Abstract

Nifedipine is an anti-hypertensive included in the Biopharmaceutics Classification System (BCS) class II to its nature of being poorly soluble in water, resulting in low bioavailability. One technique that can be used to improve the solubility of the dispersion technique. This research aimed to determine the effect of the concentration of PVP K-30 on physical characteristics, the dissolution rate of solid dispersions, and the interactions that occur between nifedipine with PVP K-30. The method of manufacturing solid dispersion is a melting-solvent method. FI - F III is a solid dispersion and F IV-VI F is a physical mixture. Formula used is the ratio of nifedipine: PVP K-30 FI (10%: 90%), F II (20%: 80%), F III (30%: 70%), F IV (10%: 90%), FV (20%: 80%), F VI (30%: 70%), and used as a control of pure nifedipine. Testing was conducted on the organoleptic test, flow rate, moisture content, melting point, dissolution testing, and FT-IR analysis. Dissolution test parameters used C (20). The data was obtained and analyzed by non-parametric test KruskalWallis and Mann-Whitney with a level of 95%. The results showed that solid dispersions aregranulated, yellow, and have a free-flowing nature. Test results C (20) FI, F II, F III, F IV, FV, F VI, and controls respectively were 89,48%; 51,75%; 22,58%; 12,88%; 24,07%; 26,34%; 24,95%. The results showed that the FI has the highest dissolved. Results of FT-IR spectra showed no interaction between nifedipine with PVP K-30.

Abstract

Endophytic fungi are potential microorganisms in lead compounds investigation as research objects for antibacterial and antifungal drug candidates. These microorganisms are able to produce various secondary metabolites, moreover they can even be regulated according to the needs of researchers. This is an advantage of endophytic fungi with good adaptability response. This study aims to explore the antibacterial, antifungal and antioxidant activity of endophytic fungi from the branch of the Melinjo tree (Gnetum gnemon L.) by qualitative analysis. This study resulted in 13 potential endophytic fungi extracts to be explored further for their compounds. Based on the results of the TLC test of the fungal extracts with 10% vanillin in H2SO4 as a reagent, it showed that the dominant compounds were alkaloids and polyphenols. The antibacterial activity test of extracts of the endophytic fungi was carried out by the disc diffusion agar method against Staphylococcus aureus and Pseudomonas aeruginosa bacteria, while antifungal agents were used for Candida albicans and Aspergillus clavatus.. Meanwhile, the qualitative test of antioxidants from fungal extracts using the DPPH spray reagent showed positive results of 8 fungal extracts. Screening of endophytic fungi from melinjo represented a source of endophytes potential as antibacterial, antifungal and antioxidant to be developed in exploration of secondary metabolites from natural products of microbial.

Abstract

In order to discover and identify antimalarial active compounds, jengkol peel extract was isolated and tested for antimalarial activity. This study aims to determine the antimalarial active compound from jengkol peel extract which functions as an inhibitor of the Malate Quinone Oxidoreductase (MQO) enzyme. In this study, the extraction process was carried out using methanol as a solvent with the maceration method. The maceration results were then concentrated using a rotary evaporator with low pressure at a temperature of 40-45°C to obtain a Crude extract of 203 grams. Then liquid-liquid fractionation was carried out with four types of solvents, namely n-hexane, ethyl acetate, methanol, and water according to the level of polarity. Each fraction obtained was 400 ml which was then tested for enzyme inhibition using the MQO enzyme formation inhibition method. The fraction that had the highest inhibitory activity was the ethyl acetate fraction of 80%. Confirmation was done again with the same separation using column chromatography so as to produce the ethyl acetate fraction which was confirmed to be identified using HPLC producing peaks at retention times of 22 and 23 minutes. The results showed that in jengkol peel extract there were active compounds that acted as inhibitors of the MQO enzyme but further tests were needed to confirm these peak compounds.
