Abstract

Aeromonas hydrophila is a gram negative and opportunistic bacteria, which could cause fish mortalityin a short time from 80%-100%. One virulent factor of A. hydrophila on common carp (Cyprinus carpio L)that could cause fish mortality is aerolysin. This research used a synthetic primers of oligonukleotide todetect aerolysin, a specific genomes of A. hydrophila on common carp (Cyprinus carpio L). The commoncarps have been feed a woof that contain garlic extract during 30 days before they challenged with A.hydrophila. Polymerase Chain Reaction (PCR) was used to detect an aerolysin gen from A. hydrophila. Theelectrophoresis result showed aerolysin gene of Aeromonas hydrophila from Veterinary Faculty of GadjahMada University (FKH-UGM) isolate was amplified with 462 bp of molecule weight. While the aerolysingen was detected in the fish kidney with 900 bp of molecule weight. Further, DNA sequence analysis of thePCR product of A. hydrophila from FKH – UGM isolate showed homolog with isolate A. hydrophila subsphydrophila ATCC 7966 complete genome with score 55.4 (71%).

Abstract

This study aims to determine the types of lactic acid bacteria grown at different pH from floc (clots) which are given carbon sources of tapioca and can increase the number of bacteria in the medium of red tilapia cultivation. The method used is the survey method and experiment by growing bacteria on different pH treatments, namely pH 2, 4 and 6, growing colonies were identified based on morphological and biochemical characteristics. The results of identification found 13 isolates of lactic acid bacteria (BAL) which only grew at pH 4 and 6 of these isolates consisted of the genus Bacillus and Streptococcus. The total bacteria in the cultivation medium ranged from 2x108 to 7x108 CFU / mL.

Abstract

Apel Mango leaf is a natural ingredient that can be used as an alternative medicine because it has phytochemical content in the form of tannins, flavonoids, saponins and alkaloids. This study aims to analyze the effect of giving a solution of apel mango leaf (Mangifera Indica) in the treatment of Pangasionodon hypophthalmus, and get the best dose for the treatment of diseases caused by E. tarda bacteria, seen from total leukocytes , leukocyte differentiation, phagocytic activity and survival of Pangasionodon hypophthalmus. This study uses a completely randomized design (CRD) with 5 treatments and 3 replications. The dosage of apel mango leaf solution used was Kn (not infected and not treated) Kp (infected and not treated), P1 (1.2%), P2 (1.4%) and P3 (1.6%) . Immersion is carried out in 5 liters of water with a mixture of mango leaf solution in accordance with different doses for 5 minutes 3 times soaking interval of 2 days , then maintained for 14 days after treatment . The results showed that soaking with apel mango leaf solution significantly affected the image of leukocytes of P. hypophthalmus (P <0.05) . Best dose contained in treatment P2 (1.4%) with an average total leukocytes ( 8,33 × 104 cells / mm 3 ) , differentiation of leukocytes (lymphocytes 84.33%, monocytes 9,00 and 6,67 neutrophils %), phagocytic activity ( 36.00% ) and survival rate of Pangasionodon hypophthalmus ( 86.67% ) .

Abstract

The aim of this research was to  determine the types of lactic acid bacteria from biofloc technology with the addition of mollase in red tilapia aquaculture containers and to determine the optimum growth of lactic acid bacteria at pH 2, 4, and 6, and also to observe the density of bacteria in the biofloc container water during maintenance. The methods used in this research are survey and experiments by taking flok from maintenance container and then identifying the lactic acid bacteria. The results of lactic acid bacteria inoculation obtained 16 isolates isolated from floc which can only grow at pH 4 and pH 6. Identification based on physical tests, chemical tests, and biochemical tests showed that the isolates were classified into Bacillus and Streptococcus. The amount of bacterial density water from containers of biofloc technology were given mollase during research reached the highest point in the third week, which averaged 109 CFU/mL.

Abstract

The vaccine is one of the alternative to improve infection of the immunity system. The purpose of this research was to understand the hematological of (Clarias gariepinus) that were with given Aeromonas hydrophila vaccine. This research was conducted on 14 April until 2 June 2019. The method was an experimental method with a Completely Randomized Design (CRD) five treatments and three times replication. The treatments appliced were: Kn: (The fishes that were not given with vaccine and infected by Aeromonas hydrophila), Kp: (The fishes that were not given vaccine and was infected by Aeromonas hydrophila, P1: Soaked with vaccine dose 1 ml/10 L, P2: Soaked with vaccine dose 1 ml/10 L, P3: (Dipping with vaccine dose 1 ml/10 L. Dipping with the vaccine is done for 15 minutes. Clarias gariepinus at average 8-10 cm TL. After 23 day, the fishes were infected with Aeromonas hydrophila, the purpose of which was to determine imumunity of fish. The results show that the vaccine for 15 minutes is able to prevent the Motile Aeromonas Septicemia (MAS) disease. The were range total erythrocyte is 2,29×106 sel/mm3, hemoglobin 12,60 g/dl, Leukocytes Differensial (Lymphocytes 71,33%, Monocytes 16,66% and Neutrophils 12,00%) and Survival Rate 63,3%.

