Abstract

Rattan manau (Calamus manan Miq.) is one of the many commercial rattan species grown in the forests of East Kalimantan. The fruit can be eaten as well as the fruits of some other types of rattan, but it is also traditionally used by locals as stomach ache or toothache. Until now there has been no research that investigates the benefits of fruit for medicinal purposes or other benefits. This study aims to determine the bioactivity potential contained in the fruit of Calamus manan Miq. including phytochemical content, antioxidant properties, toxicity properties, and antibacterial activity. Parts of fruit, pericarp, flesh and seed are tested using ethanol as a solvent. The results of study showed that the pericarp and seed have higher antioxidant activity than fruit flesh where the seed have IC50 value 25,04 ppm, pericarp 41,17 ppm and fruit flesh 170,19 ppm; based on toxicity properties using BSLT (Brine Shrimp Lethality Test) test of rattan manau fruit is not toxic; while antibacterial activity against two types of bacteria showed pericarp and seeds are antibacterial activity against to Escherichia coli with strong category, while antibacterial activity against Streptococcus mutans is weak category. Keywords: antioxidant, toxicity, antibacterial, rattan, calamus.

Abstract

The study aims to identify the secondary metabolite compounds contained in the ethanol extract Propolis Tetragonula Iridipennis and determine the antioxidant activity. Propolis is extracted from the beehive of Kelulut (Tetragonula Iridipennis) with a method of maceration of ethanol 96% produce a yield of 66.73%. Propolis extract contains alkaloid compounds, flavonoids, triterpenoids, tannins and carbohydrates. Propolis extract tested its antioxidant activity using the DPPH method (1.1-diphenyl-2-picrylhydrazyl) with a wavelength of 514 nm. Results showed a percentage of propolis extract free radicals 59% in concentrations of 100 ppm with IC50 33.74 ppm so that propolis extract is potentially as an antioxidant.

Abstract

Pemanfaatan produk propolis dan bee pollen madu tanpa sengat atau Kelulut masih sangat terbatas, demikian juga penelitiannya belum banyak dilakukan bahkan bisa dikatakan masih sangat terbatas termasuk kandungan zat ekstraktif pada bee pollen tersebut. Oleh karena itu tujuan penelitian ini adalah untuk skirining manfaat bee pollen sebagai produk kecantikan dan produk yang mengandung bahan yang dapat menangkal radikal bebas. Kemajuan teknologi dan gaya hidup manusia dimasa sekarang cenderung mudah menimbulkan banyak penyakit baru. Hal ini menimbulkan rasa penasaran akan bee pollen sebagai produk dari alam yang memiliki kemampuan sebagai antioksidan dan menghambat pertumbuhan Propionibacterium acnes. Metode ekstraksi yang digunakan dalam penelitian ini adalah ekstraksi dingin etanol 98% selama 3 x 24 jam, dilanjutkan dengan uji fitokimia, dan analisis antioksidan dengan uji DPPH dan penghambatan Propionibacterium acnes dengan metode difusi agar. Ekstrak bee pollen T. sarawakensis mengandung senyawa alkaloid, flavonoid, kumarin dan tanin. Ekstrak T. sarawakensis memiliki aktivitas antioksidan dengan nilai konsentrasi hambatan 39% pada 100 mg/mL. Ekstrak memiliki penghambatan terkuat dari Propionibacterium acnes (42% pada 500 µg/well). Hasil ini menunjukkan adanya potensi ekstrak etanol T. sarawakensis untuk dikembangkan sebagai bahan kosmetik dan jamu, namun diperlukan percobaan lebih lanjut untuk membuktikan fungsinya.

In our efforts to find new whitening agent materials, we focused on wood of Artocarpus heterophyllus which is anti melanogenesis. By activity-guided fractionation of A. heterophyllus wood extract, norartocarpetin and artocarpesin were isolated which inhibited both mushroom tyrosinase activity and melanin formation in B16 melanoma cells. This compound is a strong candidate as a remedy for hyperpigmentation in human skin that can be used for cosmetic (whitening agent).

Abstract

Oyster mushroom (Pleurotus ostreatus) is not a new food for the community. Long time ago oyster mushrooms is food but not so well known. Now oyster mushrooms have been accepted by the community as a healthy food. This is the background of this research to produce mushrooms not only as nutritious food but also natural medicine. The purpose of this study was to examine the antimicrobial potential of oyster mushrooms by determination of clear zone against Candida albicans and Propionibacterium acnes bacterias. This research was conducted at Forest Products Chemistry Laboratory, Faculty of Forestry, Mulawarman University. The resources of oyster mushroom we used in this study comes from the cultivation of the entrepreanure forestry student group in Mulawarman University. This study used successful extraction with hexane solvent, ethyl acetate, ethanol, water, and crude ethanol. The results obtained that the highest antimicrobial inhibition against Candida albicans bacteria with 47.60% with a concentration of 100 ppm but, on the antimicrobial against Propionibacterium acnes bacteria did not show any significant inhibition.

