Abstract

Colorectal cancer is the tenth most common form of malignant tumor of hospital inpatients inIndonesia. Advance approaches in anticancer development is discovery molecular-targeted drugs.Molecular targets for anticancer drug have been identified including genes associated with cellcycle control and angiogenesis. Previously, an active and selective compound against WiDr fromTithonia diversifolia (Hemsley) A. has been isolated. The aim of this study was to evaluate theeffect of the isolated active compound fromT. diversifolia on the WiDr cell cycle and angiogenesis.Isolation of the active compound was performed by preparative thin layer chromatography (TLC)method. WiDr cell cycle was analyzed by flowcytometry using propidium iodide (PI).Antiangiogenesis effect was evaluated by immunocytochemistry method using anti-human VEGFmonoclonal antibody. The results showed that the effect of the isolated active compound onthe WiDr cell cycle depended on the concentration and the incubation time periods. Atconcentration of 4 μg/mL, it inhibited the WiDr cell cycle SubG1 phase after 36 and 48 hoursincubation and G1 phase after 72 hours incubation. While at concentration of 8 μg/mL, it clearlyinhibited the WiDr cell cycle G1 phase after 36, 48 and 72 hours incubation. Furthermore, theisolated active compound at concentration of 4 μg/mL significantly inhibited the VEGF expressionuntil 47.38% compared to control. In conclusion, the isolated active compound fromT. diversifoliainhibited cell cycle and angiogenesis of WiDr cell.

Abstract

Diclofenac sodium is a nonsteroidal antiinflammatory drug (NSAID), which is drug of choice for rheumatoid arthritis. It is commercially available as enteric coated tablet and emulgel for topical dosage form.Topical drug was prepared and developed for systemic purposes in the treatment of rheumatoid arthritis, to reduce the risk of adverse drug reaction given orally. As an oral antirheumatoid drug, it caused dizziness and irritation on gastric mucosa.The study was carried out to assess the effect of semisolid formulation variation on its percutaneous absorption. The experiment was performed by preparing 4 formulas of ointment i.e. hydrocarbon base, washable base, 0/W emulsion base, W/O emulsion base and commercial cream E as a control drug. All of the formulas and standard cream were tested in vitro for the rate of dissolution and its percutaneous absorption on rabbits.The Dissolution Efficiency (DE) of washable emulsion, 0/W emulsion base,- and the commercial cream was correlated with their Ka from the experiment of percutaneous absorption. Among the three formulas, the C ointment (0/W emulsion base) showed the best correlation. The order of the correlation declined as follows C, B and E. From the result it can be concluded that the change of formulation could affect the dissolution and in turn could affect the absorption of the active compounds.Key words : ointment - cream - diclofenac - in vitro - in vivo

Abstract

Tithonia diversifolia (Hemsley) A.Gray is a kind of plant and traditionally used to cure various diseases. The previous research found that T.diversifolia had antiproliferation effect on colon cancer cell (Col2). Sample preparation using biossay guided extraction and partition method, could simplify the active compounds of the extract. Research on the selectivity of purified extract from the leaves of Tithonia diversifolia (Hemsley) A.Gray) against HeLa cells hasn’t been known yet. The aim of this research was to find out the selectivity of purified extract of T.diversifolia’s leaves on HeLa cell compared to vero cell and to find out the Fifty Percent Inhibition Concentration (IC50), and its selectivity index. Extraction of T. diversifolia‘s leaves were done by maceration method using chloroform and methanol. Both of extract were tested by MTT cytotoxic assay on HeLa cells in vitro with serial doses (0,24 - 500μg/mL). Each group was replicated 3 times. The absorbance was read using ELISA at λ 540nm. IC50 was analyzed by probit regression on SPSS 15 for Windows. Partition (purified extract) of the active extract was done using Petroleum eter (PE), and tested by MTT cytotoxic assay on HeLa cells in vitro.  The IC50 methanol extract to HeLa cell in vitro is 1006,99μg/mL, IC50 chloroform extract is 16,61μg/mL.  The IC50 of soluble PE is 325,33μg/mL.  and IC50 of insoluble PE is 3,078μg/mL. The IC50 value of insoluble PE to Vero cell is 80,30 μg/mL. Selectivity index of purified extract (PE insoluble extract) is 26.09.