Abstract

This study was conducted from October to December 2012 at the subdistrict of WestBangkinang, Riau Province. The study aimed to described the basic information ofwater quality in different technology of maintenance (traditional, semi intensive andintensive) catfish in freshwater pond especially the contains of macrozoobenthic. Watersample were taken from three station whereas catfish sample were taken in threelocation and each station consist of 4 heads in each station. The catfish sample will betaken every week for 1 month. The results of (H’) and (C), station 3 had low (H’) withdominanted of (C), and it means they got hard pressure of environment activity.Besides, Aeromonas caviae was found in all technology of maintenance freshwaterponds. The result of Aeromonas salmonicida bacteria, was founded in three sample incatfish of different freshwater pond as A. sub smithia and A. sub achromogenes. Inaddition to measurement of water quality consist of temperature 27,9-28,6 0 C; pH : 5,7-6,4; DO :4,9-5,7 ppm; BOD :11,3-14,7ppm; COD : 14,7-18,2 ppm. Physics andchemistry sediment is muddy more than 65% from each station. According toPP.82/2001 about water quality of aquaculture, it was putted on the 3 rd class.Keywords: Macrozoobenthic, water quality, aeromonas salmonicida

Abstract

The aim of this research was to isolate bacteria that had potential to act asprobiotics from the intestine to Oreochromis niloticus being cultured in RiauProvince; to investigate the optimum growth of at pH 2, 4 and 6; and to test theantibacterial potency of bacterial candidate probiotic against candidatepathogens Streptococcus agalactiae. The methods used in this research are Surveyand experiments methods were candidate in this research. Probiotic bacteria wasisolated from the inner wall of nila intestine. Twenty isolates were obtained fromfour fish. Identification based on physical, biochemical and morphologicalcharacters indicated that the isolates were classified into Bacillus andLactobacillus sp. Two of 20 isolate showed highest clear zone against S.agalactiae. They were isolate N-42 (22.7 mm), and N-62 (7.2 mm). Abstract

ABSTRACTThe heterotrophic bacterial isolate used came from the collection of the Marine Microbiology Laboratory, Department of Marine Sciences, University of Riau. This research aims to look at the effect of feeding fish mixed with heterotrophic bacteria on survival rate, absolute weight growth, heterotrophic bacterial counts and water quality. This research was conducted in May - June 2019. This research used an experimental method with a Completely Randomized Design (CRD), carried out four levels of treatment with three replications. The treatments given were P0 without the addition of bacterial isolates, P1 addition of isolate J, P2 addition of N isolates and P3 addition of all isolates. The results showed the level of life graduation obtained a value of P> 0.05, ie there was no effect of the addition of bacterial isolates to the level of survival rate. The growth of absolute weight is influenced by the addition of bacterial isolates with a P value <0.05 and the highest average is at P3 of 6.09 g. In the measurement of bacterial isolate water quality did not give effect, but the water quality is still optimal for fish maintenance with temperatures between 27-28ºC, pH between 7.0-8.0, DO between 3.8 - 4.5 mg / l, and ammonia 0.70-1.82 ppm. Keywords: Heterotrophic Bacteria, Survival Rate, Tilapia Saline (Oreochromis niloticus), Weight Growth.

Abstract

ABSTRACT Rhizophora sp. is one of the mangrove plant which potentially for antibacterial compound, because it has an alkaloids, saponins, flavonoids and tannins. This research purpose was to know the proper dose of Rhizophora sp. leaves extract in healing the MAS disease (Motile Aeromonas Septicaemia), which can be seen through the leukocytes differentiation of Pangasius hypophthalmus. The method in this research was the experimental method using the Completely Randomized Design (CRD) with one factor, four treatment levels and three times replication. The treatments were P0 (without immersion)and the immersion with leaves extract at dose P1 (1300 ppm), dose P2 (1600 ppm), and dose P3 (1900 ppm). The immersion was implemented twice in 24 hours for about 5 minutes. The fishes were used at 13-15 cm of length for 120 fishes which were infected by leaves extract. The results showed that the immersion of Rhizophora sp.leaves extract at dose 1600 ppm of leaves extract dose could treat the MAS disease (Motile Aeromonas Septicaemia) of Pangasius hypophthalmus. The total average of leukocytes was 69,61×103cell/mm3, leukocytes differentiation was 81,33% of lymphocytes, 9,66% of neutrophils, 9,00% of monocytes. The fagocyte index was 36,66% and the survival rate was 84,61%.Key Words : Leukocytes, Pangasius hypophthalmus,Rhizhopora sp.leaves extract

Abstract

This study was conducted over three months in the laboratory of Fish Diseases and Parasites, Faculty of Fisheries and Marine Sciences University of Riau Pekanbaru. The purpose of this research was to determine the structure of (Mystus nemurus) liver that were immersed with Curcuma xanthorrhiza ROXB extract and were infected by Edwardseilla tarda and to find out best dose Curcuma xanthorrhiza ROXB extract for the prevention of Edwardsiella tarda infections. The method used is an experimental method at a dose of treatment and analyzed descriptive. The treatment used is an extract of Curcuma xanthorrhiza ROXB with different concentrations, they were: Kn (Fishes were not receive any treantent), Kp (Fishes were not receive any treatment but were infected with E. tarda), P1 (immersed in curcuma with concentration 0,2 g/l), P2 (immersed in curcuma with concentration 0,4 g/l), P3 (immersed in curcuma with concentration 0,6 g/l) and were infected with E. tarda (0.1 ml of 108 of E. tarda), Fish organ (liver) were processed for histological studied (formalin fixed, alcohol series, 6 sliced and HE Stained). Result indicate that the structure of liver of the treated fish showing abnormality symptoms, such as necrosis, congestion, fatted degeneration and hemorrhage. From these, it can be concluded that immirsion in Curcuma with 0.6 g / l concentration is capable of inhibiting bacterial Edwardsiella tarda.Key word : Liver, Mystus nemurus, Curcuma xanthorrhiza ROXB, Edwardsiella tarda
