed with AAS at a wavelength of 422.7  nm. The result  showed  that  the  KLME  (2-10) % and KLEE (4-10%)  can  enhance  the dissolved calcium levels from kidney stones in vitro (p<0.05). The dissolved kidney stone's calcium in KLME and KLEE deppend on it’s concentration. The  levels of dissolved calcium  from kidney stones  in KLME  dan KLEE  series  concentration were (86.27-185.87) ppm and (95.31–177.29) ppm repectively. Abstract

INTISARI   Umbi bawang dayak memiliki kadar antioksidan yang sangat tinggi. Penelitian ini bertujuan mendapatkan formula masker gel feel off ekstrak umbi bawang dayak yang menghasilkan pembentukan film yang baik berdasarkan hasil evaluasi fisik. Masker gel peel off dibuat dalam 3 formula dengan parameter perbedaan konsentrasi PVA. Formula 1 PVA 10 %, formula 2 12%, formula 3 PVA 13%. Proses pembuatan ekstrak dengan cara maserasi, kemudian dipekatkan dengan evaporator dan dibuat sediaan masker gel peel off dengan penambahan eksipien. Hasil uji organoleptis menunjukkan parameter aroma, warna dan tekstur tidak berbeda nyata antar formula. Hasil uji fisik menunjukkan formula 3 dengan PVA 13 % menghasilkan masker gel peel off yang paling baik dibandingkan dengan formula 1 dan 2, dengan nilai viskositas 3,833 cps, pH 6, homogeny, waktu mongering < 15 menit dan uji daya sebar 5, 815 cm. Kata kunci: bawang dayak, masker gel peel off, PVA.   ABSTRACT   Bawang Dayak bulbs have a very high antioxidant content. This study aims to get the mask gel feel off formula the bawang Dayak bulb extract that results in good film formation based on the results of physical evaluation. The mask gel Peel off is made in 3 formulas with different parameters of PVA concentration. Formula 1 PVA 10%, formula 2 12%, formula 3 PVA 13%. The process of preparing the extract by maceration, then concentrated with evaporator and made gel mask preparation with the addition of excipients. The results of organoleptic tests showed the parameters of aroma, color and texture were not significantly different between the formulas. Physical test results showed that formula 3 with PVA 13% yielded the best peel off gel mask compared with formula 1 and 2, with viscosity value 3,833 cps, pH 6, homogeny, mongering time <15 min and spreading test 5,515 cm. Abstract

ABSTRACTPumpkin (Cucurbita moschata Duch ex Poiret), a fruit that contains several compounds, has activities to protect the human body from free radicals. Provitamin A is recognized as an antioxidant compound. Antioxidants can protect the body’s damage caused by free radicals. The purpose of this study was to isolate beta carotene compounds and test the purity of beta carotene isolates obtained using DR-UV and infrared spectrophotometry. Pumpkin is extracted using acetone: petroleum ether with a ratio (1: 1). Identification of beta carotene from extracts was carried out using TLC with the mobile phase of ethanol: acetone: n-hexane with a ratio (2: 1: 1) and scan spectrum using a UV-Vis spectrophotometer. Beta carotene was identified as the result of TLC elution on UV 254 and 366 nm which showed 2 spots. Detection of beta carotene spectra was carried out in the range of 350 to 550 nm and obtained beta carotene peaks at wavelength x nm. The extract purification process was carried out using Preparative TLC with the mobile phase of ethanol: acetone: n-hexane with a ratio (2: 1: 1) and obtained isolates cleaned first using chloroform: methanol ratio of 1: 1. The isolates were dried and tested using DR-UV and infrared spectrophotometry. Abstract

