Abstract

The activation of orthodontic appliance will generate mechanical force. This forceis used to depress tooth and the tissues surrounding it then stimulates alveolarbone remodeling. Alveolar bone remodeling is divided into compression andtension area. This stress becomes a signal to activate heat shock responseyielding heat shock protein (HSP) synthesis, especiallyhsp25isessentialforalveolarboneremodelingprocessitself. The aim of this study was to comparehsp25 expression in compression and tension areas. This study using guinea pigsmandibular first insicivus that given a mechanical force to see hsp25 expressionin both areas. The HSP25 expressionwas measured by counting this protein afterbeing conducted by immunohystochemistry method. It is analysed statisticallywith one way Anova with level of significant p = 0,05.

Abstract

The prevalence of oral diseases is increasing and caused by many factors. It can be caused by environmental, genetic, malignancy or side effects of dental treatment. The research detect these disease has been done, but still not getting satisfactory results. Heat shock proteins (HSP) are intracellular proteins that mediate the cytoprotective function and other essential functions. This protein is expressed and synthesized under conditions of stress/harmful or nonstress. The oral disease is a form of adverse conditions, and the course will also trigger HSP expression.

Abstract

The usage of orthodontic appliance is to correct malocclusion involves alveolar bone remodeling. This process isstimulated by mechanical force which obtained from the activation orthodontic appliance. The force was used to depressteeth and the tissues surrounding it. This stress will become a signal to activate heat shock response yielding HSPsynthesis. HSP25 plays a part in osteoclastic bone resorption and osteoblastic new bone forming. The aim of this studywas to explore the change of osteoclast and osteoblast HSP25 expression in the different mechanical force induction atpressure and strain area. The subject was guinea pig (Cavia sp), divided into 2 groups. Control group did not useorthodontic appliance and treatment group was given mechanical force as 90 g, 120 g, and 150 g. Time usage oforthodontic appliance was 5 and 10 days. Osteoclast and osteoblast HSP25 were measured by counting cells after beingconducted by immunohistochemistry method. The result showed that osteoclast HSP25 expression at pressure area on allgroups had increased value compared to strain area and was different statistically. On the other hand, osteoblast HSP25expression at strain area had increased value was compared to at pressure area and especially in group of 120 g and 10days had difference statistically. In conclusion, different mechanical force induction could influence osteoclast andosteoblast HSP25 expression. HSP25 may represent one of the factors that influences osteoclast and osteoblast cellularactivity and coupling process between osteoclast and osteoblast.

Abstract

This study was an experimental laboratory. The purpose of this study was to determine the role of probiotics in inhibitingthe activity of pathogenic bacterial oral cavity. The result of study is expected to be used for prevention and treatment ofperiodontal disease which is still complex and derived products with high quality probiotics. The main product is thedevelopment potential of probiotics in maintaining oral health, especially periodontal disease is effectively and efficiently.This study used rats and divided into 4 groups: Group I, control without treatment; group II, induction of LPS E. coli for 5days, Group III, LPS induction of E. coli + Probiotic Bacterial Lactobacillus casei injection for 5 days together, and groupIV, induction of LPS E. coli for 5 days + 5 days injection of Lactobacillus casei. Immunohistochemical examination wasdone to evaluate the activity of osteoclasts in alveolar bone resorption to detect TRAP (tartarate-resistant acidphosphatase) and Carboxyterminal Telopeptide of Type 1 Collagen (1CTP) and subsequently analyzed the data collectedstatistically. The results of this study showed that induction of LPS could trigger the occurrence of alveolar boneresorption mediated by osteoclast and Lactobacillus casei administration both at the same time with the LPS induction orafter LPS induction can inhibit osteoclastic alveolar bone resorption. In conclusion, clinically it can be interpreted that theadministration of probiotic can be given at the beginning of periodontal disease occurs before or at the time of periodontaldisease is underway.

