Abstract

Indonesia has high variety of pummelo [Citrus maxima (Burm.) Merr.], but only several cultivar are cultivated extensively. Others became vurnerable and the  germplasm has to be conserved to prevent their extinction. One of the conservation method is in vitro conservation  using a slow growth technique. Factorial experiment was used in designing the experiment, the first factor was MS medium concentration, i.e. 1/2MS and MS. The second factor was sucrose concentration, i.e. 0; 1; 2; and 3%. The results showed that low concentration of MS medium and sucrose reducing the leaf number and shoot length but increasing the root number and length.  Based on inhibition of growth,  the most reducing growth was planlet on MS without sucrose.

Abstract

Carnation has a highly economic demand of cut flower in Indonesia.  Field observations in West Java Indonesia was conducted in order to find the typical mottle symptoms that was a suspect caused by a virus disease. Identification of the virus was respectively conducted by performing ELISA test with four anti sera and characterizations held by bioassay, observing of virion particles, detecting of nucleic acid by RT-PCR and nucleotide sequencing. Total of 403 samples were collected from plants with or no virus-like symptoms. Among those all tested, 83% were found to be infected by Carnation mottle virus (CarMV), but negatively against Carnation ringspot virus (CRSV), Carnation laten virus (CLV), and Carnation vein mottle virus (CVMV) antisera. By mechanical inoculation, the virus was able to infect systemically Cenopodium quinoa and locally infect on others. However on Phalaenopsis sp and Gomprena globosa, there was symptompless found. The isometric CarMV particles size was approximately 30 nm. RT-PCR using specific primers of CP gene of CarMV successfully amplified a DNA sized 1000 bp.  CarMV West Java Indonesian (Idn-WJ) isolates possessed the highest nucleotide and amino acid homology with CarMV from Spain and was in the same cluster with CarMV from China, Taiwan and Israel.Keyword: Dianthus caryhophyllus, RT-PCR, sequence, Tombusviridae

Abstract

Tetraploid citrus are important for interploidal hybridization to create triploid seedless citrus.  Colchicine is the most commonly used as antimitotic agent to induce polyploid plants.  Tetraploid induction by colchicine in Pummelo ‘Nambangan’ was conducted in vitro using different types of explants.  The aim of this research was to induce tetraploid pummelo ‘Nambangan’ by colchicine treatment using germinated seed, shoot tip and cotyledonary node as explants.  Tetraploid shoot induction was conducted by soaking germinated seeds, shoot tips and cotyledonary nodes in 0.1% colchicine for 1, 3 and 5 h.  Regenerant shoots were grown on MS medium and their growth was observed after four weeks in culture.  Ploidy level was determined using flow cytometry analysis.  Stomata density, length and width of stomatal guard cell were also recorded. The results showed that shoot elongation was inhibited by colchicine treatment.  Soaking of shoot tip explants in 0.1% colchicine for 1 h resulted in 66.66% of putative tetraploid shoots.  Compared to diploid shoots, tetraploids had lower stomata density but bigger in guard cell size. Keywords: colchicine, tetraploid, pummelo (Citrus maxima (Burm.) Abstract

The aims of this study were to evaluate the physical and chemical change of pummelo fruit during storage with waxing and wrapping, and to determine the effect of waxing and wrapping and their  its interaction in affecting shelf life of pummelo fruits. This research was arranged in a randomized complete block design with two factors and three replications. The first factor was waxing consisted of without waxing, 10% beeswax, 5% beeswax, and chitosan. The second factor was wrapping consisted of without wrapping and wrapping using either transparent yellow or white plastic wrap. Pummelo fruits experience weight loss, reduced total soluble solid (TSS), decrease in firmness and total acidity during 48 days of storage in room temperature. Waxing using beeswax or wrapping using plastic wraps significantly suppressed weight loss and maintained fruit firmness compared to those without waxing and wrapping. However, waxing and wrapping did not significantly affect the quality of pummelo fruits. Based on the fruit’s weight loss, firmness, and panelist preferences, 5% or 10% bees wax can be used as waxing. Both transparent yellow and white plastic wraps can be used to maintain fruit weight, firmness, and appearances during storage.

Abstract

Common bean is a type of vegetable that can be consumed in the form of young pods. Potassium is one of the macronutrients needed to achieve maximum yield in common bean. Therefore this research is aimed at determining a suitable potassium source as well as an appropriate rate for drip irrigation in common bean using polyethylene mulch. The study was conducted from January to May 2018 at University Farm, IPB University. This experiment was set up as a nested randomized block design with two factors i.e. potassium sources as the main factor ( KCl, ZK, and NPK), and potassium rates (0, 37.5, 75, 112.5, 150 kg of K2O per ha). Fertilizer rates were nested in the main factor and fertilizers were applied Fertilization through a drip irrigation fertigation system with emitters placed close to the roots of plants. Fertilizer sources had significant effects on the variables of growth and bean yields. NPK resulted in a better growth and yield compared with KCl and ZK. Fertilizer sources that have very significant effect were likely related to the shape and solubility of the fertilizers, the ease of application, and also completeness of nutrient content in fertilizer. Based on the pod weight per plot, the productivity per hectare of bean pods produced in this study reached 7.52 ton.ha-1. Potassium (K2O) rate did not have significant effects on the variables of growth and yields of the bean.

