Abstract

One of well-established biological activities for chalcone derivatives is as acetylcholinesterase inhibitors, which can be developed for the therapy of Alzheimer’s disease. Assisted byretrospectively validated structure-based virtual screening (SBVS) protocol to identify potent acetylcholinesterase inhibitors, 80chalcone derivatives were designed and virtually screened. The F-measure value as the parameter of the predictive ability of the SBVS protocol developed in the research presented in this article was 0.413, which was considerably better than the original SBVS protocol (F-measure = 0.226). Among the screened chalcone derivatives two were selected as potential lead compounds to designpotent inhibitors for acetylcholinesterase: 3-[4-(benzyloxy)-3-methoxyphenyl]-1-(4-hydroxy-3-methoxyphenyl)prop-2-en-1-one(3k) and 3-[4-(benzyloxy)-3-methoxyphenyl]-1-(4-hydroxyphenyl)prop-2-en-1-one (4k).

Abstract

Dexamethasone as glucocorticoids has been widely used to reduce inflammation and tissue damage in a variety of conditions. It was commonly prescribed in combination with the other compounds such as chlorpheniramine maleate for the relief of asthma. It was important to observe the quality and safety of the combination dosage form, called pulveres or puyer in order to serve the patient oriented medication. This research aimed to develop analytical method validation and determine the content uniformity of the divided powder containing dexamethasone compounded by pharmacy in Yogyakarta and hospital in Central Java, Indonesia. A validated high performance liquid chromatography (HPLC) method was chosen to determine dexamethasone in the samples. The column of ACE 5 C18 (250 x 4.6 mm) was used in this research and the methanol:water (65:35) was used as the mobile phase at the flow rate of 1 mL/min. The UV detection was performed at 239 nm. Analytical method validation was performed on the parameters of system suitability, limit of detection, limit of quantification, accuracy, precision, and linearity. The results showed that dexamethasone contains in divided powder compounded by pharmacy in Yogyakarta and hospital in Central Java were met the acceptance requirement.

Abstract

Daidzein, one of the isoflavone aglycones contained in tempeh, has several biological activities such as anti-inflammatory, anti-oxidant, anti-breast cancer, and suppression of expression of matrix metalloprotease-9. As a fermented product from soybeans, daidzein content in tempeh was found in higher concentration compared to the soybean raw material. It was important to optimize several factors affecting extraction process such as particle size of tempeh simplicia, extraction time, and solvent in order to develop an effective method for the daidzein isolation from the tempeh or other natural products. Evaluation of extraction factors was conducted by applying variations for each factor followed by quantitative analysis using HPLC methods. The optimization condition was performed by daidzein standard and achieved with the particle size of tempeh simplicia of 1.2 mm, extraction time of 360 minutes, and 70% ethanol was used as solvent. Furthermore, the optimized condition was applied for the daidzein isolation from tempeh, a soybean fermented product.

Abstract

Resistensi terhadap beberapa antibiotik disebabkan oleh adanya penggunaan antibiotik yang rasional masih sering diabaikan. Kemangi (Ocimum basilicum L.) merupakan salah satu tanaman yang memiliki aktivitas antibakteri. Salah satu kandungan kemangi yang diduga memiliki aktivitas antibakteri yaitu minyak atsiri. Penelitian terkait aktivitas antibakteri pada kemangi sebagian besar dilakukan dengan menggunakan sampel segar maupun simplisia dari daun kemangi yang kemudian diekstraksi, namun ada beberapa uji yang menggunakan bagian batang. Metode uji aktivitas antibakteri kemangi terhadap beberapa bakteri gram positif dan gram negatif dilakukan dengan metode difusi cakram agar, difusi cakram kertas, mikrodilusi broth, mikrodilusi dalam microtiter plates, difusi sumuran agar, dan macro-dilusi. Hasil uji aktivitas antibakteri menunjukkan bahwa kandungan minyak atsiri yang terdapat dalam kemangi memiliki aktivitas penghambat aktivitas bakteri.

Abstract

Penjaminan mutu produk obat kombinasi salah satunya dilakukan melalui analisis kuantitatif. Teknik kemometrika menjadi salah satu alternatif pilihan pengenalan pola pada analisis kuantitatif senyawa obat kombinasi tanpa tahap pemisahan yang mudah diterapkan, sensitif, dan cukup terjangkau. Keunggulan teknik kemometrika seperti ini telah dibuktikan pada berbagai penelitian yang relevan. Tujuan artikel review ini adalah untuk mengetahui lebih lanjut pemanfaatan teknik kemometrika pengenalan pola pada analisis kuantitatif senyawa obat kombinasi tanpa tahap pemisahan. Penyusunan artikel review ini menggunakan metode penelusuran literatur ilmiah primer terbitan sepuluh tahun terakhir pada rentang 2011 hingga 2021. Hasilnya menunjukkan bahwa teknik kemometrika yang paling banyak diaplikasikan adalah PLS 78,95% dengan instrumen tanpa tahap pemisahan yang paling banyak digunakan sebagai kombinasinya adalah spektrofotometer UV-Vis 84,21%. Selain itu, campuran obat dari dua senyawa 71,05% pada berbagai golongan obat menjadi jumlah campuran yang paling banyak dianalisa dengan kombinasi teknik kemometrika dan instrumen analisis.

