Abstract

Pembelajaran materi keanekaragaman hayati yang kontekstual di sekolah sekitar Taman Wisata Alam Suranadi belum dilakukan karena tidak ada buku dan media VCD pembelajaran kontekstual yang menyajikan keanekaragaman hayati di sekitar sekolah. Faktor ini yang menjadi permasalahan utama mitra dan tawaran penyelesaiannya adalah dengan mensosialisasikan keanekaragaman hayati khususnya kupu-kupu yang ada di sekitar sekolah. Pembelajaran yang menyenangkan  berbasis lingkungan sekitar atau CTL (Contextual Teaching and Learning) karena siswa dapat  melihat secara nyata, mengalami sendiri materi pelajaran  yang ada di sekitar lingkungannya.  selanjutnya dengan CTL dapat meningkatkan hasil belajar  Siswa dibelajarkan dengan buku dan VCD yang berisi materi pelajaran yang ada di sekitar siswa. Hasil belajar siswa di SMPN sekitar taman wisata Suranadi masih di bawah standar KKM 75. Metode kegiatan ini adalah sosialisasi dengan penjelasan, penyajian isi buku dan penayangan VCD Pembelajaran, tanya jawab dan diskusi.   Tujuan dari kegiatan  ini untuk mensosialisasikan berbagai jenis kupu-kupu indah sebagai materi pengayaan keanekaragaman hayati di SMPN sekitar kawasan Taman Wisata Alam Suranadi. Setelah selesainya pelaksanaan kegiatan ini juga akan dilaksanakan  pendampingan pembelajaran yang dilakukan guru untuk melaksanakan pembelajaran berbasis lingkungan. Hasil pengabdian sebagai berikut 1) telah dilakukan sosialisasi berbagai jenis kupu-kupu indah sebagai materi pengayaan keanekaragaman hayati di SMPN sekitar kawasan Taman Wisata Alam Suranadi 2) peserta kegiatan pengabdian 100% mengikuti, menanggapi dan memahami materi sosialisasi.  Kesimpulan sosialisasi berbagai jenis kupu-kupu indah sebagai materi pengayaan keanekaragaman hayati di  SMPN sekitar TWA Suranadi dalam kegiatan pengabdian ini terlaksana dengan baik dengan antusiasme peserta kegiatan yang tinggi.Kata Kunci : Sosialisasi, Keanekaragaman hayati, Kupu-kupu, Suranadi

Abstract

This research was conducted to find out the capabilities of endophytic bacteria isolated from bark of the clove plants in inhibiting the growth of bacteria Streptococcus mutans, Staphylococcus aureus, Klebsiella pneumoniae and Escherichia coli and to identify endophytic bacteria that potensial to produce an antibacterial. Stages of this research include the isolation of endophytic bacteria, antibacterial test, biochemical test, and molecular identification based on 16 S rRNA. It was obtained about 29 isolates from bark of the cloves plants in which 6 of them isolates inhibited bacterial S. aureus and 3 isolates inhibited bacterial S. aureus and S. mutans. Abstract

The objective of this study was to isolate endophytic bacteria from bark of srikaya, analyzing antibacterial activity of endophytic bacteria in S. aureus, B. cereus and E. coli pathogenic bacteria, characterizing endophytic bacteria capable of inhibiting the growth of pathogenic bacteria and identification of endophytic bacteria able to inhibit the growth of pathogenic bacteria. Isolation of endophytic bacteria using TSA and NA media, bioassay on pathogen bacteria with concentration of 106 cells / ml with using Ø 6 mm wells and entering supernatant of 100 μl. Supernatant was obtained by growing endophytic bacteria in NB media shaken with shaker 150 cycles / min for 48 h at 32ᵒC then culture centrifuge at 5000 g for 30 min. Positive control using cyprofloxacin. Characterization is based on the nature of the colony, Gram paint, spore formation, and biochemical tests. The results of the study yielded 13 endophytic bacterial isolates and 4 endophytic isolates capable of inhibiting the growth of 8 pathogenic bacteria with sensitive criteria, 2 pathogenic bacteria with resistant criteria and 1 pathogen bacteria can not be inhibited its growth. Gram's paint results show that 4 endophytic isolates belong to Gram-positive, rod-shaped and spore forming cells. From the character possessed by the bacteria can be identified that the 4 bacteria endofit capable of inhibiting pathogenic bacteria are Bacillus brevis, Bacillus latesporus, Virgibacillus pantothenticus, and Bacillus circulansKeywords: Characterization, Endophytic Bacteria, Bark of Srikaya, Antibacterial