Abstract

Pruning is one of the techniques to improve plant growth and production. The aim of this research was to determine the effect of pruning on vegetative and generative growth, and fruit quality of ‘Crystal’ guava. The experiment was conducted from September 2016 to April 2017 at Cikabayan Research Station of Bogor Agricultural University, Dramaga, Bogor, Indonesia (-6.550780, 106.714531). The experiment used completely randomized design with six replications. There were three treatments i.e. a) pruning by leaving 4 pairs of leaves, b) pruning by leaving 8 pairs of leaves, and c) control (without pruning). Pruning was applied after shoot leaves grew fully. The result showed that pruning significantly increased the total number of shoots and generative shoots. Pruned plants produced more flower and fruit than un-pruned ones. Pruning by leaving 4 pairs of leaves tended to produce higher number of flower and fruit than those by leaving 8 pairs of leaves. Fruit weight and internal fruit quality were not affected by pruning.

Abstract

Penelitian ini bertujuan untuk mengevaluasi karakter morfologi dan biokimia dengan isoenzim aksesi pamelo berbiji dan tidak berbiji asal Sumedang, Kudus, Pati dan Magetan. Karakter morfologi yang berperan dalam pengelompokan aksesi pamelo adalah tebal epikarp, pinggiran helai daun, panjang kantong jus, warna kulit buah masak, lebar sayap daun dan bentuk buah, sedangkan karakter isoenzim adalah MDH (Rf 0.11 dan 0.14) dan ACP (Rf 0.24 dan 0.33). Pita ACP Rf 0.24 dapat dijadikan penanda untuk membedakan aksesi berbiji dan tidak berbiji.  Dendrogram berdasarkan karakter morfologi memisahkan kelompok aksesi berbiji dan tidak berbiji pada koefisien kemiripan 0.63, dan berdasarkan isoenzim pada koefisien kemiripan 0.49. Dendrogram berdasarkan karakter morfologi dan isoenzim dapat membedakan antara aksesi berbiji, potensial tidak berbiji dan tidak berbiji. Hasil pemetaan komponen utama kongruen dengan dendrogram, yaitu dapat memisahkan aksesi berbiji maupun tidak berbiji, berdasarkan karakter morfologi, isoenzim maupun kombinasinya.KeywordsCitrus maxima (Burm.) Up to now characters that can distinguish seeded and seedless pummelo accessions are not yet known well. The objective of this work was to evaluate morphological and biochemical (isoenzyme) characters of seeded and seedless pummelo accessions originated from Sumedang, Pati, Kudus, and Magetan. Morphological identification used vegetative and reproductive component of pummelo tree, based on IPGRI descriptor list. Isoenzyme analysis was done by using young leaves and esterase (EST), malate dehydrogenase (MDH), peroxidase (PER), acid phosphatase (ACP), as well as glutamate oxaloasetate transaminase (GOT) isoenzymes. Morphological characters that contributed in grouping pummelo accessions were epicarp thickness, leaf lamina margin, vesicle length, epicarp color, petiole wing width and fruit shape, while isoenzyme characters were MDH (Rf 0.11 and 0.14) and ACP (Rf 0.24 and 0.33). Fruit shape (pyriform), fruit axis (hollow), seeds number (<10) per fruit, and ACP band at Rf 0.24 could be used as marker to differentiate seeded and seedless pummelo accessions. Separation between seeded and seedless accessions can be done based on morphological characters occured at similarity coefficient of 0.63 while on isoenzyme characters occured at similarity coefficient of 0.49. Dendrogram based on combined morphological and isoenzyme data was able to differentiate seed bearing and seedless pummelo accessions. Principal component analysis results was congruent with that of morphological, isoenzyme, and combination of them.

Abstract

Selection and identification of effective microbes are important steps to obtain biological control agents. The objective of this research was to screen potential bacteria as controlling agents for rice sheath blight disease. The research was conducted at plant bacteriological laboratory and green house of Plant Protection Division of Bogor Agricultural University, Bogor, from May 2010 to February 2011. The trial was arranged in a completely randomized design with bacterial isolates as treatment. The result showed that 30 out of 144 bacterial isolates indicated an antifungal activity to R. solani. In vivo test indicated that 3 of the 30 isolates which have antifungal activity were able significantly to suppress the rice sheath blight disease. Those isolates were marked as TT47, SS19 and BR2, with the ability to suppress rice sheath blight disease at rate of 79.6, 56.4, and 49.4%, disease index 1.7, 3.7, and 4.3, and the disease incidence 33.3%, 73.35, and 80%, respectively. Molecular characterization of partial sequence of 16S rRNA on SS19, TT47, and BR2 isolates showed that those bacteria are Serratia marcescens, Ralstonia pickettii, and Bacillus subtilis, respectively.