Abstract

ABSTRAK Pendahuluan: Pasca perawatan ekstraksi gigi ini tentu akan menimbulkan luka. Luka pasca ekstraksi gigi secara fisiologis akan mengalami proses penyembuhan yang terdiri atas penyembuhan jaringan lunak dan jaringan keras secara bersamaan. Angiogenesis memiliki peran yang penting dalam kesiapan jaringan untuk mensuplai nutrisi. Perawatan pasca ekstraksi gigi memerlukan metode untuk mempercepat penyembuhan. Salah satunya dapat menggunakan kombinasi scaffold hidroksiapatit gipsum puger (HAGP) dengan aloe vera. Tujuan penelitian menganalisis potensi kombinasi Scaffold gipsum puger dengan lidah buaya (aloe vera) terhadap angiogenesis pada soket gigi pasca ekstraksi gigi tikus wistar jantan. Metode: Jenis penelitian eksperimental laboratorium 32 ekor tikus dibagi menjadi 4 kelompok: Kelompok diberlakukan ekstraksi gigi (K), kelompok diberlakukan ekstraksi gigi dan diberikan scaffold aloe vera (PI), kelompok diberlakukan ekstraksi gigi dan diberikan scaffold HAGP (PII), serta kelompok ekstraksi gigi dan diberikan scaffold HAGP dan aloe vera (PIII). Ekstraksi gigi dilakukan pada gigi M1 rahang bawah kiri. Tikus di euthanasia sesuai dengan masing – masing kelompok. Data dianalisis menggunakan uji Anova Least Significant Difference (LSD). Hasil: Kombinasi scaffold HAGP dan aloe vera berhasil meningkatkan angiogenesis pada hari ke-7, tetapi gagal menurunkan atau menstabilisasi angiogenesis pada hari ke-14 pada kelompok perlakuan. Hasil uji One Way Anova dan LSD menunjukkan perbedaan signifikan jumlah fibroblas pada seluruh kelompok sampel (p<0,005). Simpulan: Potensi kombinasi scaffold gipsum puger (HAGP) dan lidah buaya (aloe vera) kurang efektif dalam proses angiogenesis pada hari ke 14 pasca ekstraksi gigi tikus Wistar jantan.Kata kunci: angiogenesis; ekstraksi gigi; scaffold; hidroksiapatit gipsum puger; aloe vera ABSTRACTIntroduction: Tooth extraction will certainly cause wounds. Post-extraction wounds will physiologically undergo a healing process consisting of soft tissue and hard tissue healing simultaneously. Angiogenesis has an essential role in the readiness of tissues to supply nutrients. Therefore, treatment after tooth extraction requires healing acceleration. One of them uses the combination of hydroxyapatite gypsum puger (HAGP) and aloe vera scaffold. The study aimed to analyse the potential of the combination of hydroxyapatite gypsum puger and aloe vera scaffold on angiogenesis of the post-extraction socket of male Wistar rats. Methods: In this experimental laboratory research, 32 rats were divided into four groups: Group with tooth extraction (K) only; Group with tooth extraction, and administered with aloe vera scaffold (PI); Group with tooth extraction, and administered with HAGP scaffold (PII); Group with tooth extraction, and administered with a combination of hydroxyapatite gypsum puger and aloe vera scaffold. Tooth extraction was performed on the left mandibular M1. Rats were euthanised according to each group determined by days. The data were analysed using the ANOVA and Least Significant Difference (LSD) tests. Results: The combination of HAGP and aloe vera scaffold increased angiogenesis on the seventh day. However, it failed to reduce or stabilise angiogenesis on the fourteenth day in the treatment group. The one-way ANOVA and LSD tests showed significant differences in the number of fibroblasts in all sample groups (p<0.005). Conclusions: The potential of the combination of hydroxyapatite gypsum puger and aloe vera scaffold is less effective in the angiogenesis process on the fourteenth-day post-extraction of male Wistar rats.Keywords: angiogenesis, tooth extraction; scaffold; hydroxyapatite gypsum puger; aloe vera