Abstract

Indonesia merupakan negara hutan tropis yang luas, maka tak heran negara ini menjadi negara megabiodiversitas. Jamur tiram merupakan salah satu hasil kekayaan alam Indonesia yang memiliki potensi sebagai tanaman kesehatan dan obat. penelitian ini bertujuan untuk memunculkan potensi aktivitas antioksidan pada jamur tiram yang banyak dibudidaya di Kalimantan Timur khususnya Samarinda. Sampel kami ambil dari rumah jamur borneo dan penelitian ini dilaksanakan di Laboratorium Kimia Hasil Hutan, Fakultas Kehutanan, Universitas Mulawarman. Metode ekstraksi bertingkat menggunakan n-heksan, etil asetat, dan etanol. Metode antioksidan yang digunakan adalah penangkal radikal bebas (DPPH) dengan masing-masing tiga kali pengulangan. Hasil penelitian ini menunjukkan bahwa aktivitas antioksidan tertinggi pada ekstrak air dengan nilai penghambatan 25% pada kosentrasi 100 ppm. Pada ekstrak heksan merupakan hambatan terkecil dengan nilai penghambatan 2,41% pada kosentrasi 25ppm.

Abstract

Calamus ornatus Bl., locally known as rotan mantang or rotan seuti is one of the commercial rattan species growing in the forests of East Borneo. Its fruit is edible, like the fruits of other species of rattan, and it has been used by the local people as traditional medicine to treat stomachache or toothache. Until recently, there has not been any study investigating the benefits of the fruit for medicinal or other purposes. This research aimed to discover the bio-activity potentials of Calamus ornatus Bl. fruits by identifying its toxicity level and antibacterial potentials. The parts of the fruit (pericarp, flesh, and seed) were extracted using ethanol as solvent to extract samples. The toxicity was analyzed by using the Brine Shrimp Lethality Test (BSLT) method while the antibacterial activity was tested using a well diffusion method. Low toxicity was found in the flesh, whereas the pericarp and the seed were non-toxic. Antibacterial activity against Escherichia coli was found in the pericarp and the seed, while antibacterial activity against Streptococcus mutans was found in the pericarp and the flesh.

Abstract

Phytochemical, brine shrimp lethality and antibacterial activity tests in each fraction from Andong leaves extract(Cordyline terminalis Kunth) that came fromUnmul Samarinda botanical garden, Kalimantan Timur has been carried out. The leaf was extracted with ethanol andconcentrated by using rotaryevaporator. The crudeethanol extract was fractioned with n-hexane, and ethyl acetate. Based on the secondary metabolites phytochemical test of the Cordyline terminalis Kunth leaves show that crudeethanol extract is contain phenol, saponin and steroid.n-hexane fraction iscontain steroid.Ethylacetate fraction is contain phenol and steroid. And ethanol-water fraction is contain phenol and saponin. Brine shrimp lethality test exhibit mortality rate of Artemiasalina (L) by Probit SAS analysis to determine lethal concentration 50% (LC 50 ) value. The test showed that the most active wascrudeethanol extract with LC 50 value of 26.8788 ppm. Antibacteria activity test of extracts for Bacillus cereus bacteria (positiveGram) andEscherichia coli (negativeGram) were carried out by paper disk method. The test showed that the most active were ethyl acetate extract with minimum inhibitor consentration of 2-10% which clear zone diameter was 6,4 mm on Bacillus cereus bacteria and 6 mm on Escherichia colibacteria.Keywords: Phytochemical, Antibacteria activity, LC 50

Abstract

BACKGROUND: Brucea javanica extract has been reported to have anti-proliferative and cell death induction activities. B. javanica extract was reported to induce apoptosis through caspase cascade. Most of investigated B. javanica extracts were derived from seeds and fruits, or commercially available oil emulsion. Therefore we conducted a study on B. javanica leaf extract (BJLE) in oral cancer cells.METHODS: B. javanica leaves were collected, identified, minced, dried, extracted with distilled ethanol at room temperature for 24 hours, filtered and evaporated. Resulted BJLE was stored at 4°C. Human oral squamous cell carcinoma (HSC)-2 cells were fasted for 12 hours and treated with BJLE in various concentrations for 24 hours. Cells were then quantified with 3-(4,5-dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium bromide (MTT) assay, demonstrated with 4',6'-diamidino-2-phenylindole (DAPI) staining. To find out mitochondrial membrane permeability (MMP), mitochondrial membrane potential (ΔΨM) was analyzed.RESULTS: BJLE reduced percentage of viable HSC-2 cells in a concentration dependent manner. BJLE induced apoptosis in HSC-2 cells. With treatment of 50 μg/ml BJLE, fragmented nuclei were seen. ΔΨM of HSC-2 cells treated with 50 μg/ml BJLE were shifted to the left, meaning that BJLE induced reduction of ΔΨM and attenuation of MMP.CONCLUSION: Our results suggested that BJLE could induce apoptosis by attenuating MMP.KEYWORDS: Brucea javanica, leaf, apoptosis, HSC-2, MTT, DAPI, mitochondria, permeability
