Abstract

Aegelin is an alkaloid which isolated from Maja (Aegle marmelos Corr.) leaves. The molecular structure of the compound contains a chiral carbon atom. In our earlier publications the existence and influence of a chiral carbon atom have not been discussed yet. The existing of the chiral carbon atom gave rise two pair of doublets at d 3.67 J=3.4 Hz and d 3.43 J=8.5 Hz in the proton nmr spectrum. The doublets are signal of two proton which attached on adjacent atom of the chiral carbon atom which is confirmed bythe two dimension nmr spectrum (HMQC). The two protons are magnetic unequivalent which cause their chemical shifts are different. The protons appear as two doublet with different J value because its couple to a proton attached at the chiral carbon atom and the spin-spin splitting is observed. The existence of the two pairs of the doublets at d 3.67 d,J=3.4 Hz and d 3.42 d,J=8.5 Hz because the isolated aegelin from E.marmelos leaves contains of its enantimeer.Key words: E. marmelos, aegelin, enantiomeer, spectroscopy

Abstract

A research about inhibitory activity on various cancer related protein kinases of several marine sponges collected in Indonesia was performed. This study aims were to isolate and to identify structures of biologically active sponge metabolites.A combination of a chemically-and biologically driven approach for drug discovery was employed. Sponge extracts were tested by protein kinase inhibitory assay by in vitro method in parallel to the usage of TLC, and HPLC coupled to UV spectrophotometry and mass spectrometry to isolate the chemically most interesting substances. Structure identification of active compounds was performed by using NMR spectroscopy and massspectrometry methods.This study showed that only Axynissa sp. fractions were found active in protein kinase inhibitory assay. Two bisabolene type sesquiterpenoids, (+)- curcuphenol (1) and (+)-curcudiol (2) were identified as active compounds from the sponge. Compound 1 showed SRC protein kinase inhibition with an IC50value of 7.8 μg/mL while 2 inhibited FAK with an IC50 value of 9.2 μg/mL.Key words: protein kinase inhibitor, Axynissa sp.

Abstract

Seventeen traditional medicinal plants of South East Sulawesi are collected at Arboretum Prof. Mahmud Hamundu Universitas Haluoleo, have been determined successfully by Herbarium Bogoriense staffs, Cibinong Bogor. The plants came from Konawe and KonaweSelatan districts. Endemicity of those plants as medicinal plants were based on ethnobotanical study using information from “Tokoh Masyarakat Tolaki” and “Tokoh Masyarakat Jawa”. Collected traditional medicinal plants at those places, but have not known in Java, are assumed as specialized plants of Tolakinese. All of the plants were determined at Herbarium Bogoriense. Moreover, detailed study of those plants including biology, chemistry and pharmacy aspects were done by electronically literatures study using science direct programmed. According to the literatures, Sterculea cf. oblongata R.Br. and Talinum triangulare Willd have not been reported the research yields. Abstract

Oleandrin is one of cardenolida compound isolated from an active fraction of Nerium indicum Mill leaves. (fam. Apocynaceae), which have cytotoxic effect on several human cancer cells, and also to normal cells in vitro. Another compound which was potential as renal anticancer has also been isolated from N. indicum. However, its chemical structure has not been discovered. The aim of this study was to identify the potential compound as selective to renal cancer present in the leaves of N. indicum.The potential compound was isolated from the active fraction using Preparative TLC and structure elucidation was done by using spectroscopic methods (UV, IR, MS and NMR).Base on this spectra and by comparison with oleandrin data, it was indicated that the potential compound as Renal anticancer was as 16,17-dehydrodeacetyl-5a-oleandrin.Key word: N.indicum, oleandrin, cytotoxic, renal anticancer, spectroscopic

Abstract

Hypertension is a disease which need a complex therapy and high cost of drugs. This fact urge exploration of new antihypertensive drug. The aim of the research was to asses to look at the activity of curcumin and its derivatives bisdemethoxy curcumin and bis(demethoxydehydroxy) curcumin on the contraction of isolated organ smooth muscle induced by an α1-adrenergic receptor agonist (phenylephrine), and to assay relaxation effect on this organ. In this study, aortic organ represented a vascular system. This research was conducted using an isolated organ technique with isotonic transducer. The results have shown that curcumin and its analogs bisdemethoxy curcumin and bis(demethoxydehydroxy) curcumin could inhibit the contraction of isolated organ smooth muscle, and showed relaxation effects of this organ. The inhibitory effect on the isolated organ might be mediated by non-competitive antagonism mechanism on a1-adrenergic receptor, whereas the relaxation effects were suggested through nitric oxide (NO) independent pathway.Key words : curcumin, isolated organ, vasodilation, relaxation

Abstract

Endophytic microbes is a potentially new field for producing the promising secondary metabolite and a few people utilizing them. The purpose of this research is to find out the fungi which have an ability to produce new pathogenic eukaryote inhibiting antibiotic. The researches steps were isolation, selection based on the ability of isolates to utilize carbonsources and their inhibitory effect by bioassay test with indicator microbes such as Fusarium oxysporum f.sp. licopersicae, Bacillus subtilis and Candida albicans. Selected isolates was determined by their inhibitory effect value was more than 4.0. Paper chromatography technique was applied to analysis the character of antibiotic using many kinds of eluents. Optimization was done to increase the production and inhibitory effect of produced antibiotic. Isolation research step found 86 endophytic fungi isolates from many kinds of plants tissue from Yogyakarta. Selection results showed that 9 isolates had inhibitory effect value more than 4.0 and four isolates that were JA-2, MB-1, NGK-1 and KMD-7 higher than 5.0. Antibiotic production was influenced by carbon sources or kinds of substrates, for examples JA-2 isolate grown at PDB medium produced higher inhibitory effect to B. subtilis than Antibiotic-3 and GY; NGK-1 grown at Antibiotic-3 medium produced higher inhibitory effect to B. subtilis and C. albicans than others; MB-1 grown at Antibiotic -3 medium produced higher inhibitory effect to F. oxysporum than others. Finally isolates JA-2 and NGK-1 was chosen as selected isolates for development of new antibiotic.Key words : Secondary metabolite, endophytic fungi, Indonesia

Abstract

The flowers of kamilen (Matricaria chamomilla, L.) has been used as antifungal in Indonesian traditional medicines. Previous study showed that the extract of flowers inhibited Candida albicans activity. In order to determine the active fraction, the using bioactive-guided fractionation method was used.The air-dried flowers was extracted from methanol. Methanolic extract was fractionated by using n-hexane, chloroform and ethyl acetate. n-Hexane fraction was separated by vacuum liquid chromatography (VLC) by busing silica gel H 60 with mobile phase n-heksan : ethyl acetate and chloroform : methanol by gradient polarity. VLC results fraction was separated by preparative thin layer chromatography with silica gel GF254 and mobile phase n-hexane : chloroform (1:1). The anti-fungal activities of Candida albicans was done by difuse method.Based on the TLC appearence , nine fractions were obtained (FK 1-9) from VLC, and FK-3 was the most active fraction in inhibiting Candida albicans. The result from preparative TLC of FK-3 was 5 subfractions (FP 1-5) Subfraction FP-3 and FP-5 were most active. that had inhibiting radical zone diameter 18 mm in 3% consentration.Key words : Kamilen ( Matricia chamomila, L), Candida albicans, anti-fungal.