Abstract

Neutrophils were part of leucocyte cells which dominate in blood circulation. When the microorganism entered the body, neutrophils kill it immediately. The steps that were migration, ingestion, swallowing, and microbicidal activity or intercellular killing. The contents of mangosteen peel is presumed to help neutrophils beat bacteria. Samples numbered 28 consist of 7 treatment groups, namely K 1 (negative control), K II (positive control), K III 100% MPI (Mangosteen Peel Infution), K IV 75% MPI, K V 50% MPI, K VI 25% MPI, and K VII 100% MPIwithout neutrophils. Isolated neutrophils exposed to MPI appropriate treatments groups above and then incubated for 3 hours. Subsequently exposed to S. mutans and incubated again for 3 hours. After that, the sample werecultured in BHI-A media and incubated for 1x24 hours. Furthermore, bacterial colonies were counted with a colony counter method. The result showed a significant difference between MPI treatment groups with control groups. The conclusion of this research is thenatural mangosteen peel infution hasthe potential to increase microbicidal activity of neutrophils and can act as an antibacterial.   Keywords: mangosteen (Garcinia mangostana L.),microbicidal,neutrophils, S. mutans.

Abstract

Background. Porphyromonas gingivalis (P. gingivalis) is plaque bacteria caused periodontitis with proteolitic characterised that makes some damages to the periodontal tissues. Lymphocytes are chronic inflamed cells which have specific characteristic when periodontitis was occured. Papaya is known as herbal plants, which contains substances which have antimicroba and antiinflamation activities in its leaves. The purpose of this research is to determinate the effect of papaya leaves extract to the number of limfocyte cells to the male-wistar rats gingival that was periodontitis. Twenty male-wistar rats that were divided to the five groups with different treatments. The first group is control of other group, the second group is treatment group that was induced with P. gingivalis and assembled with a ligature, while the remaining of groups were added with the extract of papaya leaves sondation by certain concentration 25% for the third group, 50% for the fourth group, and 75% for the last group after getting some treatments as the second group. The result of the research showed that the highest lymphocytes cells was found to group that was induced by P. gingivalis and assembled with a ligature. In the contrary, the lowest lymphocytes cells was found to the fifth group that was treated with the extract of papaya leaves 75%. It concluded that papaya leaves extract decreased the number of lymphocytes in periodontitis male-wistar rats gingival. The most effectively of papaya leaves extract that was used is 75%. Advanced research about the compound of the extract of papaya leaves which most influential and the mechanism to the decrease of the number of lymphocytes cells are needed. Abstract

Perimenopause is trantition phase of nomal ovulation cicle to menopause that signed decreasingestrogen hormone production. In this phase, women will be physiology and psychology alteration related fluctuationof estrogen hormone. The estrogen level alteration is one of cause reducing of bone density in perimenopausewomen. When estrogen level decreases, it will induce osteoclast activity and finally cause bone resorption. In boneresorption osteoclast secretes protease that can dissolve collagen which is in organic matrix and free bone mineral.Mature collagen is bound by pyridinium crosslink that it will be degraded in bone resorption process, such asdeoxypyridinoline. Deoxypyridinoline have suggested as collagen degradtion marker of bone resorption in serumand urine. This study was objective to identify deoxypyridinoline in saliva of perimenopause women. The subjects ofthis study were perimenopause women who were 40-55 years old. The subjects had to fulfill informed consent and beundergone intra oral examination, including caries and periodontal index index. After that the subjects collectedtheir whole saliva. Deoxypyridinoline in saliva was identified by Liquid chromatography Mass Spectroscopy. The resultshowed that deoxypyridinoline level of perimenopause women was about 23.802 ppm. This level was higher thanproductive women. The conclusion was deoxypyridinoline level in saliva of permenopause women