ABSTRACT Breast cancer is the most common cancer found in women and is the dominant cancer in Indonesia. Cancer treatments such as chemotherapy can affect the quality of life of patients including physical, psychological, and social. The purpose of this study is to measure the quality of life of breast cancer patients. This research used cross sectional study design. The patient quality of life was measured using the EORTC QLQ-C30 questionnaire. The subjects of this study were breast cancer patients in RSUP Dr. Kariadi Semarang meeting the inclusion criteria. The cognitive function domain, social function, emotional function, role function, and physical function were 86,15 ± 17,82; 84,10 ± 21,13; 82,56 ± 17,72; 77,95 ± 29,77; 76,72 ± 22,95, respectively. Whereas in symptom scale group from pain, fatigue, insomnia, appetite loss, financial difficulties, nausea and vomiting, constipation, dyspnea and diarrhea were 32,82 ± 30,33; 32,65 ± 21,14; 31,79 ± 34,58; 31,79 ± 32,51; 30,77 ± 32,44; 23,59 ± 25,67; 17,95 ± 30,9; 4,62 ± 14,28; 4,10 ± 13,83, respectively. For global health status / quality of life obtained 72.18 ± 18.94. The highest score on the functional scale is the cognitive function domain and the lowest score is the physical function.On the scale of symptom the domain that has the highest score is the pain domain and the lowest score is the diarrhea domain. Abstract

ABSTRACT Avocado’s leaves and Curcuma xanthorrizarhizoma contain compounds of flavonoids, alcaloids,triterpenoids and curcumin, where the compounds are effective as antidiabetic agents. This study aims to prove the effect of antidiabetic combination of extracts on insulin resistence type-2 diabetic rats then to compare it with a single extract. This research used pre-test and post-test control group design. The 30 test animals were divided into two treatment groups. Five control groups of rats were they given 1,25 mL aquabides/KgBW/day, and 25 rats were divided into five groups and given were given 1,8 IU insulin/KgBW/day for 14 days. Group I was control of 12,5 mL CMC-Na/KgBW/day 0,5%, group II was positive control of 150 mg metformin/KgBW/day, group III was avocado’s leaves extract, group IV was extract of Curcuma xanthorriza rhizoma, and group V was combination extract. The extracts of group III-V were given the same dose, that was given 200 mg/KgBW/day for 14 days. The meansurement of antidiabetic effect was done by comparing the GDP value of rats before and after treatment. The GDP concentrations of rats were analyzed using T-paired testand Wilxocon test, whereas the comparative data of percentage GDP decrease was tested using Kruskall Wallis test then followed by Mann-Whitney test. The results showed that the effect of antidiabetic combination of extract in insulin resistence type-2 diabetic rats as the percentage GDP decrease was 22.03%. The effect of decreasing GDP concentration at the ratio of 1:1 extract did not differ significantly with the single extract of 200 mg Curcuma xanthorrizarhizoma/KgBW/day. Abstract

ABSTRACTLycopene is one of the phytochemical compounds of the carotenoid group, the main producer of red pigments found in watermelons. Watermelon is a round-shaped vines containing vitamin C and vitamin A and has high antioxidant levels. This study aims to identify lycopene compounds in watermelons. Extraction of watermelon flesh by maceration method. Identification of lycopene compounds with UV-Visibel spectrophotometry instrument followed by identification with the TLC method and fractionation of extracts using preparative TLC with chloroform: ethanol (1: 1) solvents, and identification of fractionation results with Fourier Transform Infrared Spectroscopy (FT-IR). The results of the identification of watermelon fruit extract using the TLC method obtained three spots and the results of UV-Vis spectrophotometry showed three peaks namely between 440-520 nm. The identification of fractionation of watermelon extract using FT-IR obtained a constituent group of lycopene structures, but did not show the R-CH = CH-R group that is owned by lycopene, whereas the results of the test with UV-Vis spectrophotometry appearing at 206 and 245 which indicate the presence of other compounds. From the results of the study it can be concluded that the testing of known extracts of watermelon fruit contains lycopene compounds, but the testing of lycopene identification on the results of fractionation of watermelon fruit extracts was not found with certainty the existence of lycopene because the FT-IR results were only shown and contained on the results UV-Vis spectrophotometry was found to be the wavelength of other compounds.keywords: Watermelon, Lycopene, UV-Vis Spectrophotometry, IR Spectrophotometry
