Abstract

Infectious diseases are estimated to be the cause of half of all deaths worldwide. About 50-75% of deaths in hospital are reported to be due to infection. This number continues to increase rapidly in line with the development of bacterial resistance to certain antibiotics. One of the causes of resistance due to antibiotic abuse. The number of resistance events that occur encourages new discoveries in order to overcome the problem of resistance. Actinomycetes are Gram-positive microscopic bacteria that are widespread in nature. These bacteria can live in various environmental conditions that contain lots of nutrients. Actinomycetes population increases in the presence of organic matter that undergoes decomposition. Acinomycetes have the ability to produce antimicrobial compounds. The need for antibiotic compounds and chemotherapy compounds that are effective with low toxicity to the host, and the resulting waste can be degraded by the environment, is a problem that needs serious attention. The existence of resistance to antibiotics causes the need to develop new types of antibiotics from natural ingredients and microorganisms that can control microbapatogens. Compounds obtained from natural materials, especially plants and microbes, provide promising results in the development of new antibiotic compounds, among the existing types of microorganisms, Actinomycetes are the most potential source of producing antibiotics. Apart from antibiotics, actinomycetes also produce bioactive compounds of high economic value in the health sector as antiviral and anti-cancer, while in agriculture as herbicides, insecticides, and anti-parasitic compounds. This causes the need to explore actinomycetes which have the potential to produce antibiotic compounds against bacteria. This study aims to determine the antibacterial activity of secondary metabolites of actinomycetes isolates against cuts infected with Staphylococcus aureus resistant methicillin (MRSA) in winstar rats (rattus norvegicus). This type of research is carried out in an experimental laboratory. The results showed that the potential for secondary metabolites of actinomysetes isolates against MRSA bacteria was classified as strong with an inhibitory index of 10.1.

Abstract

The degree of smoking habits in the urban community is still high, although carried out health promotion of harmful substances in cigarettes, one of which is cadmium (Cd). The dangers of cadmium in cigarette smoke causes harm to active and passive smokers. Increased exposure of cigarettes will lead to an increase in the burden of disease in groups at risk such as people with Diabetic Type 2 (DMT2). The purpose of this study was to determine the differences in levels of cadmium (Cd) in the blood of active smokers and passive smokers patients with DMT2 in Puskesmas Kedungdoro Surabaya. The type of this research is a comparative with cross-sectional study design conducted in the Toxicology Laboratory of the Department of Health Analyst and Research and Standardization Center of Industry in August 2020-May 2021. The independent variable in this study is active smokers and passive smokers patients with DMT2, and the dependent variable in this research is the content of cadmium (Cd) in the blood. Analysis of the levels of cadmium in the blood using Atomic Absorption Spectrophotometer by the method of acidifying carbon furnaces. The results showed range levels of cadmium in blood in 8 people suffer from Type 2 Diabetic as active smokers are 0.605-1.5535 µg/dL. While the range levels of cadmium in the blood of the 8 people who suffer from Type 2 Diabetic as passive smoking are 0.5648-2.7045 µg/dL. Based on the results of statistical tests using test Independent Sample T-Test, it can be concluded that there is no difference in the levels of cadmium (Cd) in active smokers and passive smokers patients with DMT2 in Puskesmas Kedungdoro Surabaya.

Abstract

Motor vehicles emit exhaust gas emissions that produce pollutants, one of the compounds contained is carbon monoxide (CO). CO may cause intoxication in humans if exposed for a long time especially street vendors because the work area is filled with vehicle fumes containing CO gas. High CO levels in the air indicate HbCO levels to increase. The increased of HbCO levels will cause abnormal hemoglobin levels and induce vasoconstriction which makes blood pressure increase. The purpose of this study was to prove the correlation between CO levels in the air with HbCO, hemoglobin, and blood pressure levels to street vendors in the Gading Fajar Sidoarjo Street Vendor Area. This research uses the Conway diffusion cell method and is an analytic observational study with a cross sectional design implemented at the Surabaya Health Laboratory Center and the Hematology Laboratory of Medical Laboratory Technology Department Poltekkes Kemenkes Surabaya in March 2022. Blood tests was carried out on 25 street vendors using random sampling technique. The results showed that average air CO level was 11.0 ppm and 13.3 ppm; mean HbCO levels 0.87%; mean hemoglobin levels 14.4 g/dL; mean systolic blood pressure levels 128 mmHg; and the mean of diastolic blood pressure levels 80 mmHg. The data were analyzed using SPSS. The outcome of normality test showed that the data was not normally distributed. Statistical tests was performed using Spearman correlation. The conclusion showed there was correlation between air CO level with HbCO level, hemoglobin level, and blood pressure level on street vendors in the Gading Fajar Sidoarjo Street Vendor Area. If CO levels in the air increase, then HbCO levels and blood pressure also increase. Street vendors are expected to always use Personal Protective Equipment (PPE) such as mouth masks when working to reduce the risk of exposure to CO gas and doing a healthy lifestyle as a preventive effort to avoid health problems.

Abstract

The prevalence of MRSA in various hospitals in the world ranges from 2-70% with an average of 20% .In recent decades there has been an increasing prevalence of S. aureus and MRSA in the world. Population-based studies in North America and Europe indicate the prevalence of S. aureus is between 18-30%. The overall prevalence of MRSA in Asia has reached 70%, while publications and prevalence of MRSA in Indonesia are still very limited and very difficult to obtain. The prevalence of MRSA in Atmajaya Hospital, Jakarta, reached 47%. Yuwono also reported in 2010 MRSA at Dr. RSUP. Moh. Hoesin Palembang reached 46%. Amplification were carried out under PCR conditions as a result of optimization, namely the results of PCR optimization were carried out for 30 cycles with the following thermal conditions. Denaturation phase at 94oC for 30 seconds, annealing at 52.0oC for 30 seconds, and extension at 72oC for 1 minute. Amplification results showed that of the 30 samples only 90% (27 samples) were detected to have the Mec-A gene, 10% (3 samples) were negative. Keywords: Mec-A; gene; Methicillin Resistant Staphylococcus aureus

Abstract

HIV (Human Immunodeficiency Virus) is a virus that causes decreased immunity. Examinations that can be done for HIV testing include a rapid HIV test and HIV viral load. The rapid HIV test is a test that is used to find out the antibodies in the sample, while the viral load test is to find out the amount of HIV. The purpose of this study was to compare HIV using a rapid test with HIV viral load in people with HIV. This type of research is observational with a cross-sectional approach. The sample in this study were 30 samples from HIV sufferers who carried out an HIV viral load examination using the Molecular Rapid Test (TCM) method at the RSUD Haji Provinsi Jawa Timur, then a rapid HIV test was carried out using the immunochromatography method. The results of the HIV rapid test and HIV viral load were processed in the form of a bar chart and tested with the McNemar statistical test. The results of the McNemar test can be concluded that there is a significant difference between the results of the HIV viral load examination of HIV viral load by the Molecular Rapid Test (TCM) method is superior to the HIV rapid test. However, rapid tests can be used in diagnosing HIV infection.
