Abstract

Indonesian marine environment is one of the most vulnerable waters from oil pollution in the world. Therefore, a hard effort to minimize oil pollution impact is really needed. Physical and chemical approaches have been already popular to combat oil pollution; one of the other promising techniques is bioremediation, the use of microorganism to detoxify or remove pollutants. Research and application of bioremediation on soil environment has been started in Indonesia; however, in marine environment it is still need to be studied due to more complicated aspects and also difficulties. Development of bioremediation study on marine environment in Indonesia has been started from enumeration, isolation and identification of oil degrading (hydrocarbonoclastic) bacteria. Researches on taxonomic and functional genes have been conducted. Biostimulation and bioaugmentation studies are ongoing research which conducted from laboratory scale and microcosm scale to field experiment (sand column). To have a manual or guidelines on conducting bioremediation in marine environment is not easy and still many steps have to be done. Several aspects concerning with this study such as the diversity of polluted sites characteristic, oils characteristic, oceanographic conditions and engineering has to be studied comprehensively.

Abstract

Petroleum pollution has bad impact for life in marine ecosystems, particularly in coastal areas. One method of handling the pollution is bioremediation using bacteria that are able to degrade and utilize petroleum hydrocarbons as carbon source. One technique of bioremediation is biostimulation, that is the addition of nutrients that can improve the process of oil degradation by degrading bacteria. Nutrient slow-release fertilizer are used, one of which is Gramafix. The purpose of this study was to determine the optimal concentration of Gramafix in increasing degradation of petroleum contaminants by indigenous bacteria. Four concentrations of Gramafix as treatments are P1 (0,085 g), P2 (0,171 g), P3 (0,341 g) and P4 (0,682 g), as well as the negative control treatment (no fertilizer and bacteria) and positive control (with bacteria, without fertilizer). Observations were carried out  four times, on 0, 7, 14 and 28 days of incubation. The parameters used are heavy oil using the gravimetric method, the total number of bacterial cells using Acridine Orange Direct Counting and environmental factors such as nitrogen contents, phosphorus contents, temperature, dissolved oxygen, pH and salinity. Analysis of the data using a completely randomized design (CRD) with ANOVA and Duncan test. The results showed the P3 can improve the process of oil degradation by bacteria that degrade the highest percentage of 65,91% in the 28 days of incubation, it also has more bacterial cells than other treatments. The result of this study is the addition of 0,341 g Gramafix is optimum for enhancing the degradation of petroleum hydrocarbons by degrading bacteria. Abstract

CULTURE INDEPENDENT APPROACH FOR BACTERIAL COMMUNITY ANALYSIS. Analysis of bacterial community can be through two approaches, through cultivation (culture dependent) and without cultivation (culture independent). Culture dependent approach is conventional method which only covered few bacteria because not all bacteria could be cultured. Culture independent approach with molecular techniques based on DNA communities can provide more information about the structure and diversity of bacteria in nature, both culturable bacteria and unculturable bacteria. 16S rRNA gene is commonly target gene used in bacterial communities analysis. Other specific target genes also being developed for specific groups of bacteria. Several methods are developed for the analysis of molecular markers 16S rRNA or other specific genes, including Denaturing Gradient Gel Electrophoresis (DGGE), Terminal Restriction Fragment Length Polymorphism (TRFLP), and Single Strand Conformation Polymorphism (SSCP).

Abstract

APPLICATION AND EFFECTIVITY OF BIOREMEDIATION TECHNIQUE FOR OIL CONTAMINATED COASTAL WATERS. Oil pollution is often occurs in Indonesian marine waters and it has a significant impact on ecological and economical aspects. Physical and chemical approaches are common to be used to combat this pollution. Bioremediation is a promising method using biological approach that not been used yet for oil spill clean up in Indonesian coastal environment. The success stories were reported mostly from sub-tropic region. This technique is potential to be used in Indonesian due to tropical climates and enormous microbial resources in Indonesian marine waters. However, intensive research and field test are still needed to develop an appropriate method for application in Indonesia. One of bioremediation technique limitations is the suitability for each polluted sites. This paper will discuss about the application and the affectivity of bioremediation technique in other countries and research development of this technique for coastal environment in Indonesia.
