Abstract

Twenty growing cross breed etawah goat were treated with clenbuterol every two days. Three levels ofclenbuterol , 5, 10 and 20 ?g/kg BW were injected intramuscularly from the thigh side. The experimentwould like to see the effect of clenbuterol on the metabolism and carcase quality. It was found thatclenbuterol did not affect the gross energy, digestibility, metaboliseable energy and urinary nitrogenexcretion.Clenbuterol will increase the feed intake up to 51.49%. Althoush not affecting protein intake, itsignificantly increased the retain protein from 70.51 up to 149.37%. Subcutaneous carcase weights werereduced between 29.6 up to 51%. Data from carcase quality showed that clenbuterol increase half emptybody weight indicating that clenbuterol is affecting protein metabolism. Dose -respond curve of clenbuterolagainst half empty body weight follows the Michaelis Menten equation and solved by Lineweaver-Burks.Result indicated that the half weight maximum was 5120 gram and Km of clemnbuterol was 9.50 x 10-8M. Clenbuterol is affecting the bone weight carcase but not affecting the intermusculer carcase.

Abstract

A study has been carried out to map the distribution pattern of poultry from traditional market toreduce the transmission risk of avian influenza virus. The data were collected from threes markets wherepoultry are sold, namely in Bringkit of Badung Regency, Kumbasari of Denpasar City, and Kediri ofTabanan Regency. Data collections was based on interviews using questionnaire. Poultry from all marketsare distributed throughout Bali. Poultry are traded mainly for religious ceremony and immediatelyslaughtered as it arrives at the consumer’s house. The distribution pattern of poultry seems to play asignificant role in the disseminations of avian influenza virus. The right implementation of biosecurity intraditional markets is highly recommended to curb the risk.

Abstract

As chromosome of different species differs in the size, shape, and number, their caryotpes areexpected to provide a distinctive and characteristic feature of the organism. The caryotypes ofseven amphibian species : Limnonectes gruniens, L modestus, Rana (Hylarana) chalconota,Fajervarya cancrivora, Polypedates celebensis, Bufo celebensis and B biporcatus of South EastSulawesi-origin were investigated. The result showed that the number of diploid chromosomes inthe seven anuran varied between 22-26. The highest diploid chromosomes were observed in Rchalconota, F cancrivora (26), followed by L gruniens, L modestus (24) and the lowest were observed22 in P celebensis, B celebensis and B biporcatus (22). In general, all the the seven anuran sharesfour metacentric chromosomes pairs which is in pairs No 5, 6, 9, and 10 respectively. Howeverdifferences were observed in other chromosomes pairs numbers among the seven species.

Abstract

It has previously been demonstrated that a G/A Single Nucleotide Polymorphism (SNP) at nucleotideposition 1,892 of coding sequence of chicken Mx gene confers susceptibility/resistance to avian viral diseases.The aim of this study was to assess the geographical distribution of G/A alleles in relation to differentgenetic backgrounds of a wide range of chicken populations. Using Polymerase Chain Reaction- RestrictionFragment Length Polymorphism (PCR-RFLP) methods, 492 samples from 15 breeds of indigenous chickenpopulations from Java, Sumatera, Kalimantan and Sulawesi islands were genotyped. Allele and genotypefrequencies of each population were calculated. Deviations from Hardy-Weinberg equilibrium were testedand inbreeding coefficient FIS estimated. Overall, the susceptible allele G had a frequency of 37.27% whilethe resistant allele A had a corresponding frequency of 62.73%. No clear relation of the geographicaldistribution of the G/A alleles to genetic backgrounds was found. The distribution of this SNP acrosspopulations seems to be affected by genetic drift rather than selection.

Abstract

Bali cattle are one of Indonesia national asset which need to be conserved as they have many advantages.They are however susceptible to many infections diseases such as jembrana disease. Currently, thedisease is prevented by vaccination using vaccine derived from jembrana disease virus (JDV)-infected balicattle. An alternative vaccine using JDV-infected lymphocyte culture is expected to increase the virusyield and is therefore likely to increase the antibody response in the vaccinated animals. A study thereforewas therefore conducted to compare the anti-capsid antibody response of Balb/c mice immunized withvaccine derived from the spleen of infected cattle (spleen vaccine) and those immunized with vaccinederived from infected lymphocyte culture (culture vaccine). As many as 16 female Balb/c mice weredivided into two groups, Each group was vaccinated 4 times weekly respectively with spleen and culturevaccines. The antibody response against the capsid protein of JDV was determined using enzyme-linkedimmunosorbent assay (ELISA) and the absorbance reading of mice sera from each group was compared. Tstudentand univariate analysis showed that the average absorbance reading sera of sera derived frommice vaccinated with spleen vaccine (0.15) was not significantly different from those vaccinated withculture vaccine (0.18). It appears that culture vaccine is able to induce anti-capsid antibody response ashigh as spleen vaccine.