Abstract

Aktivitas sitotoksik ekstrak larut aseton batang kayu MezzetiaparvifloraBecc. dievaluasi pada sel HeLa dan dibandingkan dengan sel normal vero untuk memastikan penggunaan sebagai tanaman obat tradisional untuk pengobatan tumor. Percobaan dengan sel normal vero menggunakan uji MTT menunjukkan persentase viabilitas sel sebanyak 96,8% pada konsentrasi 1000 ug dan tidak meningkat dengan meningkatnya konsentrasi ekstrak. Sedangkan pada percobaan dengan sel HeLa menunjukkan aktivitas sitotoksik yang rendah dengan persentase viabilitas sebesar 87,4% pada konsentrasi 1000μg/ml. Oleh karena itu ekstrak ini dikategorikan sebagai tidak beracun dan diperlukan studi selanjutnya untuk mengeksplorasi mekanisme yang bertanggung jawabterhadap efek antikanker dari tanaman tersebut.

Abstract

Angiogenesis merupakan peristiwa pertumbuhan pembuluh darah baru, yang memungkinkan sel kanker mendapatkan suplai nutrisi dan oksigen, sehingga dapat terus bertahan hidup. Pasak bumi (Eurycoma longifolia, Jack) merupakan salah satu tanaman asli Indonesia yang memiliki potensi anti kanker. Ekstrak metanol, butanol, kloroform, dan air dari akar pasak bumi terbukti memiliki efek sitotoksik terhadap beberapa sel kanker. Penelitian ini bertujuan untuk mengetahui efek antiangiogenik ekstrak metanol akar pasak bumi pada CAM embrio ayam. Uji penghambatan angiogenesis dilakukan dengan membagi telur berembrio umur 8-9 hari dalam 8 kelompok perlakuan. Kelompok I sebagai kontrol paper disc, kelompok II sebagai kontrol bFGF, kelompok III sebagai kontrol bFGF+ pelarut DMSO 0,8%, kelompok IV, V, VI, VII dan VIII sebagai kelompok uji penghambatan angiogenesis, pada paper disc diberi bFGF 1 ng/μL dan ekstrak metanol akar pasak bumi berturut-turut dengan dosis 20, 40, 60, 80 dan 100 μg/mL. Setelah diinkubasi selama 3 hari (umur embrio12 hari), telur dibuka dan isi telur dikeluarkan, kemudian CAM yang melekat pada cangkang diamati secara makroskopik dan mikroskopik. Ekstrak metanol akar pasak bumi memberikan aktivitas antiangiogenik mulai kadar 40 μg/mL. Peningkatan konsentrasi ekstrak metanol akar pasak bumi meningkatkan aktivitas penghambatan angiogenesis.

Abstract

Background : Traditionally some benalu species were used to treat tumour in Indonesia. Some research report indicated the present of anti tumour activity, although its toxicity study by BST was not performed yet. It is necessary to observe the toxic level of the benalu leaves and whether they are safe to be consumed. Objectives : To determine the toxic level of species of benalu leaves (Dendrophtoe pentandra L. Miq. (1), Macrosolen tetragonus (bL) Miq. (2 and 7), Helixanthera parasitica Lour. (3), Dendrophtoe falcate (Lf.) Ettings (4), Dendrophtoe constricta Dans. (5), Macrosolen cochinchinensis (Lour). Tiegh. (6))Methods : The leaves of benalu were separately extracted with chloroform, followed by methanol. The chloform and methanol extracts toxicity level were screened using Brine Shrimp Lethality Test (BST) at the dose of 1500, 1000 and 500 ug/mI. The level of the toxicity was determined by counting the death percentage of the Artemia saline larvae after 24 hours of adding the extract at the dose indicated. The extract was considered toxic when 100% A. saline larvae was killed at the dose < 1000 ug/ml, and the extract was mildl toxic when 100% death percentage was observed at the dose of 1500 ug/ml.Results : Practically all the chloroform extracts were non toxic since they were not able to kill 100% A. saline larvae at the dose of 1000 ug/mI. Although when the doses were raised to 1500 ug/mI, the 100% death percentage was still not obtained. Similarly, the methanol extracts were also non toxic at the dose of 1000 ug/ml. However, M. cochinchinensis (6) and M. tetragonus (7) were considered to be mild) toxic as their methanol extracts were able to kill 100% A. saline larvae when the dose was raised to 1500 ug/mI.Conclusion: The different test result (at 1500 ug/mI) shown by two methanol extracts of similar species M. tetragonus {2b (killed 52%) & 7b (100%)} should give an idea that the benalu are practically non toxic and save to be consumed for tumor treatment.Key words : toxicity screening, brine shrimp lethality test (bst) - benalu leaves - benalu hosts - anti tumor.

