Abstract

Palm oil mill effluent (POME) has high chemical oxygen demand (COD) content of 44.444 mg/L causing the most suitable processing technology is anaerobic digestion. This research aimed to study soluble COD removal efficiency and bacteria growth rate as VSS in acclimatization process of rumen fluid as inoculum addition in POME treatment. Acclimatization I was conducted in circulating bed reactor (CBR) with working volume 20 L, ratio of glucose : POME inside the reactor is 50% : 50% (%v/v). Meanwhile, acclimatization II was conducted in upflow anaerobic sludge blanket (UASB) with working volume 10 L, ratio of glucose : POME inside the reactor is 0% : 100% (%v/v). Acclimatization I and II was done in 20 and 50 days with soluble COD removal efficiency 80,36% and 65,41%, respectively. Meanwhile, bacteria growth rate as VSS has amount of 277 mg/L/day and 429 mg/L/day, respectively.Keywords: palm oil mill effluent, acclimatization, mixed culture bacteria, glucose, soluble COD, VSS

Abstract

Eupenicilium Javanicum is a mold that has high heat resistance. The purpose of this study was to determine the effect of temperature (T: 80 ° C, 85 ° C, 90 ° C) at soluble solid SS: 10 ° Brix on the thermal inactivation of E. Javanicum spores. Then determine the value of D and Z. This study uses the thermal method of pasteurization process and spore calculation using the spread plate method. The D-values obtained at SS 10 ° Brix for temperatures 80, 85 and 90 ° C were 10.82; 4.63; and 1.45 minutes and the Z-value obtained is in the at 11,47 ° C. It was shown that haiger temperature resulted in the decreasing of the timeneeded to inactivate the spores. The results of this study emphasize the importance of temperature to the D and Z value for pineapple juice pasteurization.Keywords: Ascospore, Inactivation, Pasteurization, Pineapple juice, E. Abstract

Hydroxyapatite is a bioceramics that is widely used as a bone and tooth grafting material, catalyst and adsorbent. Hydroxyapatite is a calcium phosphate compound that can be synthesized from natural ingredients with high calcium content such as coral reefs. Coral reef is a marine biota that has a CaCO3 content of 98.8%. This study aims to synthesize hydroxyapatite through the formation of Precipitated Calcium Carbonate (PCC) from coral reefs. The research procedure begins with the process of calcination of coral reefs to form CaO which is then made into PCC with carbonation methods. PCC as a source of calcium is synthesized into hydroxyapatite using precipitation processes with variations in temperature (30°C, 50°C and 70°C) and aging time (7 hours, 14 hours and 21 hours). Synthesized hydroxyapatite was analyzed by X-ray Diffraction (XRD) and Scanning Electron Microscopy (SEM). The results of XRD analysis showed the formation of hydroxyapatite compounds from 2 sudut angles in accordance with the data (ICDD 01-074-0565). Hydroxyapatite which was synthesized at a temperature of 50⁰C and aging time gave the best results with a monoclinic crystal structure, a degree of crystallinity of 96.0772% and a crystal size of 20.1846 nm. Abstract

The utilization of enzymes in bioprocess industry increases annually. Enzyme ability to accelerate the reaction and its biodegradability make it preferable to be utilized as catalyst. Cellulase and xylanase are ones of common enzymes that used in industries, such as pulp and paper, detergent, syrup and more. Pineapple peel waste has potential lignocellulose content to substitute pure substrate such as cellulose and xylan which is not efficient in cost. Solid-state fermentation allows the microorganism to live in condition that suits its habitat. This research aims to investigate temperature of fermentation’s (25, 30 and 35 oC) effect on cellulase and xylanase enzyme production in solid-state and comparing the enzyme activities resulted with submerged fermentation method ones. Fermentation for enzyme production was done for 96 h and the pH of medium was adjusted at 4. Enzyme activity was obtained with DNS method using spectrophotometry visible at 540 nm. The highest enzyme activities were obtained at temperature 30 oC which resulted 0,054 U/mL for cellulase and 0,077 U/mL for xylanase.Keywords : cellulase, enzyme, solid-state fermentation, submerged fermentation, xylanase