Abstract

White pomegranate (Punica granatum L.) is one of potential plant that can be use as an antimicrobial. The objective of this study was to determine antimicrobial activity of endophytic bacteria from white pomegranate (Punica granatum L.) in inhibiting the growth of pathogenic microbia (Staphylococcus aureus, Streptococcus viridans, Shigella dysentriae, Klebsiella pneumoniae and Fusarium sp. fungi). The stages of the research were isolation endophytic bacteria, antimicrobial assay, and characterization of potential endophytic bacterial. Endophytic bacterial were isolated from bark, leaves, flower, and fruit peel of white pomegranate taken from Gebang Baru, Mataram District. Antimicrobial assay was then performed using agar diffusion method, using supernatant of endophytic bacteria, water as negative control or ciprofloxacin as positive control. The potential isolates were characterized using Gram staining and biochemical test. The results showed that 2 endophytic bacterial isolates (DNX2 and BNG1) show strong activity against S. aureus, 3 endophytic bacterial isolates (BTG1, DNX11, and BNG1) show strong activity against S dysentriae, and 1 isolate KLBX11 shows medium activity against Fusarium sp. fungi. Those isolates are gram-possitive with bacill-shaped and produce spores. It can be concluded that it is possible to obtain potential endophytic bacterial from white pomegranate are potential as an alternative source for antimicrobial compounds.

Abstract

Batujai Dam is one of the largest dams in Central Lombok with an area of 890 ha of inundated water. The water from the Batujai Dam is used for irrigation of agricultural land, flood control, freshwater fisheries and the development of micro-hydro power plants. Batujai Dam is also a reservoir for several rivers in Central Lombok, namely:  the Leneng River, the Sade River and the Dodokan River. This function makes the Batujai Dam a place for the accumulation of various forms of pollutants carried by the inlet rivers. Microalgae is one of the ecological parameters that can function as a bioindicator of water quality. The purpose of this study was to determine the water quality in the Batujai Dam in terms of the microalgae diversity index and the measurement of environmental factors. Determination of sampling point and sampling time using purposive sampling method. Measurement of water quality is carried out by determining the diversity index and measuring environmental factors such as pH, temperature, DO, BOD, phosphate and nitrate. Microalgae data for determining the diversity index was analyzed using the Microsoft Excel 2010 program. The results showed that the identified microalgae consisted of 5 classes, 16 orders and 40 species. The overall diversity index of microalgae in the Batujai dam is 1.712 with a DO value of 5.09 and a BOD of 4.0175. Based on these data, it can be concluded that the water quality in the Batujai Dam is in an unhealthy condition (lightly polluted). The allotment for the use of water from the Batujai Dam is suitable for freshwater fish farming, livestock and agricultural irrigation, and it is not recommended for drinking water sources and water recreation infrastructure/facilities.

Abstract

Bacillus thuringiensis is one of the bacteriae species that be able to kill insects. The latest research showed that the cell plasm of this bacteria contains crystalline protein with natural insectisides properties. Therefore the insecticidal effect of this species on insects which is decreasing cultivation plant production is interest to be examined. One of the insect species that are well-known causes of loss of farm production in fruit flies (Drosophila melanogaster). Therefore this research is designed to isolate, purify, and thenassess Bacillus thuringiensis toxicity against Drosophila melanogaster as animal treatment. Bacillus thuringiensis was successfully isolated from soil of  cows cage by using Lauria Bertani medium contain 0,25 M buffer solution Acetic, pH 6.8. The result shows that Bacillus thuringiensis isolate is a gram-positive bacteria. Position of endospore is located between the middle and the end of the cell (subterminal spore). Toxicity test was conducted by mixing 0.5 g/L, 1 g/L, 2 g/L, 4 g/L of pure Bacillus thuringiensis isolate into  Drosophila melanogaster medium. The treatments effect were monitored for 15 consecutive days. Toxicity test shows that the treatment has no significant effect on the treated animal mortality. This result shows that crystal-producing Bacillus thuringiensis strain isolated from cows cage soil do not have insecticidal properties for Drosophila melanogaster. It is concluded that Drosophila melanogaster is a non-target species of Bacillus thuringiensis.

Abstract

Coastal areas have potential that can be developed by the community, one of which is beach tourism. Tourism activities around that take place around the coast can affect the aquatic ecosystems especially the community structure of phytoplankton. Phytoplankton are aquatic organisms that act as primary producers. Bacillariophyceae is a class of phytoplankton that is commonly abundant in marine waters. The objectives of this study were to determine the species composition, species abundance, and community structure of Bacillariophyceae in the waters of Klui Beach. This type of research is descriptive-explorative. The Haphazard sampling was applied for determining the sampling sites. Phytoplankton data were analyzed using the abundance formula, diversity index, uniformity index, and dominance index. The results showed that the composition species of Bacilllariophyceae consisted of 48 species which included in 17 orders. Average species abundance was 298.09 ind/L (low category). The spesies diversity index was 2.37 (moderate category), the species uniformity index was 0.86 (high category), and the species dominance index was 0.13 (low category).