Abstract

The effectiveness of homeopathy combination in eliciting nonspecific cellular defense mechanism indairy cows is demonstrated in this study. Two groups of 2-7 years old, in 3-5 months term of pregnancydairy cows which suffering clinical mastitis were used id this study. At 3 weeks and 4 weeks prae partusanimals received their first homeopathy treatment, i.e. Group I: Coenzyme comp®, and Group II: a combinationof Mucosa comp® and Traumeel®, respectively. At one week and two weeks post partus all animals in thetwo groups received their second treatment, i.e. a combination of Lachesis comp® and Traumeel®.Subsequently, at 3 and 4 weeks post partus all animals in the two groups received their third treatment,i.e. a combination of Carduus comp® and Coenzyme comp®, respectively. All homeopathy solutions weregiven at 5 ml subcutaneously. Ten ml of blood samples for haptoglobin analysis were drawn from eachanimal and blood smears for White-Blood-Cell counting and differentiation and phagocytosis activity andcapacity observation were prepared from each animal at 3 and 4 weeks prae partus; and at one week, 3, 5,and 7 weeks post partus, respectively. Our results indicated that although there were non significantdifference in the phagocytosis activity and capacity between animals in Group I and Group II; animals inGroup II showed a marked increase in phagocytosis activity and capacity. Based on haptoglobin analysis,animals in Group II showed a steadily low haptoglobin level in the blood (0.08 mg/ml) up to 3 weeks postpartus and a tendency to further decline until the end of the observation. Based on the WBC counts anddifferentiation, animals in Group II also showed a better response compared to animals in Group I.In conclusion, the use of homeopathy combination is capable in eliciting nonspecific cellular defensemechanism; this phenomena were clearly shown in the combination of Mucosa comp® and Traumeel®. Theuse of this homeopathy combination should be considered at prae partus allowing the recognition of thehost defense mechanism.

Abstract

Cryoprotectant is one of determining factors for the success of stallion semen cryopreservation. Formost mammalian sperm cryopreservation,glycerol has been widely used as the cryoprotectant. Glycerol inhigh concentration is toxic to the sperm but in low concentration it has protective effect on the semen. Theobjectives of this experiment were to find appropiate glycerol concentration by using skim milk extender inorder to maintain motility and viability of spermatozoa of stallion frozen semen. Semen was collectedfrom three sexually mature stallions by using artificial vagina. The semen characteristics and qualitywere examined both macro and microscopically. Prior to extension, semen was centrifugated at 3000 rpmfor 15 minutes. The pellet was re-extended with skim milk extender with glycerol 5% (G5) 7.5% (G7.5), and10% (G10). The extended semen as then packed in mini straw (0.3ml), equilibrated at 5 oC for 2 hours,frozen in the liquid N2 vapor for15 minutes and then stored in liquid N2 container until futher evaluation.The frozen semen, was thawed at 37 oC for 30 seconds. The percentages of sperm motility and viabilitywere examihed. The result of this research showed that the sperm motility and viability in G5 was (23.8 %;35.6 %) significantly hihger (P<0.05) as compare a with G7.5 (18.1%; 28.6%) and G10. (11.8%; 23.1%). Therecovery Rate in G5 (35.4%) was significant high er (P<0.05) than G7.5 (26.9%) and G10 (17.6%). In conclusion,glicerol of 5% is recomended for cryopreservation of stallion semen extended with skim milk.

Abstract

Fenugreek seed (Trigonella foenum-graecum) contains saponin diosgenin, wich has an antifertility effect on spermatozoa so it can be used as an oral contraceptive drug. This study was aimedto investigate the effect of fenugreek seed extract to spermatogenic process of rabbit, especially onviability spermatozoa. “Completely randomized control group post-test only design” was used inthis study. The animals were divided into four groups; one control group and three treatmentgroups with six replicates (P0 = control group; P1 = group were given 10 % fenugreek seed extract,1 cc/day; P2 = group were given 20 % fenugreek seed extract, 1 cc/day; P3 = group were given 30 %fenugreek seed extract, 1 cc/day). The extract was given orally once a day in 50 days. After treatment,testicles were sectioned and stained with Hematoxylin Eosin; for qualitative and quantitativemicroscopic analysis. The result of this study showed that the number of spermatozoa were decreasedsignificantly (p<0,05) after receiving 10% fenugreek seed extract 1 cc per day. In conclusion,fenugreek seed extract could reduce the number of spermatozoa.