Abstract

The reference method for simultaneous analysis of drugs is chromatography, however, this technique is expensive, complex, and needs excessive sample preparation; therefore, some simple methods like UV spectroscopy is proposed. Assisted with multivariate calibration, it is possible to analyze drugs using UV spectroscopy without prior separation. This study is intended to use UV spectroscopy coupled with multivariate calibration of partial least square (PLS) for simultaneous analysis of paracetamol (PCT), propyphenazone (PROPI), and caffeine (CAFF) in tablet dosage form. The calibration model is prepared by developing a series 20 mixture of PCT, PROPI and CAFF with certain composition randomly and its absorbance is measured at wavelength of 220-313 nm with an interval of 3 nm. The performance of calibration model was assessed by coefficient of determination (R2), root mean square error of calibration (RMSEC) and root mean square error of cross validation (RMSECV). The R2 values for the correlation between actual values of PCT, PROPI and CAFF and predicted values using UV-spectroscopy combined with PLS are 0.9994; 0.9878; and 0.9919, respectively. The calibration errors expressed with RMSEC are 0.027%, 0.082% and 0.043% for PCT, PROPI and CAFF, respectively. While, during cross validation using “leave one out” technique, RMSECV values obtained are 0.062%, 0.095% and 0.982%, respectively for PCT, PROPI and CAFF. The level of drugs obtained are 226.76 ± 14.49 mg/tablet (equivalent to 90.70% from labeled claim) for PCT,  135.74 ± 11.23 mg/tablet (equivalent to 90.49% from labeled claim) for PROPI and 51.69 ± 2.35 mg/tablet (equivalent to 103.38% from labeled claim) for CAFF.

Abstract

The limitations of science and students' awareness of reproductive health causes such students susceptible to various diseases caused by viruses, fungi, or bacteria. This program aims to provide education about "Reproductive Health for Students in Yaketunis Yogyakarta through the media “Pakem Braille", and examples of diseases that can be caused due to lack of maintaining health and hygiene of reproductive organs. The program is expected to increase interest in learning, especially science and awareness of students of MTs Yaketunis in maintaining reproductive health, producing the media "Pakem Braille" and learning modules, and also an article publication. The method used in educational activities consisted of a lecture with and without the media "Pakem Braille", where the media will be used as props in the educational process. Overall the students' understanding regarding the material of reproductive health in humans increased by 77.78%. Students’ interest also increased, it is seen from the increase in the number of questions asked by students amounted to 61.11%. Knowledge and awareness of reproductive health-related students increased to 73% and the result obtained was 55.6% of students categorized as having sufficient awareness and 38.9% of students considered to have good awareness of the importance of maintaining reproductive health.

Abstract

A cyclooxygenase-2 (COX-2) inhibitor celecoxib has been previously reported to have cytotoxic activities towards gastric, prostate, ovarian, colon and breast cancer cell lines. This article reports that the cytotoxic activities of celecoxib could be resulted from its activity as a potent ligand for estrogen receptor alpha (ERα). Aided by molecular docking simulations, an in silico test to examine whether celecoxib is a ligand for estrogen receptor alpha (ERα) was performed followed by in vitro test employing cytotoxic assay using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) method. The compound was extracted from Celebrex®. Measured by using UV spectrophotometric method at 255.5 nm, it was identified that the content of celecoxib was 102.15 mg/271.48 mg capsule content. The in silico test indicated that celecoxib is a potent ligand for ERα. This finding was confirmed experimentally by an in vitro test that celecoxib has a comparable activity as an ERα ligand to tamoxifen, a drug of choice for breast cancer treatment.

Abstract

Food supplement which contains vitamins and stimulants such as caffeine were classified as energy drink. TLC-densitometry method was chosen to determine the pyridoxine, nicotinamide, and caffeine in the energy drink sample. TLC plates of silica gel 60 F254 was used as the stationary phase and methanol : ethyl acetate : ammonia 25% (134:77:10) was used as the mobile phase. The correlation coefficient for each pyridoxine, nicotinamide, and caffeine were 0.9982, 0.9997, and 0.9966, respectively. Detection and quantitation limits of from the three analytes were 4.05 and 13.51 µg/mL; 13.15 and 43.83 µg/mL; 5.43 and 18.11 µg/mL, respectively. The recovery of pyridoxine, nicotinamide, and caffeine were within the required limit range of 95-105%. The percent of RSD were below the limit value of 5.7% for caffeine and nicotinamide and 8% for pyridoxine. The content amount of pyridoxine in the sample 1 and 2 were 33.59 ± 0.981 and 30.29 ± 2.061 µg/mL, respectively. The content amount of nicotinamide in the sample 1 and 2 were 106.53 ± 3.521 and 98.20 ± 3.648 µg/mL, respectively. The content amount of caffeine in the sample 1 and 2 were 249.50 ± 5.080 and 252.80 ± 2.640 µg/mL, respectively. Robustness test results showed that the most optimal method conditions should be applied for the analysis.