Abstract

The purpose of this study was to determine the effect of lemongrass extract (Cymbopogon nardus) in inhibiting the growth of the bread fungus Aspergillus oryzae by using a combination of lemongrass extract with n-hexane as a solvent. This type of research is experimental research. The population in this study were all types of mushrooms isolated from bread. The sample used was the fungus Aspergillus oryzae isolated from bread. Lemongrass extraction was carried out by maceration method with n-hexane and ethanol as solvents separately, followed by mixing the two extracts with a combination of n-hexane extract: ethanol extract respectively as follows: 1:1; 2:1 and 1:2 before evaporation. The inhibition test of lemongrass extract against the fungus Aspergillus oryzae was carried out by growing the fungus on PDA media which had been supplemented with lemongrass extract with a concentration of 0.6%; 0.8% and 1%. Data were analyzed using the ANOVA test at a significance level of 95%, followed by the Honest Significant Difference (BNJ) test. The results showed that the combination of n-hexane extract-ethanol extract 1:1 and 1:2 showed low inhibition at all concentrations tested, while the combination 1:2 with 1% lemongrass extract concentration was the best combination in inhibiting the growth of the fungus Aspergillus oryzae ( 1/4 control mycelium colony diameter) and showed 74% inhibitory activity (strong inhibitory category). From the research, it was concluded that lemongrass extract using a combination of ethanol solvents, N-Hexane extract - ethanol extract with a 1:2 combination applied with a concentration of 1% inhibited the growth of the fungus Aspergillus oryzae in the category of strong inhibition, with an inhibitory activity of 74%.

Abstract

Abstrak: Kurikulum Mata Pelajaran IPA SMP tahun 2017 di Kota Mataram  menuntut para guru IPA untuk bisa meningkatkan keterampilan dalam mengembangkan perencanaan  dan mengimplementasikannya secara baik. Salah satunya adalah guru IPA SMP bisa mengembangkan perangkat pembelajaran. Untuk dapat mengembangkan perangkat pembelajaran IPA sesuai dengan standar,  para guru IPA harus memiliki keterampilan dalam mengembangkan perangkat pembelajaran, Para guru masih terpaku dengan meodel yang disarankan kemnetrian terkait, sementara  banyak model pembelajaran berbasisi siswa aktif salah satunya model 5E (Engage, Explore, Explain, Extend dan Evaluate).  Oleh karena itu perlu dilakukan kegiatan untuk mengmbangkan keterampilan  bapak/ibu guru IPA anggota MGMP IPA dalam perangkat pembelajaran model 5E. Desain yang digunakan adalah post tes no control group design. Populasi adalah semua guru IPA anggota MGMP Kota Mataram, sampel yang digunakan sebanyak 40 guru. Pengumpulan data dilakukan dengan mengevaluasi produk berupa perangkat pembelajaran hasil kegiatan workshop. Data dianalisis deskriptif kuantitatif. Hasil menunjukan bahwa perangkat pembelajaran yang disusun oleh guru peserta (90%) sudah baik dalam merencanakan langkah-langkah 5E. Tetapi dalam kaitannya dengan tagihan dan pengetahuan sebagian besar perangkat pembelajaran kurang mencakup keluasan dan kedalaman materi yang ditagih oleh kompetensi dasar pengetahuan dan keterampilan. Dengan demikian walaupun langkah-langkah pembelajaran berbasis siswa aktif dan memenuhi tagihan kurikulum tetapi tingkat asesmen belum memenuhi tagihan. Berdasarkan hasil penjelasan di atas dapat disimpulkan bahwa  90% bapak ibu guru IPA dapat menyusun perangkat pembelajaran cukup baik khususnya terkait sintaks  5E. Kata Kunci : pembelajaran IPA, model 5E, kajian kritis

Abstract

Abstrak: Kegiatan pengabdian pada masyarakat ini secara umum bertujuan memberi keterampilan kepada guru-guru Biologi SMA dalam memelihara hewan percobaan Drosophila, serta teknik-teknik dasar yang diperlukan dalam menggunakan hewan ini sebagai bahan percobaan pada pokok-pokok bahasan yang terkait dengan mekanisme pewarisan sifat. Kegiatan pelatihan ini dilaksanakan di SMAN 2 Labu Api dan diikuti oleh 23 orang peserta yang merupakan guru-guru Biologi SMA yang tergabung dalam MGMP Biologi Lombok Barat. Materi pelatihan meliputi Teknik Kultur Drosophila, Teknik Isolasi Betina Virgin, Teknik Menyilangkan mutan Drosophila, dan Teknik Mengatasi Kontaminasi Kultur. Hasil observasi menunjukkan bahwa dari 23 orang peserta pelatihan, 96% peserta belum pernah menggunakan Drosophila dalam praktikum, dan 100% peserta tidak memiliki keterampilan memelihara dan menggunakan Drosophila sebagai bahan praktikum di sekolah tempat mereka bertugas. Monitoring dan diskusi selama 10 hari pasca pelatihan menunjukkan bahwa hampir 74% peserta telah mampu melakukan dengan baik teknik-teknik dasar yang diperlukan dalam kultur Drosophila. Materi pelatihan tersulit yang dialami peserta adalah Teknik Mengatasi Kontaminasi Medium Kultur. Hingga 10 hari pasca pelatihan tidak seorang pun peserta melaporkan keberhasilan merekan dalam mengerjakan teknik ini. Meskipun demikian pelatihan ini berhasil karena sebagian besar materi pelatihan mampu dikerjakan dengan baik oleh mayoritas peserta pelatihan. Kata Kunci: pelatihan, kultur Drosophila
