Abstract

This study aims to determine the breaking dormancy of candleberry seeds which soaked with organic growth regulator Basmingro and its effect on seed viability. Research will be carried out in Limba U2, South City, Gorontalo City, for about 6 months. The materials used are candleberry seeds, organic PGR Basmingro. Candleberry seed soaked in a solution of organic PGR Basmingro in 5 treatment concentration (control (without PGR); PGR 0.02%, 0.03%, 0.04% and 0.05%) and 4 replications. Data were analyzed quantitatively by using Environmental Design Complete Randomized Design (CRD) and analysis of variance (ANOVA). Data are significantly different tested further by Duncan Multiple Rate Test (DMRT). The results are not significantly different, because no germination of candleberry seeds, caused by yet influential PGR is treated to break dormancy due to hard seed coat seed candleberry. So that further research needs to be done by increasing the dose and duration of soaking candleberry seeds with PGR organic Basmingro.

Abstract

This research aims to determine the occurrence on induction of protocorm in garlic on the media MS minimal macro and micro nutrient added coconut water. The materials used are garlic, coconut water and nutrient minimal macro and micro. The result was the establishment of direct induction of green protocorm from garlic explants. Conclusion of this research is the addition of 15% coconut water in medium MS minimal macro and micro nutrient can induce formation of protocorm directly from garlic explants.

Abstract

ABSTRACTMandarin’s SoE is national variety originated from Mount of Mutis, Sub District of SoE, of Timur Tengah Selatan (TTS) District, East Nusa Tenggara (NTT). The genetic diversity of citrus can be induced by gamma ray irradiation on embryogenic callus cells thus producing new mutants. Genetic diversity detection can be based on morphological and ISSR markers. The aim of this research was to obtain information on the genetic diversity on putative mutants mandarin SoE induced by gamma ray irradiation based on morphology and markers ISSR. ISSR markers used are ISSR 1, 4, 6 and 8. Analysis of morphological diversity produced a dendrogram with the level of similarity between individuals each irradiation dose 83-95% with 5-17% genetic distance. Dendrogram analysis based on the genetic diversity ISSR markers showed high levels of 51-100% similarity and genetic distance 0-49%. Individuals samples obtained from gamma irradiation, based both morphological and ISSR markers, was different from individual's genetic make up before irradiation.Keywords: cluster, gamma ray, genetic distance, genetic diversitys, similarity ABSTRAKJeruk keprok SoE adalah jeruk varietas unggul nasional yang berasal dari Pegunungan Mutis, Kecamatan SoE, Kabupaten Timur Tengah Selatan (TTS), Provinsi Nusa Tenggara Timur (NTT). Keragaman genetik jeruk ini dapat diinduksi dengan iradiasi sinar gamma pada sel-sel kalus embriogenik untuk menghasilkan mutan yang solid. Deteksi keragaman genetik yang terbentuk dapat dilakukan secara morfologi maupun molekuler dengan marka ISSR. Tujuan Penelitian ini adalah untuk mendapatkan informasi mengenai keragaman genetik yang terjadi pada mutan harapan jeruk keprok SoE hasil iradiasi sinar gamma berdasarkan morfologi dan marka ISSR. Marka ISSR yang digunakan adalah ISSR 1, 4, 6 dan 8 pada beberapa mutan harapan jeruk keprok SoE. Analisis keragaman secara morfologi menghasilkan dendrogram dengan tingkat kemiripan antar individu masing-masing dosis iradiasi 83-95% dengan jarak genetik 5-17%. Dendrogram analisis keragaman genetik berdasar marka ISSR memperlihatkan tingkat kemiripan 51-100% dan jarak genetik 0-49%. Individu-individu sampel yang diuji hasil iradiasi gamma, baik secara morfologi dan marka ISSR, telah memiliki susunan genetik yang berbeda dari susunan genetik individu sebelum diiradiasi.Kata kunci: grup, jarak genetik, kemiripan, keragaman, sinar gamma

Abstract

Tujuan kegiatan pengabdian adalah untuk pembuatan rumah bibit untuk penyemaian, penanaman dan pemanfaatan hasil berbagai macam tanaman sayur-sayuran sebagai bahan makanan untuk para anggota Dasa Wisma Mandiri “Anggrek Bulan”. Kegiatan dilaksanakan setelah diperoleh dana pengabdian Kolaboratif Fakultas, dibelikan alat dan bahan yang diperlukan, diantaranya baja ringan (Canal C) dan paranet. Hasil kegiatan berupa rumah bibit (rumah semai) yang dapat dimanfaatkan oleh para anggotanya untuk melakukan proses penyemaian beberapa jenis tanaman hortikultura terutama jenis sayur-sayuran.

Abstract

Techniques in the fold of shallot plant breeding can be done to obtain or improve existing characters to be better than before. One of them is the mutation induction technique with a chemical mutagen, namely colchicine. The purpose of the study was to find out the effect of colchicine concentration on growth, yield, character diversity and obtain value of Lethal Concentration 20-50% (LC20-50) for Tajuk Vriety M1 of shallot. The research was conducted in July-October 2019 in Libuo Village of Dungingi District of Gorontalo City. The study was conducted using a design randomized block design (RBD) with six level of colchicine solution concentration (0, 0.1, 0.2, 0.3, 0.4 and 0.5% (w/v)) with three repeats. The results of the variety analysis showed that a solution of colchicine concentration of 0-0.5% had the effect on the morphology of leaf colour, the number of saplings and the percentage of seeds growing abnormally and character of lear colour shallot varieties of mutant putative generation M1, but had no significantly effect on plant height, number of bulbs, wet and dry weights. CurveExpert1.3 analysis result, the lethal concentration value of colchicine solution in shallot bulbs can be obtained at a concentration of 0.33 (w/v) at LC30. Abstract

Tujuan kegiatan pengabdian ini adalah untuk melakukan penyuluhan dan pelatihan pembuatan pupuk organik cair dari limbah dapur pada warga di Desa Patoameme Kecamatan Botumoito Kabupaten Boalemo. Kegiatan ini berupa penyuluhan dan pelatihan yang dilaksanakan pada bulan November 2022. Kelompok sasaran pengabdian adalah warga desa seperti ibu-ibu atau bapak-bapak maupun remaja dari Desa Patoameme tersebut. Pupuk organik cair dari limbah dapur yang difermentasi dan dihasilkan dalam kurun waktu 15 hari dapat langsung dimanfaatkan atau diaplikasikan pada pertanaman sayur-sayuran atau pada tanaman hias yang ada di halaman rumah warga masing-masing.