Abstract

Mahkota Dewa [Phaleria macrocarpa (Scheff). Boerl.] is used traditionally to treat cancers in Indonesia. Extract methanol of the leaves displayed a small LC50 value (63.16 μg/ml) on BST (Brine Shrimp Lethality test) assay; therefore a phytochemical study of this extract was undertaken. A new benzophenoic glucoside was isolated from the methanol extract and identified as 4,5-dihydroxy,4’-methoxybenzophenone-3-0-β-D-glucoside (Phalerin) based on its spectroscopic data. Phalerin was cytotoxic to myeloma cell line (NS-1) (in vitro) having IC50 of 83 μg/ml or 1.9 x 10-1 mM.Key words: Phaleria macrocarpa, Phalerin, myeloma

Abstract

Kembang bulan [Tithonia diversifolia (Hemsley) A. Gray] merupakan kandidat tanaman berpotensi antikanker yang telah dibuktikan sebelumnya memiliki efek sitotoksik terhadap sel kanker WiDr, IC50=61,55 ug/mL. Kunyit (Curcuma domestica Val.) banyak digunakan dalam pengobatan tradisional, mengandung kurkuminoid yang memiliki aktivitas antikanker dengan memodulasi fungsi imun yang dimediasi oleh limfosit. Dengan adanya efek imunostimulan dari kunyit apabila dicampur dengan kembang bulan sebagai kandidat antikanker diharapkan akan bersinergis lebih baik. Tujuan penelitian ini untuk mengkaji potensi kombinasi ekstrak etanol T. diversifolia dan C. domestica pada hewan model kanker dengan cara menghitung berat badan, jumlah nodul dan volume nodul hewan coba. Penelitian ini adalah penelitian kuasi eksperimental. Kombinasi ekstrak etanol 70% daun kembang bulan dan rimpang kunyit dibuat suspensi dan dibagi menjadi 3 dosis yaitu dosis 40:150mg, 80:150mg, dan 160:150mg. Kemudian diberikan peroral pada masing-masing kelompok tikus yang telah mengalami kanker payudara setelah diinduksi dengan DMBA. Kemudian diamati berat badan, jumlah nodul, dan volume nodul tikus yang dibandingkan dengan kelompok kontrol negatif dan kelompok kontrol positif (doksorubisin). Persentase perubahan pada masing-masing kelompok perlakuan dibandingkan dan diuji secara statistik menggunakan analisis Anova. Hasil menunjukkan bahwa perbandingan berat badan tikus antara kelompok kontrol doksorubisin dan kelompok dosis 40:150mg, 80:150mg, dan 160:150mg menunjukkan tidak berbeda signifikan. Perbandingan volume nodul kanker payudara antara kelompok kontrol dan kelompok uji menunjukkan hasil yang tidak berbeda signifikan. Kombinasi ekstrak etanol  kembang bulan dan rimpang kunyit tidak mempunyai efek signifikan terhadap berat badan dan nodul kanker payudara

Abstract

BACKGROUNDThe leaves of Nerium indicum Mill. have been utilized traditionally to cure cancer. By using Bioassay guided extraction and isolation method, three compounds (NiO-1, NiO-2, NiO-3) were isolated from an active fraction of the CHCl3 extract of N. indicum leaves. NiO-2 identified as 5α- oleandrin is the best cytotoxic compound on HeLa cervical cancer cell in vitro among the other two. However mechanism of action of the compound hasn’t been evaluated yet. The aims of this study were to determine the mechanism of action at molecular level.METHODSThe action mechanism of 5α-oleandrin on HeLa cervical cancer cells was analyzed by staining the cells with Hoechst 33342; the agarose gel electrophoresis was aimed to determine the DNA fragmentation and the western blotting was aimed to determine the Bcl-2 and Bax protein expression.RESULTSIncubation of HeLa cervical cancer cell with 5α-oleandrin at the concentration 3,47x10-4 mM (24 hours) followed by staining with Hoechst 33342, a broken up light blue color of nucleus was observed (compared with intensive color of untreated control). By gel electrophoresis (at the same concentration of the tested compound), a smear band at about 200 bp was observed. In addition, cells treated with 5α-oleandrin displayed a decreasing of the Bcl-2 protein expression and increasing of the Bax protein expressionCONCLUSION5α-oleandrin induced HeLa cervical cancer cells DNA fragmentation observed by the presence of a smear band at about 200 bp indicative the apoptotic occurrence. 5α-oleandrin induces apoptosis by reducing the Bcl-2 protein expression but the Bax protein expression increases.
